    Notes: Abstract – This study examines the frequency of oral disease in an adolescent population, and assesses the relationship to Actinobacillus actinomycetemcomitans. A total of 470 eighth grade students from San Antonio, Texas, were examined clinically for number of teeth, frequency of gingival inflammation, frequency of sites with BOP. and frequency of sites with 3-5 mm pockets, and pockets 〉5 mm. The population ranged in age from 12 to 17 yr and was 93% Hispanic. Heavy accumulations of plaque and calculus were frequently observed and were associated with gingival inflammation, as 95.6% of the students exhibited bleeding on probing, and 99.6%. of the students presented with at least on quadrant of inflammation upon visual examination. Significantly, 25.7% of the students exhibited early-onset periodontitis (HOP) with 1.7% diagnosed as LJP. Many students exhibited substantial levels of plaque and calculus, but no clinical evidence of loss of attachment. Subjects with periodontitis (EOP or LJP) presented with elevated systemic IgG antibody to actinomycetemcomitans serotype b and subgingival plaque samples positive for the microorganism. These results describe the prevalence of EOP/LJP in an adolescent Hispanic population from South Texas. The findings support that actinomycetemcomitans may represent a pathogen in periodontitis and while oral health care may be poor, contact with the microorganism appears to be required to initiate disease in this population.

    Notes: A procedure was developed for the preparation of single cell suspensions from gingival and periodontal tissues using collagenase digestion followed by gentle mechanical disruption. The procedure was evaluated on rat gingival tissues. One-hour incubation was found to produce the greatest number of cells with the highest viability, and the largest recovery of mononuclear cells. Collagenase did not appear to effect recovery, viability or B cell surface markers on human peripheral blood cells. Human gingival and periodontal tissues were obtained from 35 patients: 20 with adult periodontitis; 8 with juvenile periodontitis; 7 with normal gingivae or chronic gingivitis. Scaling, followed by probing measurements and then surgery of the affected sites was routinely performed. In single cell suspensions from tissue obtained from surgery, significantly fewer mononuclear cells were recovered from the tissues of normal/chronic gingivitis patients than cells/mg of tissue recovered from adult periodontitis or juvenile periodontitis groups. The maximum contribution of blood mononuclear cells to the gingival cells averaged 0.5+0.2%. This method is useful for recovering gingival (periodontal) cells for phenotypic and functional studies.

    Notes: Stimulated parotid salivas, collected from 162 young adults, were screened in ELISA Tor IgA antibody to Actinobacillus actinomycetemcomitans. Subjects with elevated levels of IgA antibody (HSA) were age- and sex-matched with subjects with low antibody levels (LSA) for subsequent study. Infection with A. actinomycetemcomitans, elevated serum antibody levels to A. actinomycetemcomitans, and presence of multiple sites 〉5 mm were generally found only in the HSA group. There was a significant association between the presence of deep pockets and elevated serum antibody in these subjects. Thus, screening saliva for IgA antibody may be a useful, noninvasive technique to identify individuals with the potential to develop periodontal disease or those who are currently responding to an infection with A. actinomycetemcomitans.

    Notes: The cellular and humoral responses in ovalbumin (OVA) presensitized germfree rats fed OVA (SAF), in an otherwise antigen-free diet, were compared to sham-sensitized animals that were also fed OVA (SSAF). Sensitized animals demonstrated relatively more lymphocytes in their gingival tissues. An initial predominance of T lymphocytes gave way to a predominance of B-lymphocytes. Splenic lymphocytes from presensitized animals showed significant blastogenesis at the time antigen feeding was initiated. Sensitized animals showed elevated blastogenesis throughout the feeding period whereas the sham-sensitized animals showed some elevated responses only after 38 days of antigen feeding. The plasma IgG response to OVA was markedly elevated in the sensitized animals, in contrast to a low or nonexistent IgG antibody response in the sham-sensitized rats. On the other hand, IgA anti-OVA responses in intestinal perfusates were elevated in the SAF group for the first 14 to 21 days of feeding. Early salivary IgA responses (0 to 21 days) were essentially the same between the groups, but later (38 to 84 days) both IgA and IgG responses in saliva were elevated in the SAF group. Sensitized animals demonstrated significantly elevated periodontal bone loss after 84 or 128 days of OVA feeding. The potential effects of “mixed” hypersensitivity on periodontal disease in the rat are discussed.

    Notes: The immune response, gingival inflammation and periodontal bone loss were examined in rats receiving adoptive transfer of Actinobacillus actinomycetemcomitans (Aa)-sensitized T cells and oral infection with Aa. Thirty-eight, 30-day-old, male Rowett phenotypically normal (rnu/+) heterozygous rats were placed into three groups: 1) Aa-sensitized T cell transfer group (AaT), 2) sham-sensitized T cell transfer group (ST) and 3) no transfer control group (C), infected with Aa, and then sacrificed at 120 d of age. Blastogenesis of splenocytes to PHA and Con A was observed in AaT and ST animals that exceeded the values in C animals. Positive antigenic lymphoproliferation and delayed-type hypersensitivity (foot pad swelling) to Aa were found in AaT group rats, but weak responses were observed in ST and C group rats. Serum IgG, salivary IgA and IgG antibody concentrations to Aa were elevated in AaT and ST groups compared with the control group after infection with Aa. The numbers of gingival T and B lymphocytes in the AaT group were increased in comparison to the other groups. There was also an obvious increase in horizontal bone loss in the AaT group rats. It is suggested that certain subsets of antigen-specific T cells may augment destructive functions on periodontal tissues.

    Notes: A rapid method for collection of static crevicular fluid (SCF) on filter papers is described. Standardization of a Periotron® for volume quantitation of the SCF allowed accurate collection of crevicular fluid samples from periodontal disease patients. A method for elution of SCF was developed and was shown to provide nearly total recovery of antibody activity from the filter papers. Analysis of specific antibody to suspected periodontal pathogens in eight individuals indicated a site-to-site variation in antibody levels within the oral cavity. Also, SCF from certain sites exhibited antibody levels that were significantly greater than serum, supporting the concept of specific local antibody synthesis in gingival tissues.
