    Notes: Summary A line of flax, homozygous for four genes controlling resistance to flax rust, was transformed with T-DNA vectors carrying the maize transposable elements Ac and Ds to assess whether transposition frequency would be high enough to allow transposon tagging of the resistance genes. Transposition was much less frequent in flax than in Solanaceous hosts such as tobacco, tomato and potato. Transposition frequency in callus tissue, but not in plants, was increased by modifications to the transposase gene of Ac. Transactivation of the excision of a Ds element was achieved by expressing a cDNA copy of the Ac transposase gene from the Agrobacterium T-DNA 2′ promoter. Progeny of three plants transformed with Ac and 15 plants transformed with Ds and the transposase gene, were examined for transposition occurring in the absence of selection. Transposition was observed in the descendants of only one plant which contained at least nine copies of Ac. Newly transposed Ac elements were observed in 25–30% of the progeny of some members of this family and one active Ac element was located 28.8 (SE=6.3) map units from the L 6 rust-resistance gene. This family will be potentially useful in our resistance gene tagging program.

    Notes: Summary A pathway for biosynthesis of the crown-gall opine, agropine is proposed and three potential new precursors characterised. The location of genes involved in the three steps of this pathway was determined by site directed insertions and deletions in the TR region of the octopine Ti plasmid, pTiB6Trac. The proposed biosynthetic pathway for agropine which involves three T-DNA genes is in contrast to the biosynthesis of octopine and nopaline where single T-DNA genes are involved.

    Notes: Summary The conjugative behaviour of nopaline and agropine Ti-plasmids has been investigated. Using a technique which avoids enrichment of transconjugants on a mating medium we have shown that preculture in the presence of agrocinopines A or B of donor strains harbouring nopaline Ti plasmids promotes plasmid transfer whereas preculture of the same strains in the presence of nopaline has no such effect. Similarly, preculture in the presence of agrocinopines C or D promotes Ti-plasmid transfer from strains harbouring agropine Ti-plasmids.

    Notes: Summary We report four new opines that have been discovered in sterile crown gall tumour tissue. These compounds have been partially characterised as phosphorylated sugar derivatives. Two were detected in tumours induced by nopaline strains of Agrobacterium tumefaciens and two in tumours induced by an agropine strain. These compounds are involved in interesting interactions with the nucleotide bacteriocin, agrocin 84 and we propose that they be called agocinopines.

    Notes: Summary Conjugal transfer of the nopaline Ti-plasmid pTiC58 is inducible by the phosphorylated opines, agrocinopines A and B. Although the uninduced level of transfer is negligible (〈 10−7 per donor), some transconjugants have been isolated from crosses performed in the absence of agrocinopine. These transconjugants harbour mutant Ti-plasmids that transfer constitutively (〉10−3 per donor). These mutants have several other correlated phenotypes including constitutive uptake of agrocinopine A, supersensitivity to agrocin 84 and the ability to prevent the excretion of agrocin 84 when they are present in the same cell as the agrocin 84 biosynthetic plasmid pAt-84a.
