    Notes: Abstract  Polymorphism of the HLA-G gene in a Japanese population was investigated employing polymerase chain reaction (PCR)-single-strand conformation polymorphism (SSCP) analysis, PCR sequence-specific oligonucleotide (SSO) analysis, and DNA direct sequencing. Nucleotide sequence variations in exons 2, 3, and 4 of the HLA-G gene in 54 healthy Japanese individuals were examined. In addition, seven Japanese samples carrying common HLA haplotypes were analyzed. In total, nine single-base substitutions compared with the sequence of G * 01011 were identified: one in intron 1 (nucleotide position 970), one in exon 2 (the third base of codon 57: G → A), three in intron 2 (1264, 1276, and 1292), three in exon 3 (the third base of codon 93: C → T, the third base of codon 107: A → T, and the first base of codon 110: C → A), and one in intron 3 (2334). The substitution at codon 110 was non-synonymous and led to an amino acid substitution from leucine to isoleucine. The other three nucleotide substitutions in exons were synonymous. Through analysis of combinations of the exon 2, 3, and 4 nucleotide sequences we identified four alleles, which we provisionally designated GJ1, GJ2, GJ3, and GJ4. The allele frequencies were estimated to be 0.33, 0.16, 0.45, and 0.06, respectively. Nucleotide sequences of GJ1, GJ2, and GJ4 were identical to G * 01011, the clone 7.0E, and G * 01013, respectively. GJ3 was a newly observed allele and was officially designated G * 0104 by the WHO Nomenclature Committee in January 1996. Strong positive associations were observed between HLA-G alleles and HLA-A, -B, or -DRB1 alleles.

    Notes: Molecular dynamics simulation is conducted to investigate the effect of notch depth onthe deformation and fracture behavior of a single crystal copper which is expected to a conductivematerial of micro-devices. In the stress – strain relationship, a normal stress increases withincreasing applied strain. Then, the normal stress decreases rapidly. When the stress decreases, thedislocation emits from a notch root and the stacking fault is formed on the {111} plane, which isslip plane of the fcc crystalline structure. The maximum stress decreases with notch depth. Thenon-damaging defect size is quite small. The shear stress in the slip direction at dislocation emissionis constant irrespective of the notch depth. The criterion of the dislocation emission is given by thecritical value of the resolved shear stress in the sliding direction

    Notes: Agar/gelatin mixed gels with the same composition but with a different two-phase disperse structure were prepared and their mechanical properties compared. The agar/gelatin mixture was first kept at temperature above the gelling temperature of gelatin but below that of agar and stirred for the selected period, before cooling it below the gelling temperature of gelatin. For the low rupture stress system the agar concentration was 0.7% (w/w), while the gelatin concentration was 4.5% (w/w) to achieve the same rupture stress as the agar gel. The mixing temperatures selected were 20 and 37C. For the high rupture stress system, the agar and gelatin concentration was 2.8 and 10.4% (w/w), respectively, to achieve the same rupture stress. The mixing temperatures selected were 37 and 40C. The both mixed gels prepared by this method consisted of a dispersed phase of agar and a continuous phase of gelatin. The rupture stress of the mixed gels decreased as the content of the dispersed phase increased. The rupture stress had a tendency to be lower as the size of the dispersed particles increased. These results suggest that the interface between the dispersed phase and the continuous phase plays an important role as Griffith's crack, with the rupture of mixed gels occurring from that place.

    Notes: Abstract The polypeptide encoded by the partial fragment of cDNA of phenylalanine ammonia-lyase (PAL; EC 4.3.1.5), PALcDNAl (Osakabe et al., 1995, Plant Sci. 105: 217–226), isolated from Populus kitakamiensis (P. sieboldii x P. grandidentata), was expressed in Escherichia coli cells. The polypeptide was purified and an antiserum raised against it. The antiserum recognized a protein of 77 kDa on nitrocellulose blots after sodium dodecyl sulfate-poly-acrylamide gel electrophoresis of total protein and the partially purified PAL protein from P. kitakamiensis. Moreover,the antiserum recognized a protein on the blot after non-denaturing polyacrylamide gel electrophoresis of P. kitakamiensis proteins and this protein had PAL activity. Furthermore, the antibody inhibited PAL activity of extracts from stem tissues. These results showed that the antiserum against the partial PAL peptide recognized only the PAL subunits in extracts of P. kitakamiensis. Immunolocalization studies of P. kitakamiensis tissues revealed that the PAL protein was specifically localized in the xylem and the phloem fibers and no immunogold signal was found in the epidermis, the cortex, the pith, or the cambium of either stems or leaves.

    Notes: Continuous production of L-alanine with conjugated enzyme systems of alanine dehydrogenase (AlaDH) and lactate dehydrogenase (LDH) or alcohol dehydrogenase (ADH) was carried out with NAD regeneration in an ultrafiltration hollow-fiber capillary reactor (HFCR) which was proposed as a test bioreactor with very small scale. In the AlaDH/LDH system, pyruvate is the intermediate product for L-alanine so that an optimal point existed in pyruvate concentration for the production rate of L-alanine. NAD cycling number of 4850 and L-alanine productivity of 61.7 mmol/L h were obtained at the best condition. In the AlaDH/ADH system, however, the substrate inhibition in the AlaDH reaction by pyruvate should be considered and the best results of NAD cycling number and L-alanine productivity were 2700 and 13.5 mmol/L h, respectively. In consideration of concentration distribution and mixing in the axial direction on an HFCR, performance of the reactor was theoretically analyzed with a multistage stirred tank reactor model combined with the kinetic model based on all the elementary reactions involved. Although quantitative discrepancy existed in some cases, the present theoretical model could explain experimental results and is expected to be generally applicable to standard hollow fiber reactors.

    Notes: A polyelectrolyte complex gel was prepared by mixing chitosan and k-carrageenan solutions in the presence of NaCl. To study the effect of ambient pH on the swelling behavior, the diameter of the cylindrical gel(4 mm x 8 mm) immersed in aqueous NaOH, KOH, or HCl solution was measured. In the range of pH 10-12, the diameter increased with time, and the gel reached swelling equilibrium within 6 days. In an NaOH solution of pH 10.5, the maximum swelling occurred, and the volume of the gel at equilibrium was 10.2 times as large as the initial one. At pH below 9 and at pH 13, no swelling was observed. Thus, the swelling of the complex gel prepared in this study was sensitive to a rather narrow range of pH. The swelling equilibrium in the presence of NaCl was also investigated at various pH. © 1993 John Wiley & Sons, Inc.
