    Notizen: The effects of type of mixing element in the screw configuration and feed composition on specific mechanical energy input and product attributes during twin-screw extrusion were investigated. The mixing elements studied were kneading, reverse screw and combination of kneading and reverse screw elements. The feed composition was changed by altering the ratio of rice flour to pink salmon muscle. Incorporation of mixing elements increased the specific mechanical energy, and water solubility index; but decreased apparent density and Warner-Bratzler shear stress. Expansion ratio was maximum with kneading elements. However, reverse screw and combination produced extrudates with lesser expansion than that obtained using conveying screws with no mixing element. Increasing the fish solids content in the feed decreased specific mechanical energy, expansion ratio, and water solubility index; but increased the shear stress and apparent density. The shear stress for 0% and 30% fish solids blends were not affected in the expansion ratio range of 3 to 10. Water solubility index and shear stress were linearly related to specific mechanical energy and apparent density respectively.

    Notizen: Metallothioneins (MT) are sulphur-rich, low molecular-weight, intracellular metal-binding proteins with a possible role in carcinogenesis of some human tumours. Metallothionein expression in gallbladder cancer has not been studied previously.〈section xml:id="abs1-2"〉〈title type="main"〉Methods and resultsImmunohistochemical expression of metallothionein was studied in 42 gallbladders (27 cases of carcinoma of the gallbladder, eight chronic cholecystitis and seven cases of normal gallbladder). Metallothionein expression was significantly higher in cases with carcinoma of the gallbladder (70.37%) as compared to chronic cholecystitis (25%) and normal gallbladders (0%). There was a trend suggestive of increasing MT expression with increasing histological dedifferentiation of carcinoma of the gallbladder.〈section xml:id="abs1-3"〉〈title type="main"〉ConclusionsThe increased expression of MT in cases of carcinoma of the gallbladder may represent an increased exposure to heavy metals, which are known carcinogens, and may have a role in gallbladder carcinogenesis. Metallothionein over-expression in carcinoma of the gallbladder may be relevant to the poor prognosis and chemoresistance seen in these cases.

    Notizen: The effects of fish protein hydrolysis time and fish solids level on macroscopic properties of extrudates were investigated. Arrowtooth flounder muscle was autolyzed for varying times; hydrolyzed muscle was recovered, dried, blended with rice flour at varying levels, and extruded using a twin-screw extruder. Hydrolysis beyond 5 min had little effect on extrudate properties. Addition of hydrolyzed fish solids strikingly decreased the apparent density and breaking strength but increased extrudate porosity and expansion ratios. In general, the effect was greater when the level of fish solids was higher. Opposite and expected trends were observed for blends with unhydrolyzed fish solids. Fish solids level played a dominant role in increasing true density.

    Notizen: The effects of 29 screw configurations on rice flour extrudates were investigated. The moisture content, screw speed and feed flow rate during all extrusion experiments were 15%, 400 rpm, and 12kg/h, respectively. Temperature profile in the 8 barrel sections from feed to die end were set at 0, 30, 30, 30, 70, 100, 150, and 150°C. Incorporation of kneading block (KB) and reverse screw element (RSE) in screw profiles significantly influenced apparent density, product expansion (radial, axial and overall), and breaking strength. Apparent density and overall expansion were functions of die temperature. KB was the best element for maximizing radial expansion. Product hardness (breaking strength) decreased with increasing radial expansion.

    Notizen: Abstract Xanthogranulomatous cholecystitis exists in asmall but significant proportion of routinecholecystectomy specimens. A few recent reports haveshown a possible association of this disease withcarcinoma of the gallbladder. All cholecystectomizedspecimens were prospectively evaluated over a period oftwo and half years in a single surgical unit to examinethe incidence of xanthogranulomatous cholecystitis and its association, if any, with carcinoma ofthe gallbladder in an area that is prone to gallbladderdiseases. A total of 460 cholecystectomies wereperformed for various gallbladder diseases. Histological confirmation revealed chronic cholecystitis in311 (67.6%) cases, carcinoma of the gallbladder in 62(13.5%), acute cholecystitis in 29 (6.3%),xanthogranulomatous cholecystitis in 41 (8.9%), andxanthogranuloma and carcinoma of the gallbladder in one case(0.2%) only. Almost all cases were suspected to havechronic cholecystitis on clinical and ultrasonographicfeatures. Two specimens on gross examination showed mass lesions, and hence were suspected to becarcinoma of the gallbladder. Subsequent frozen sectionand histopathology demonstrated xanthogranulomatouscholecystitis. Only one case of xanthogranuloma was found to be associated with carcinoma of thegallbladder but no firm association could be establishedbetween xanthogranulomatous cholecystitis and carcinomaof the gallbladder.

    Notizen: A method was developed to record interactions between mobile non-adherent immunocytes by time-lapse cinematography and then to study the same cells by immuno-electron microscopy, using monoclonal antibodies against surface components. For this purpose a modified stage was designed to fit an inverted microscope. The attachment included a device to cool the culture chamber with N2 gas, a micro-injector for monoclonal antibody and immuno-gold treatment, and two pairs of washing needles to change the medium without disturbance. The technique was first employed to study the formation of aggregates around the antigen-presenting cells in cultures containing cells from hyper-immunized animals. Recently peripheral blood cells from normal subjects and patients with immune deficiency syndromes were stimulated with pokeweed mitogen, cluster formation was recorded, and the cells were processed for immuno-electron microscopy.
