    Notes: Summary Continuous decolorization of kraft black liquor by mycelial pellets ofCoriolus versicolor in the presence of glucose as co-substrate is discussed. A linear relationship was developed between the rate of decolorization and the liquor concentration. The rate constant was equal to 0.00961 gmyc−1 h−` at 22°C. During the continuous experiments the pellets exhibited no apparent loss of activity when the liquor concentration was in the range of 400 CU l−1 to 5000 CU l−1. However, in the repeated batch experiments a loss of activity was observed which was dependent on the initial liquor concentration. The half-life of pellets was equal to 4.7, 9.4 and 20.2 days for the initial liquor concentration of 1380, 31 780 and 6990 CU l−1, respectively. The production of the extracellular enzyme, laccase, was followed but could not be used as an indicator of the ligninolytic activity. The involvement of the intracellular enzymes ofC. versicolor in the decolorization process is described.

    Notes: A method for positioning an oriented fragment within the unit cell is presented. It is based on a correlation between a model and observed data which is performed in Fourier rather than Patterson space. Symmetry-related molecules are located in the electron density map calculated in space group P1, with the phases derived from a model that is correctly oriented but arbitrarily positioned in the unit cell. It is shown that considering all symmetry elements simultaneously substantially increases the sensitivity of the method and makes it less susceptible to the errors in the model. The procedure also automatically incorporates a penalty for the overlap of symmetry-related molecules, and the stringency of this requirement is easily modified. The method has been tested on two different proteins and the results compare favorably with other translation functions.

    Notes: Abstract A white-rot fungus Coriolus versicolor was immobilized by entrapment in calcium alginate beads. Treatment of a kraft mill effluent with the immobilized fungus in the presence of sucrose resulted in 80% loss of color of the effluent within 3 days. The minimal concentration of sucrose required for the decolorization was 10 mM. Other carbon sources (xylose, glucose, glycerol, and ethanol) could also be used.

    Notes: Summary The plasmid pRH271, harboring a xylanase gene cioned fromBacilius subtilis, has been transferred into a mutant ofE. coli SK2284 which allowed the release of part of the xylanase in the culture supernatant. Kinetic parameters of this recombinantE. coll strain were determined in microscale batch culture with and without the selective pressure of antibiotics. No significant difference in µmax was observed for the nontransformedE. coli strain when compared to the recombinant strain. However, K5 values for glucose were two times higher in the case of the recombinant strain. Preliminary study of xylanase production in a large batch farmenter was also described.

    Notes: Refiner mechanical pulp was biologically treated with several higher fungi in order to test their potential for increasing the strength of paper. It was among the white-rot fungi that the best results were obtained. Polyporus versicolor gave the best overall improvement in handsheet properties with no reduction in tear. The strength improvement is due to attack on lignin and to an increase in fiber flexibility as measured by water retention values and by acidic group content of the treated pulps. The brown-rot fungi had a detrimental effect on paper properties.

    Notes: Screening fifteen strains of white-rot fungi for their ability to decolorize combined bleached kraft effluent showed that Coriolus versicolor in liquid culture removed over 60% of the color of the effluent within six days in the presence of sucrose. Treatment of the same effluent with this fungus, immobilized in beads of calcium alginate gel, resulted in 80% decolorization after three days in the presence of sucrose. Caustic extraction E1 effluent was also decolorized by the immobilized fungus. Decolorization was achieved more rapidly at pH 5.0 than at pH 7.0. Recycled beads could remove color efficiently and repeatedly in the presence of air but not under anaerobic conditions.

    Notes: The growth kinetics of the yeast Saccharomyces cerevisiae and the production rate of ethanol have been studied in batch fermentation under anaerobic conditions in a 20-L fermentor. Two substrates were used in fermentation trials: a synthetic mixture of three fermentable sugars, D-glucose, D-mannose, and D-galactose, and a low-yield liquor originating from a bisulfite cooking process. The Monod model adequately described the system in relation to the specific growth rate μx and the specific product formation rate μP. Different fermentation parameters (growth rate, substrate utilization, and product formation) were determined for the synthetic mixture and the bisulfite liquor. It was observed that the specific growth rate is much lower in spent sulfite liquor than in a synthetic medium. However, the specific product formation rate remains the same in both media.
