    Notes: Abstract Prevention and control of the spread of IBR/IPV virus in cattle presumes exact diagnosis even with large sample collections. The requirements are fulfilled firstly by the application of the most sensitive testing methods for virus specific antibodies, such as the enzyme-linked immunosorbent assay (ELISA), and secondly by the introduction of bovine milk as a specimen for sampling. Data are presented demonstrating the application of a commercial ELISA system for evaluation of large sample collections and its optimization for this purpose.

    Description / Table of Contents: Zusammenfassung Die Antikörperquantifizierung ist im EIA auch ohne Standardkurve möglich. Bei der sogenannten Alpha-Methode wird nur eine Testverdünnung benötigt, das Meßsignal korrigiert und dann über eine Rechenformel die IU/ml ermittelt. Diese neue Einpunktquantifizierung wurde in sieben unabhängigen Arbeitsgruppen durch Vergleich mit kommerziellen EIA's bewertet, die eine Standardkurve oder Titerbezugsgerade verwenden. Zur IgG-Quantifizierung bei Rubella wurden insgesamt 1480 einzelne Untersuchungsproben und drei Vergleichs-EIA's eingesetzt. Beim IgG gegenToxoplasma gondii waren es über alles 743 Proben und zwei Vergleichstests. Die mittels Alpha-Methode erzielten Ergebnisse besaßen eine voll praxistaugliche Präzision und Richtigkeit. Zusätzlich wurden der technische Aufwand und die Reagenzienkosten reduziert. Voraussetzungen und Einschränkungen werden vor dem problematischen Hintergrund einer Immunstatusdefinition diskutiert.
    Notes: Summary Antibody quantification by EIA is possible without a standard curve. Following the so-called alpha method only one test dilution is used, the resulting absorbance is corrected and the IU/ml will be calculated by means of a mathematical formula. This new kind of a single point measurement was evaluated in seven independent laboratories by comparison with commercial EIAs using a standard curve or a titer calibration line. For the quantification of IgG against rubella virus this study comprised 1,480 individual samples and three comparison EIAs. For IgG againstToxoplasma gondii a total of 743 samples was evaluated in two comparison tests. The results obtained by the alpha method show a precision and accuracy more than sufficient for routine testings. Also the technical expenses and reagent costs were reduced. Prerequisites and limitations are discussed against the background of the problem of immune status definition.

    Notes: Summary Eleven cistrons of genes active in lytic growth of phage P22 were classified by phenotypic expression of their amber mutants in nonpermissive Salmonella typhimurium. Seven cistrons code for late functions according to their lysis positive phenotype. Of the remaining four cistrons one codes for lysozym synthesis, two for phage directed DNA synthesis and one seems to be engaged in regulation of late gene expression.

    Notes: Summary Phage mutants were isolated with amber mutations in genes necessary for establishment of lysogeny. These mutants form turbid plaques on su + strain 527R1 and clear plaques of different types on LT2. According to complementation tests, fourteen mutants fall in the c 2 gene, four in the c 3 gene but no amber mutants were found belonging to the c 1 gene. Pulse labelling experiments to follow DNA synthesis after phage infection were done with the mutants classified by complementation tests. Furthermore the labelling experiments demonstrated that the nonleaky c 3 amber mutants displayed the same DNA synthesis pattern as c 1 missense mutants. Since these c 3 amber mutants complement missense c 1 mutants it is concluded that the c 3 and c 1 genes must act together for the first transient repression of DNA synthesis, i.e., seven minutes after infection. It is suggested that clear plaque forming c 1 amber mutants cannot be isolated because of polarity leading to defectivity of lysogenic as well as of lytic functions.
