    Notizen: Alkaline phosphatase was used in an immuno-enzymatic procedure to detect tissue-bound and circulating antibodies in pemphigus vulgaris. Pemphigus antibodies were revealed by a direct method using alkaline phosphatase conjugated goat anti-human IgG. Cryostat sections were incubated with the specific antiserum, and alkaline phosphatase activity was then revealed histochemically either by Gomori's technique or by the azo dye method. The sections were examined by light microscopy and intercellular staining was demonstrated in the epidermis. The indirect method, in which the patient's serum was incubated with sections of normal skin, gave similar results. Using parallel sections, an immunofluorescent technique was used to demonstrate tissue-bound and circulating antibodies. The alkaline phosphatase method appeared to be slightly less sensitive than the immunofluorescent method.

    Notizen: Résumé Un agent stimulant la reproduction des virus a été trouvé dans les liquides des œufs fécondés, non-infectés. Cet agent a stimulé la multiplication des virus Semliki Forest jusqu'à 105 fois dans les cellules L929. Nous avons constaté que le méchanisme de ce phénomène stimulant n'est pas basé principalement sur l'effet suppressif de l'interféron endogénique.

Die meisten ribonukleasereichen Peritonealzellen haften nicht an plastischen Oberflächen. Die Ribonuklease-Aktivität von nichtadhärenten Zellen wird erhöht, wenn Mäuse mit Cortison behandelt werden, und diese Wirkung ist besonders auffallend im Gesamtbestand der peritonealen Zellen.

    Notizen: Abstract Sera of mice immunized with ribosomal fractions of Candida albicans showed the presence of anti-C. albicans antibodies, detected by the gel-immunodiffusion, agglutination and immune adherence tests. Candida infections are among the most prevalent opportunistic yeast infections, attacking debilitated individuals, and against which there is no effective prophylactic treatment currently available (1, 2, 3, 4, 5). In view of the succes reported in experimental immunizations with ribosomal fractions from various bacteria and some fungi, as summarized by Youmans and Tewari (7, 8), a similar approach for immunization in experimental candidiasis appears reasonable. The present work describes preliminary results on circulating antibodies elicited in the course of immunizations with ribosomal fractions of Candida albicans. Ribosomal preparations were obtained from mechanically disrupted cell-pellets of C. albicans by differential centrifugation and purification in a 15% sucrose and 5% ammonium sulfate solution (in sodium-magnesium-Tris buffer), using a modification of the procedure described by Rubin (6). Concentration of ribosomal-RNA was determined by the absorbance at 260 nm; ribosomal-protein concentration by the Lowry reaction; and purity of the ribosomal preparation checked by the ratio of absorbance at 260 nm to 280, and at 260 to 235 nm. Mice (ICR strain) were immunized with these ribosomal preparations in amounts of 50–100 μg ribosomalprotein/mouse, by 2–3 subcutaneous inoculations with Frend's adjuvant, with a 10–21 day interval between the inoculations.

    Notizen: Abstract Enzyme linked immunosorbent assay was found to be a convenient method for the investigation of antibodies in mice immunized with Candida albicans ribosomes. Antibodies against the ribosomal antigen were detected in all the sera of mice (ICR and BALB/ c) immunized with ribosomes and incomplete Freund's adjuvant and in some of the sera of mice immunized with ribosomes only; the titer of antibodies varied from 1∶320 to 1∶10 240. Vaccination of mice with ribosomal protein and IFA resulted in a high titer of antiribosomal antibodies. Treatment of ribosomes with pronase abrogated the capacity of the ribosomes to elicit anti ribosomal humoral responses, suggesting that the antibodies detected were directed against the protein moiety of the ribosomes. The presence of antibodies in sera of immunized mice could not be correlated with the protection afforded by the ribosomal vaccination.

    Notizen: Abstract Fifty samples of human milk obtained on the third day after delivery were analysed for antibody titers against E. coli antigens, using the passive haemagglutination test (PHA). Antigens were prepared from two “enteropathogenic” E. coli strains (0111:B4 and 0.26:B6) and one “saprophitic” strain (088) by two methods of extraction for each bacterial strain: 1. with water at 100°C, and 2. with veronal buffer, pH 8.4. The milk samples were classified into three groups: after the first parturition [17], after the second parturition [14], and after the third or subsequent parturition [19]. The mean antibody titer was highest in the first group and lowest in the third group. The immunoglobulins in the milk samples, characterized by the radial immunodiffusion test, were found to be IgA and IgM. The concentration of IgA was relatively higher than that of IgM in all the groups; no IgG was detected. The third group showed the lowest concentrations of immunoglobulins. In an attempt to characterize the type of anti-E. coli antibodies, several samples were fractionated on DEAE-cellulose. Using the PHA test, it was possible to demonstrate that only the IgA fraction contained E. coli antibodies. These results suggest that the anti-E. coli antibodies in human milk are mianly of the IgA type and that a decrease of immunoglobulin concentration can be observed after three or more parturitions.
