    Notes: Abstract Changes in phosphorus metabolism were studied by examining the incorporation of32P and33P into cells ofCandida utilis growing in phased culture during a 6 h cell cycle and a post-cycle period of 6 h. Three different chemically defined media were used; these were phosphorus, nitrogen and carbon limited. The patterns of incorporation of phosphorus into RNA, DNA, lipid and cold water extractable phosphate fractions showed a non-uniform behaviour during both cell cycle and post-cycle periods. The patterns were different in all three types of media. The results showed that a cell can grow and develop at a fixed growth rate in different ways: so that the pattern of behaviour during a cell cycle is not stereotyped for a given doubling time, but largely depends upon the nutrient environment in which the cell exists.

    Notes: Summary Growth, sporulation, insecticidal crystalline protein (ICP) production and plasmids of Bacillus thuringiensis var. kurstaki (HD-1) were investigated during batch and continuous phased cultivation using a laboratory scale cyclone fermentor. When grown in batch culture at 28°C, 93% of the cells sporulated and produced ICP within 10 h of commencement of stationary phase. The batch culture runs were completed within 50 h of inoculation. A predominantly sporogenous and crystalliferous cell population was also obtained by second, stage processing of culture harvested from the first 10 to 25 cycles of continuous phased cultivation. In contrast, after 25 or more cycles of cultivation the population in the continuous phased culture shifted towards a predominantly asporogenous and acrystalliferous one. Culture conditions in continuous phased cultivation did not affect the plasmid content of B. thuringiensis (HD-1), yet influenced sporulation and plasmid-coded ICP production.

    Notes: Continuous phased growth produces a culture in which most of the cells in the population are in the same stage of their development. The cell, thereby amplified by the size of the synchronous population, may be examined in the phased culture at any desired growth rate. Changes taking place in the cell after the cell cycle, i.e., post-cycle changes, may be examined by a modification of the procedure. Further systematic applications of the method permit a rational approach to problems of cell growth and metabolism.The phasing technique recognizes the cells as the fundamental unit for experimental investigation, and offers a great potential in the analysis of the cell throughout its cycle, a relatively unexploited field in cell physiology and fermentation. Experiments with yeasts and bacteria illustrate some of the applications and progress made so far.

    Notes: A simple inexpensive apparatus with a working volume of 10 liters of culture is described. Details of construction and procedures for operation of the cyclone column vessel are given. The vessel is self-contained, so that experimental parameters of temperature and aeration are individually controlled; homogeneous mixing and representative sampling of the culture, besides control of foam without need for antifoam, are obtained. The vessel may be used in single or multistage systems for aerobic or anaerobic cultivation of organisms in batch, chemostat, or phased cultures.

    Notes: Acid soluble extracts obtained at 30 min intervals from cells of C. utilis growing in synchrony in a phased culture (cycle time 51/2 hr) were fractionated on a Dowex-1-formate column. The series of fractionation profiles showed changes in number and amounts of components over the cell cycle. Transient accumulations of numerous components over the complex pool were observed. The significance of the changes are discussed in relation to practical applications and cell metabolism.

    Notes: An inexpensive scale-up of the cyclone column apparatus (Can. J. Microbiol., 11, 893 (1965)) is described which is suitable for pilot plant studies on the 10 liter scale. Details of its construction and operation are given.Experimental with phased cultures of C. utilis growing in the equipment are reported during which studies of dissolved oxygen and oxygen uptake were made over the cell cycle. Changes in both oxygen parameters occurred during the cycle and were useful as monitors of the growth. Cell of C. utilis were found to be very sensitive to changes in oxygen supply during the early stages of the cell cycle.

    Notes: Growth of Saccharomyces rouxii, an osmophilic yeast that produces glycerol and arabitol, was investigated using chemically defined media in continuous culture. Total polyol production increased with increasing aeration and growth rate. At very low aeration levels ethanol was produced but as aeration levels were increased ethanol could not be detected, formation of d-arabitol remained constant and formation of glycerol increased.In fully aerobic cultures with a replacement time of twelve hours, nitrogen-limited growth resulted in accumulation of glycerol in the medium. In glucose-limited growth glycerol tended to decrease. It was probably metabolized as a secondary carbon source. The optimum temperature for polyol production was 30°C while that for cell growth was 27°C.
