    Notes: Summary An open incubation technique was used to measure S mineralisation in a range of New Zealand soils. For most of the soils studied, the release of S as sulphate was curvilinear with time, and during a 10-week incubation, the amounts of S mineralised ranged from less than 3 μg S g-1 soil to more than 26 μg S g-1 soil. The best predictor of mineralised S appeared to be the amount of C-bonded S in the soil (explaining 59% of the variation in mineralised S between soils). Examination of the soils after incubation also revealed that the bulk of the mineralised S was derived from the C-bonded S pool. Hydriodic acid-reducible forms of organic S appeared to make little contribution to mineralised S. Attempts were made to predict total potentially mineralisable S (S o) from incubation data using an exponential equation and a reciprocal-plot technique. However, the dependence of estimated values of S o on the length and temperature of incubation cast doubts on the validity of this approach.

    Notes: Abstract: β-Bungarotoxin, a snake neurotoxin purified from the venom of Bungarus multicinctus, caused a significant increase in the in vivo release of glutamate from the superfused sensorimotor cortex of awake animals. A smaller effect on GABA release was observed, but no change was detected in the release of six other amino acids measured. The effects on glutamate and GABA release were entirely blocked by tetrodotoxin (1 μM) and were reversible when the cortical tissue was washed with saline.

    Notes: Abstract: The incorporation of [U-14C] protein hydrolysate and [U-14C]leucine into the trichloroacetic acid (TCA)-insoluble membrane and the soluble synaptoplasm proteins of synaptosomes was studied. Following treatment with the depolarizing agents veratrine, Tityus toxin, or potassium, the specific radioactivity of both precursor pool and proteins was measured to examine the link between protein labeling and the fall in the free amino acid pool due to depolarization-induced release of glutamate and aspartate. By reducing the size of the fall in precursor pool due to depolarization by using a nontransmitter amino acid such as leucine (as compared with the usual use of protein hydrolysate), it was shown that the amount by which the pool is reduced is proportional to the change in the protein labeling observed. These results confirm that membrane depolarization causes a large increase in the labeling of membrane-bound proteins as compared with the soluble synaptosomal proteins.

    Notes: Abstract: The incorporation of amino acids into brain proteins following brachial plexus stimulation (BPS) was studied in anaesthetised Sprague-Dawley rats following injection of radioactive precursors of both neuronal and glial compartments. Following intraperitoneal injection of [14C]glucose, which is the major neuronal pool precursor, BPS resulted in a significant increase of 379% (P± 0.001) in the incorporation of carbon from [14C]glucose into TCA-insoluble proteins in the contralateral sensorimotor cortex as compared with the ipsilateral area of the same animal. This increase was abolished totally when tetrodotoxin (10 μg ml-1) was applied topically to the surface of the stimulated area. Following intraperitoneal injection of [14C]acetate, which is considered to be mainly a glial cell precursor, the same afferent electrical stimuli caused a significant decrease of 21% in the incorporation of amino acids into proteins in the stimulated versus unstimulated sensorimotor cortex. With [4-3H]phenyl-alanine or [l-14C]leucine as precursors a significant decrease (12%) or no change was recorded, respectively. A similar decrease in protein synthesis in the stimulated sensorimotor cortex was achieved using different routes of injection. No significant changes were observed in the ratio of the specific radioactivities of the total amino acids of the two hemispheres using either precursor. In vitro, synaptosomes showed a large increase in incorporation into proteins after treatment with electrical pulses, both with [14C]glucose and with [U-14C]acetate as precursors.

    Notes: Abstract— Stimulation (AES) of the brachial plexus of anaesthetised rats resulted in an increased incorporation of carbon from [U-14C]glucose into TCA-insoluble proteins in the contralateral cerebral hemisphere, as compared with the ipsi-lateral hemisphere. The greatest change was observed in the sensori-motor cortex grey matter.Following intraventricular injections of [U-14C]glucose, the changes caused by brachial plexus stimulation were variable, depending on which hemisphere received the label. The injection itself severely inhibited the incorporation into protein. Neither the injection, nor stimulation affected the conversion of [U-14C]glucose into amino acids or its relative distribution between the two hemispheres.

    Notes: Abstract— In this work we have studied the effect of afferent electrical stimulation (AES) of the contralateral brachial plexus on the release of glutamine and glutamate from the cat's brain into the cerebral venous blood, at rest and during continuous infusion of L-glutamine and sucrose solutions.(1) In the resting state, before stimulation, there was a net outflow of glutamine from the brain into the cerebral venous blood, but no release of glutamic acid. (2) AES caused release of glutamate and increased 3.5-fold the release of glutamine. The increase in release of glutamine and glutamate was found to be reversed very shortly after stimulation. (3) Steady intravenous infusion of a 0.3 M-gluta-mine solution for 10 min changed the negative arterio-venous difference in glutamine to a positive one and increased the content in brain by 15×20%. In this case AES caused a singificant drop, to zero of the glutamine arterio-venous difference. (4) At the onset of pentamethylenetetrazole (PTZ) seizures, like AES, there was a significant reduction of the level of glutamine in the cats’cerebral cortex. This reduction vanished when the animals were infused with L-glutamine solution but not with 0.3 M-sucrose solution that was used as an inert electrolyte. (5) The kinetic behaviour of the glutamine transport is compatible with a carrier-mediated process, but not with passive diffusion.

    Notes: Previous studies have revealed that, in areas affected by severe drought, yielding ability and stability of barley (Hordeum vulgare L.) could be improved by increasing the level of heterozygosity. One possible approach to increase the heterozygosity level is to improve the outcrossing rate. In view of that, the genetic variability and phenotypic correlation coefficients involving four floral traits related to outcrossing behaviour were investigated in 274 drought adapted barley lines from the International Center for Agricultural Research in the Dry Areas (ICARDA's) current breeding programme. A significant quantitative variation existed in anther length and width, stigma length, and anther extrusion, which indicate that selection for these traits may be effective. Anther extrusion showed weak but highly significant positive correlations with anther and stigma length. Somewhat higher positive correlations were found between anther and stigma length. The results indicate that selection for high anther extrusion and longer anthers and stigmata may increase outcrossing in cultivated barley and consequently improve yielding ability through enhanced heterozygosity.

    Notes: : Tityustoxin, the active component of the venom of the Brazilian yellow scorpion Tityus serrulatus, caused specific release of the neurotransmitter amino acids glutamate, aspartate and GABA in vivo from the superfused sensori-motor cortex of conscious unanesthetised rats and in vitro from rat cortical synaptosomes. The effects on synaptosomes appear to be due to a depolarising action. Synaptosomal potassium levels were depleted by the toxin. The action was also blocked both in vivo and in vitro by tetrodotoxin and was Ca2+ -dependent. The uptake of [U-14C]GABA was inhibited by tityustoxin but this action was prevented by tetrodotoxin (1 μm). Since the release of [U-14C]GABA from synaptosomes due to the tityustoxin was also prevented by ietrodotoxin under identical circumstances, it is concluded that the tityustoxin has a primary action on release of neurotransmitters rather than on uptake.
