    Notes: Abstract: Presynaptic correlates of evoked neurotransmitter release include a rise in cytosolic free calcium level and the calcium-dependent liberation of unesterified arachidonic acid. It has been proposed that lipoxygenase metabolites produced from arachidonic acid may constitute an endogenous feedback system for the modulation of neurotransmitter release. The results of the present study are in agreement with this hypothesis. It was demonstrated that membrane depolarization evoked the release of endogenous glutamate from hippocampal mossy fiber synaptosomes, as well as the accumulation of intraterminal free calcium. The presence of 12-lipoxygenase products attenuated both the induced release of glutamate and the increase in calcium content, whereas 5- or 15-lipoxygenase metabolites were ineffective. A role for lipoxygenase products in the negative modulation of mossy fiber secretion processes was further indicated by the observations that low concentrations of the lipoxygenase inhibitor nordihydroguaiaretic acid (0.1–10 μM) potentiated the glutamate release and calcium accumulation induced by membrane depolarization. Therefore, we suggest that 12-lipoxygenase metabolites provide a presynaptic inhibitory signal that limits neurotransmitter release from hippocampal mossy fiber terminals.

    Notes: Background : Although serology is the main Helicobacter pylori test used by general practitioners in the UK, there is no information available on variation in requesting rates.Aim : To explore the reasons for any variation in H. pylori serology testing by general practices in the UK using qualitative methods.Methods : Serology requesting rates were determined using laboratory and population data. Staff from randomly selected practices in the lowest and highest quintiles of testing attended focus groups to discuss the management of H. pylori and dyspepsia. Transcribed data were analysed using an interpretative phenomenological approach.Results : Serology submission varied 600-fold (0.1–59/1000 population/year) and H. pylori positivity rate 17–100%. Low-testing practices were less aware of the benefits of H. pylori testing and had shorter endoscopy waiting times. They preferred endoscopy diagnosis over serology test. Three high-testing practices had a high non-white population with high H. pylori positivity. Most staff knew little about the predictive value of serology, the availability of urea breath test on prescription or the existence of a stool test.Conclusions : Seroprevalence of H. pylori is still high in dyspeptics, especially in non-white populations. Laboratories and primary care trusts should audit H. pylori requests and endoscopy referrals, target education at high endoscopy referrers and low H. pylori testers and inform clinicians of the more accurate H. pylori tests and NICE dyspepsia guidance.

    Notes: Sclera was obtained from the eyes of New Zealand white rabbits and immediately stored at –79°C. Later, after thawing, a piece 4 mm square was implanted into each of the anterior chamber of 14 eyes and under the renal capsule of 28 kidneys of rabbits of the same strain, Allografts of skin, muscle or tendon were implanted subcapsularly in the contralateral kidneys as controls. Two animals were sacrificed postoperatively each week up to 4 weeks, 6 animals at 6 weeks, 4 each at 8 and 9 weeks and 6 at 12 weeks. Serial paraffin sections of the implants and host tissues were examined by light microscopy. Osteogenesis occurred in 7 of 12 implants of sclera implanted beneath the renal capsule at 6 and 12 weeks, but was not seen prior to 6 weeks or at 8 or 9 weeks. Osteogenesis did not occur in sclera implanted into the anterior chamber of the eye or in control implants.

    Notes: The value of autogenous marrow as a bone graft material was examined. Holes ∼ 2 mm in diameter were made in. the right and left parietal bones of 9 middle –aged Wistar-strain albino rats weighing ∼ 500 g. Bone marrow was obtained from the medullary cavity of the femur and immediately placed into the left hole, while the right served as a control. All animals were killed 12 weeks postoperatively. Their calvaria were examined radio graphically and by routine histological methods. It was found that whereas the control defects never healed, those grafted with autogenous marrow were consistently filled with a bone ossicle. These findings suggest that bone marrow grafts, may be of value in periodontal therapy.

    Notes: Osseous coagulum is regarded by some clinicians as a useful material for grafting in procedures aimed at bone replacement. Its value in this regard has been assessed experimentally. Holes ∼ 2 mm in diameter were made in the right and left parietal bones of 15 middle-aged Wistar-strain albino rats weighing ∼ 500 g. Osseous coagulum was obtained from the interparietal bone, and was placed into the left hole, while the right served as a control. Three of the animals were injected with tetracycline; these animals were sacrificed at 4, 8 and 12 weeks postoperatively, and undemineralized sections of their calvaria were viewed in ultraviolet light to assess accretion of new bone. The remaining 12 animals were sacrificed at postoperative intervals of 1, 2, 3, 4, 8 and 12 weeks, and serial sections of their calvaria were examined by routine histological methods. The observations indicate that, in this system in which repair is normally poor, osseous coagulum consistently did not appear to have induced the formation of new bony callus. These findings bring into question the value of using osseous coagulum as an autograft in clinical periodontal procedures aimed at restoring the attachment apparatus.
