    Notizen: Abstract: The relative roles of the superoxide and hydroxyl radicals in oxidative stress-induced neuronal damage were investigated using organotypic hippocampal slice cultures. Cultures exposed to 100 µM duroquinone, a superoxide-generating compound, for 3 h developed CA1-selective lesions over a period of 24 h. The damage accounted for ∼64% of the CA1 subfield, whereas CA3 showed just 6% damage, a pattern of damage comparable to that observed following hypoxia/ischaemia. Duroquinone-induced damage was attenuated by a spin-trap agent. In contrast, hydroxyl radical-mediated damage, generated by exposure to 30 µM ferrous sulphate for 1 h, resulted in a CA3-dominant lesion. The damage developed over 24 h, similar to that observed with duroquinone, but with ∼45% damage in CA3 compared with only 7% in CA1. These data demonstrate a selective vulnerability of the CA1 pyramidal neurones to superoxide-induced damage and suggest that of the free radicals generated following hypoxia/ischaemia, superoxide, rather than hydroxyl radical, is instrumental in producing neuronal damage.

    Notizen: A scheduling system is proposed and developed for a special type of flow shop. In this flow shop there is one machine at each stage. A job may require multiple operations at each stage. The first operation of a job on stage j cannot start until the last operation of the job on stage j - 1 has finished. Pre-emption of the operations of a job is not allowed. The flow shop that the authors consider has another feature, namely time lags between the multiple operations of a job. To move from one operation of a job to another requires a finite amount of time. This time lag is independent of the sequence and need not be the same for all operations or jobs. During a time lag of a job, operations of other jobs may be processed. This problem originates from a flexible manufacturing system scheduling problem where, between operations of a job on the same workstation, refixturing of the parts has to take place in a load/unload station, accompanied by (manual) transportation activities. In this paper a scheduling system is proposed in which the inherent structure of this flow shop is used in the formulation of lowerbounds on the makespan. A number of lowerbounds are developed and discussed. The use of these bounds makes it possible to generate a schedule that minimizes makespan or to construct approximate solutions. Finally, some heuristic procedures for this type of flow shop are proposed and compared with some well-known heuristic scheduling rules for job shop/flow shop scheduling.

    Notizen: Summary α-Crystallin is a major eye lens protein, composed of two types of subunits, αA and αB. The αA subunit is restricted to the lens, but αB-crystallin has recently also been detected in non-lenticular tissues, including the nervous system. With the use of a polyclonal antiserum directed against a synthetic C-terminal peptide of human αB-crystallin, the presence of αB-crystallin could be demonstrated immunohistochemically in astrocytes in the brains of patients with Creutzfeldt-Jakob disease (CJD). Most intensive localization was observed in the spongiotic tissue representing abundant progressively changed astrocytes in CJD. In agematched control brains weak positive reaction was located in individual oligodendroglia cells and subpial astrocytes. Prominent increase of αB-crystallin in pathological glia in CJD may represent a response to stress.

    Notizen: Abstract. αB-crystallin is a member of the small heatshock protein family. Under pathological conditions, the expression of αB-crystallin increases in proliferating astrocytes, which suggests that this protein, in addition to glial fibrillary acidic protein (GFAP), can be a marker for gliosis in neurodegenerative diseases. Immunoblotting and immunohistochemical methods were used for the detection of αB-crystallin in the brains of Alzheimer's disease (AD) patients and nondemented controls. An increase in αB-crystallin expression was found in the brains of AD patients. Immunoreaction was present in reactive astrocytes, microglia, and oligodendrocytes, indicating that all types of glia respond to the stress associated with AD pathology. Colocalization of GFAP and αB-crystallin was found in fibrous astrocytes. However, the intensity and range of αB-crystallin expression appeared to be limited as compared with the large increase in the number of GFAP-positive astrocytes. This indicates that expression of αB-crystallin is not a marker for gliosis in AD. Immunoreactivity to αB-crystallin in both astrocytes and microglia was found mainly restricted to areas with senile plaques and neurofibrillary tangles, suggesting the association of αB-crystallin with amyloid deposition AD.
