    Notes: The sensitivity of the immunofluorescence microphotometry method for the demonstration of antibodies against platelets was compared with that of the agglutination reaction, the indirect antiglobulin consumption test, and the complement fixation method. For this purpose titres of sera containing hetero- or iso-antibodies were measured by various methods, with of the same donor. For the immunofluorescence method an anti-rabbit immunoglobulin conjugate, and an antihuman immunoglobulin conjugate, anti-human IgG conjugate, and an anti complement conjugate were used. Fixation of platelets with acetone appeared to be unsuitable for the immunofluorescence complement method On the whole, the highest titres were obtained with immunofluorescence microphotometry.

    Notes: The standardization is described of an immunofluorescent microphotometric method that offers a solution for the problems generally encountered in immunofluorescence procedures on platelets. Fluorescein isothiocyanate (FITC) conjugated .anti-immunoglobulin serum and anti-IgG serum were used. Sera, containing isoor hetero-antibodies against platelets that had given positive reactions with one or more of the conventional serological techniques, were used as positive controls. The method described proved to have a good reproducibility. Significant differences in fluorescence were obtained using the above-mentioned positive sera and normal control sera. It was found that platelets stored in sodium-azide could be used as antigen. A number of variables that may influence the difference in fluorescence obtained with positive and normal sera were studied.

    Notes: After a survey of the literature dealing with the demonstration of platelet autoantibodies by immunofluorescence techniques, the results are given of a study in which immunofluorescence microphotometry was used for this purpose. The serum of 58, the platelets of 34, and the megakaryocytes of. 2 patients -with thrombocytopenia were investigated. In 21 of 52 sera (40%) in which the presence of platelet autoantibodies could be expected, positive results were obtained that could not be due to isoantibodies, either because the patients had not been pregnant and had not received blood transfusions or because the reactivity of the serum with the patient's own platelets was demonstrated. The platelets of 28 patients with thrombocytopenia not due to a platelet defect or decreased thrombopoiesis were investigated. In the platelets of 15 (54%) of these, significant differences in fluorescence were'found with anti-immunoglobulin conjugate as well as with anti-IgG, -IgA, -IgM, or -complement reagents. It was concluded that in these patients in vivo sensitization of the platelets with autoantibody was demonstrated. In two patients an indication of the in vivo sensitization of the megakaryocytes was also obtained.

    Notes: The purpose of this study was to characterize the destruction of sensitized erythrocytes by human blood monocytes in vitro The incubation in vitro of human monocytes with 51Cr-lalbelled human erythrocytes sensitized with IgG rhesus alloantibodies anti-D EAIgG anti-D) resulted in release of51Cr from the erythrocytes (lysis) as well as uptake of 51Cr-labelled erythrocytes by the monocytes (phagocytosis). The lysis of EAIgG anti-D by monocytes was not dependent on phagocytosis, because cytochalasin B, which inhibited phagocytosis of EAIgG, enhanced lysis. In contrast, hydrocorlisone and colchicine inhibited lysis, but had no effect on phagocytosis. These agents did not affect binding of EAIgG anti-D to monocytes. The effect of these agents on lysis corresponded to their effect on release of lysosomal enzymes by monocytes. The release of lysosomal enzymes, when induced by EAIgG anti-D, was, likewise, enhanced by cytoehalasin B and inhibited by hydrocortisone and colchicine. A significant correlation was found between lysosomal enzyme release and lysis. Together, these results strongly suggest that lysosomal enzymes. released by the monocytes when incubated with anti-D-sensitized erythroeytes, are responsible for the cytotoxic activity of these cells towards sensitized erythrocytes. The action of these enzymes only occurs over a short range, probably at the site of attachment of the erythrocyte, becuuse only erythrocytes that were bound to the monocytes were lysed. The finding of other investigators that removal of monocytes from suspensions of human mononuclear leucocytes results in a strong reduction in the cytotoxic activity of these leucocytes towards sensitized erythrocytes in vitro. was confirmed.

    Notes: In view of the importance of the species of animals used fat the preparation of specific heteroantisera against human blood cells, comparative immunization experiments Were carried out in goats, rabbit, chickens, guinea pigs, and rats, using lysates leukocytes, granulocytes, lymphocytes, monocytes, and thrombocytes. The antisera obtained were tested by the indirect immunofluorescence technique. The strongest antisera were obtained in goats, but these antisera required extensive absorptions to make them specific for the immunizing cell, whereas rabbit antisera needed only few absorptions. Antisera from both of these animals could be rendered specific for lymphocytes, granulocytes and platelets. In the guinea pig only immunization with lymphocytes and granulocytes resulted in useful antisera, which were all specific for granulocytes after appropriate absorptions. Antisera obtained from rats could be rendered specific for granulocytes and lymphocytes by absorption. Antibodies against platelets could not be detected in any of the rat antisera. Chickens, however, produced strong anti-platelet anti-bodies. A specific anti-granulocyte antiserum could also be obtained in these animals. Specific anti-monocyte antisera could not be prepared from any of the animal sera. Immunizations with leukocyte lysate resulted in granulocyte-specific antisera in most animals but not in chickens. All antisera produced in chickens were specific for platelets. It can be concluded from these results that animals of different species may react very differently to the antigens of some human blood cells.

    Notes: An analysis of the HLA data of 100 myasthenia gravis (MG) patients and their first degree relatives and spouses is presented. The family studies confirmed the excess of B8 in the myasthenia gravis patients and showed that the excess was not limited to the A1, B8 haplotype. An excess of B8 homozygous patients was found from the matings of two B8 heterozygous parents, suggesting the existence of a dominant B8 linked MG susceptibility. B8 homozygous individuals run a 4.7 times higher risk of acquiring myasthenia gravis than B8 heterozygous individuals.

    Notes: Of 118 Dutch patients suffering from ankylosing spondylitis (AS) 81.4% were found to be positive for the HLA antigen B27. The B27 frequency proved to be significantly higher in patients in whom the disease had an early onset.In addition to B27, another HLA antigen may be associated with AS; the antigen Bw16 was found to be significantly increased in B27 negative AS patients.HLA phenotype frequencies were also determined in 109 patients with idiopathic inflammatory bowel disease (IBD). In fifty-eight ulcerative colitis (UC) patients a raised incidence of A11 was noticed. In fifty-one patients with Crohn's disease (CD) the antigen B18 showed an increased frequency. Both deviations were statistically significant. In thirty-nine patients suffering from both AS and IBD 50% proved to be B27 positive, which is significantly different from the B27 frequency in patients with AS alone. In the B27 negative patients with AS and IBD an increased frequency of Bw16 was also shown.

    Notes: Summary An extended family is described in which three members had thrombocytopenia. These affected members had chromosomal changes resembling those found in Fanconi's anaemia, though they lacked the development defects associated with that syndrome. One had bone-marrow hypoplasia and died of squamous cell carcinoma of the mouth at the age of 27. In addition, all three had platelet autoantibodies not found in any other family members tested. There was no linkage between the thrombocytopenia and HLA groups. The nature of the association of thrombocytopenia, platelet autoantibodies and chromosomal abnormalities in this family remains doubtful.
