    Notizen: Effects of starter cultures, Lactobacillus sake and Pediococcus acidilactici, on the survival of Yersinia enterocolitica were studied in Turkish dry sausage (sucuk). Inoculated Y. enterocolitica (approximately 5.0 log CFU/g) was eliminated completely in sausages by L. sake (about 7.0 log CFU/g) and P. acidilactici (about 7.0 log CFU/g) by the 3rd d of fermentation but was not totally eliminated in natural fermentation without starter(s) after completion of 4 d of fermentation and 12 d drying (p 〈 0.05). pH decreased from 6.3 to about 4.7 in starter culture treatments and from 6.3 to 5.6 in natural fermentation after completion fermentation and drying (p 〈 0.05).

    Notizen: : We have developed and tested a 1-step resuscitation thin agar layer (TAL) system (nonselective agar overlaid on selective agar) to effectively recover heat-, acid-, cold-, and salt-injured foodborne pathogens in liquid foods. Each TAL unit was designed for the recovery of one injured population. To improve operation efficiency we have developed a single 4-compartment Petri dish to simultaneously recover heat-injured Escherichia coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium, and Yersinia enterocolitica. No significant difference occurred between the single recovery unit and the 4-compartment unit for enumeration of 4 heat-injured pathogens (P 〉 0.05). This 4-compartment TAL system is at least 4 times more efficient than the single TAL system.

    Notizen: Membrane fractions obtained from Escherichia coli, Gluconobacter oxydans, and Acetobacter xylinum significantly stimulated rapid growth of Listeria monocytogenes (strains 101, 103, and Scott A) under atmospheric conditions of the OmniSpec Bioactivity System. L. monocytogenes demonstrated a shorter lag phase and faster growth compared to culture systems without the membrane fractions as determined by shorter color detection times, the durations of time for a significant color change to occur. The growth stimulating effect increased as the concentrations of membrane fractions increased. The use of membrane fractions to recover facultative Listeria spp. also lowered the detection limit of the method to 〈 10 cells/ml by increasing small cell numbers to the detectable level (107 cells/m) faster under aerobic conditions. The application of membrane fractions with the OmniSpec Bioactivity System shortened the detection time for the bacteria by 0.5–10 h, depending on the specific membrane and initial cell concentrations, compared to the conventional OmniSpec method. This method is very useful as a tool for studying food microbiology. Membrane fractions produced in our laboratory effectively dissipated oxygen in Fraser broth and had growth-enhancing activities comparable to that of commercial OxyraseTM.

    Notizen: Microbial growth curves are essential components in microbiological studies and are modeled conventionally by nonlinear fitting to one analytical expression such as the modified Gompertz equation. This paper discusses the potential of artificial neural networks (ANNs) for modeling bacterial growth curves. These ANNs are efficient approximators for highly dimensional complex functions because of their high nonlinearity and tolerance to noisy data. Therefore, ANNs can provide great flexibility in developing generalized models by extracting the real behavior directly from the experimental data. Such models can be designed to include the effect of time as well as a multitude of parameters pertaining to experimental conditions. The approach was applied to modeling time-dependent growth curves of Escherichia coli O157:H7 as affected by sodium chloride concentration and of Shigella flexneri as affected by incubation temperature, pH, and initial count. The developed ANNs were able to approximate the experimental growth curves with high accuracy. The advantages as well as limitations of the proposed methodology are presented.

    Notizen: Growth of Arcobacter butzleri was evaluated in Brain Heart Infusion broth incubated aerobically, microaerobically, and with OxyraseTM supplementation (anaerobically). At initial concentrations of101 to 103 cells/mL, A. butzleri populations reached 7.5 to 8.0 log CFU/ml in 48h at 37C in OxyraseTM -supplemented broth. The organism quickly declined in the other two systems to undetectable levels during the initial 24h of incubation. Only moderate population levels (ca. 3 log CFU/ml) could be detected in aerobic and microaerobic systems after 56h incubation. Growth of five Arcobacter spp. strains was evaluated at 30C in Brucella-blood broth, modified Cary and Blair Transport Medium, and a biphase cultural system. Strains were inoculated at a level of ca. 103 cells/ml and incubated with and without OxyraseTM supplementation to the system for 36h. Both the Brucella-blood broth and the biphase system supported good growth of the strains, with counts reaching ca. 8 to 9 log CFU/ml. Modified Cary and Blair Transport Medium was the least effective cultural system. OxyraseTM provided slight to moderate stimulation of growth for most strains.

    Notizen: The effect of volume of liquid medium (Hunt broth) supplemented with Oxyrase® on growth of Campylobacter jejuni was evaluated by using four sizes of containers and two concentrations of Oxyrase® (0.15 units/mL and 0.6 units/mL). We used 13×100 mm screw-capped tubes containing 1, 3, and 6 mL of medium; 18x150 mm screw-capped tubes containing 1, 5, 10, and 20 mL of medium; 45x45x120 mm bottles containing 25, 50, and 100 mL of medium; and Stomacher® 400 bags containing 250 mL of medium. Counts of four strains of C. jejuni were determined on blood agar plates at 0, 6, 12, 20, and 28 h of incubation. For 13×100 mm tubes, 3 mL of broth resulted the highest bacterial counts with increases of 4–5 log CFU/mL at 12 h and 7–8 log CFU/mL at 20 h. For 18x150 mm tubes, 10 mL and 20 ml of broth both provided increases of 3–4 log CFU/mL and of 6–7 log CFU/mL at 12 h and 20 h, respectively. Increases of 3.5 log CFU/mL at 12 h and of 7–8 log CFU/mL at 20 h were found in bottles containing 100 mL of broth. Overall, 0.15 units/mL Oxyrase® enhanced Campylobacter growth as effectively as 0.6 units/mL in all containers with no significant differences (p〉0.05). However, supplementation with 0.6 units/mL of Oxyrase® gave slightly higher bacterial counts than addition of 0.15 units/mL in containers with high ratios of head space volume to medium volume (H:M). Ratios between 1.0 and 3.0 generally gave better results with all strains. The dissolved oxygen levels in each container were lower with high medium volume than with low medium volume. This study positively supported the efficiency of using Oxyrase® for recovery of C. jejuni under normal atmospheric conditions. In addition, various sizes of containers could be used for culturing campylobacters by maintaining the H:M ratio between 1.0 and 3.0.

    Notizen: Our improved method enables detection of catalase-related activities as a biochemical marker for estimating cooking end-point temperature (EPT). The broad range of inactivation temperature in chicken tissues was first determined to be 〉 68°C and 〈C 72°C. Then samples were heated to EPTs from 69 to 71.5°C at 0.5°C intervals. The catalase activity at 23 and 37°C was followed up to 120 min. The probability of obtaining a positive result with an EPT of 69°C was 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:00221147:JFDS494:ges" location="ges.gif"/〉0.99 after 45 min when incubated at 23°C. This probability decreased to 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:00221147:JFDS494:ges" location="ges.gif"/〉0.03 as EPT increased to 71°C. Higher incubation temperatures (37°C) increased the probability of positive results.

    Notizen: : The recovery of 5 pathogens from minced chicken (MC) and liquid whole egg (LWE) was determined with tryptone soya agar (TSA), selective medium, and thin agar layer (TAL; selective medium overlaid with TSA) to evaluate high-pressure effects at 200, 300, 400, and 500 MPa for 1 and 5 min at 20 °C. At 400 MPa for 5 min, Escherichia coli inactivation was 4 log in MC and 5.5 log in LWE, whereas no Salmonella Typhimurium, Yersinia enterocolitica, or Listeria monocytogenes were detected in either product. Staphylococcus aureus was the most pressure-resistant. In general, counts on the 3 media were not different. The TAL method allows differentiation of the inoculated pathogens from background microbiota.

    Notizen: : Pasteurized apple juice with CO2 (0, 1, and 4%) and cinnamon (0 and 0.3%) was inoculated with Escherichia coli O157:H7 at 104 CFU/mL, and stored at 5 and 20 °C. Counts on nonselective and selective media, and thin agar layer (TAL; selective medium overlaid with nonselective medium) were determined at 1 h and 1, 3, 7, and 14 d. Inactivation was greater at 20 °C. Samples with 1 and 4% CO2, alone and combined with cinnamon, presented 〈 0.7 log CFU/mL in 3 d. Counts in apple juice inoculated at 102 CFU/mL, a low-level E. coli O157:H7 contamination, were nondetectable at 3 d. The TAL method was as effective as nonselective medium to recover injured cells.
