Notes: SYNOPSIS. Steady-state oxygen kinetics of Trypanosoma mega reveal the presence of 3 oxidases. These include an oxidase which is sensitive to salicylhydroxamic acid (SHAM) but insensitive to sodium azide. This oxidase could be the L-α glycerophosphate oxidase present in bloodstream trypanosomes. In addition, an oxidase is present which is azide-sensitive but SHAM-insensitive. This oxidase is inhibited by CO and is probably cytochrome aa3. A 3rd oxidase is insensitive to both azide and SHAM but is inhibited by CO and is possibly cytochrome o. Reciprocal plots of T. mega reveal the presence of 2 oxidases that are inhibited by CO. These results are discussed in the light of previous evidence suggesting the presence of several oxidases and a branched electron transport system in T. mega.

Notes: Abstract A mass spectrometer with membrane inlet was used to measure methane and oxygen utilization rates at various methane concentrations in Methylosinus trichosporium and a locally isolated strain of a methane-oxidizing coccus (OU-4-1). The apparent Km for methane was found to be 2 μM for M. trichosporium and 0.8 μM for strain OU-4-1. These Km-values are 10–30 times lower than most previously reported values. The ratio of oxygen to methane utilization rates was 1.7 for M. trichosporium and 1.5 for strain OU-4-1 corresponding to a growth yield of 0.38 and 0.63 g dry weight/g methane, respectively.

Notes: Summary The acoustic behavior of four species of Vespertilionid bats flying in the field was studied using high speed tape recorders and ultrasonic detectors. The bats can be identified solely on the basis of their cries when using a ‘divide-by-ten’ detector. Three species (Eptesicus serotinus, Nyctalus noctula, andPipistrellus pipistrellus) show considerable flexibility in their acoustic behavior, whileMyotis daubentoni has a stereotyped acoustic behavior under our study conditions. Several aspects of the cry repertoire can be correlated with the bats' activities and acoustic environment. During aerial chases and when circling the roost,E. serotinus, N. noctula andP. pipistrellus emit ultrasound that is distinctly different from their orientational cries. Such ultrasound may have a social function.

Notes: Summary The kinetics of methane uptake by Methylococcus capsulatus (Bath) and its inhibition by ammonia were studied by stopped-flow membrane-inlet mass spectrometry. Measurements were done on suspensions of cells grown in high- and low-copper media. With both types of cells the kinetics of methane uptake are hyperbolic when oxygen is in excess. The apparent K m and K max for methane uptake are both higher in low-copper cells than in high-copper cells. Ammonia is a simple competitive inhibitor of methane uptake in high-copper cells when the oxygen concentration is above a few μM. The findings agree with the assumption that ammonia is a week alternative substrate for particulate methane monooxygenase. In low-copper cells the effect of ammonia is complicated and cannot be explained in terms of current assumptions on the mechanism of soluble methane monooxygenase. Our data indicate that ammonia inhibition is likely to be a more serious problem in connection with cultivation in low-copper medium than in high-copper medium.

Notes: Summary A turbidostatic and oxystatic fermentation system was used to study the growth kinetic ofMethylococcus capsulatus (Bath). Dissolved oxygen and methane concentrations were measured continuously with membrane inlet mass spectrometry. The specific growth rate was found to increase from 0.25 h−1 to 0.37 h−1 and the saturation constant for methane was found to decrease from 71 μM to 1.3 μM as the copper content of the medium was varied from a very low to a high value.
