Summary 1. The present study contains investigations on the effect of erythropoietin and somatotropic hormone on the maturation of reticulocytes from rabbitblood with a high number of reticulocytes. 2. The reticulocytosis was produced by means of repeated bleeding, the erythrocytes were washed and incubated in a standardized medium under reproducible conditions for over 12 hours. Samples of the reticulocyte suspension were taken in regular intervals, and the number of reticulocytes as well as the reticulocytogram (Percentage of the different maturation stages-O-IV according to Heilmeyer- in the total reticulocyte count were determined. 3. The maturation of the reticulocytes was significantly accelerated under the influence of 20 and 50 units of erythropoietin/4,5 ml cell suspension, the reticulocytogram was not affected in comparison to the control experiments. 4. Somatotropic hormone had no effect on the maturation time of the reticulocytes, i.e. the reticulocytogram in a dose of 2,10 or 40 units/4,5 ml cell suspension. 5. The results demonstrate that erythropoietin affects not only the stem cells of the erythropoiesis and the cells of the erythron, but also the anuclear, direct precursors of the erythrocyte. The biochemical principles of the procedure are discussed.

Zusammenfassung 1. Die vorliegende Arbeit enthält Untersuchungen über den Einfluß von Erythropoietin und somatropem Hormon auf die Reifung der Retikulozyten aus retikulozytenreichem Kaninchenblut. 2. Das retikulozytenreiche Blut wurde durch wiederholte Aderlässe erzeugt, die Erythrozyten wurden gewaschen und in einem standardisierten Medium unter reproduzierbaren Bedingungen über 12 Stdn. inkubiert. In regelmäßigen Abständen wurden Proben der Retikulozytensuspension entnommen und die Retikulozytenzahl sowie das Retikulozytogramm (Anteil der einzelnen Reifungsstadien-0-IV nachHeilmeyer-an der Gesamtretikulozy tenzahl) bestimmt. 3. Die Reifung der Retikulozyten war unter der Einwirkung von 20 und 50 Einheiten Erythropoietin/4,5 ml Zellsuspension deutlich beschleunigt, das Retikulozytogramm blieb gegenüber den Kontrollansätzen unbeeinflußt. 4. Somatropes Hormon hatte in einer Dosierung von 2, 10 oder 40 Einheiten/4,5 ml Zellsuspesion keinen Einfluß auf die Geschwindigkeit der Retikulozytenreifung bzw. auf das Retikulozytogramm. 5. Die Ergebnisse beweisen, daß Erythropoietin nicht nur an den Stammzellen der Erythropoiese und den Zellen des Erythrons, sondern auch an den kernlosen unmittelbaren Erythrozyten-Vorstufen angreift. Die biochemischen Grundlagen dieses Vorganges werden diskutiert.
