    Notes: To assess the differences in proteolytic activity of acute and chronic wound environments, wound fluids were collected from acute surgical wounds (22 samples) and chronic wounds (25 samples) of various etiologies, including mixed vessel disease ulcers, decubiti and diabetic foot ulcers. Matrix metalloproteinase (MMP) activity measured using the Azocoll assay was significantly elevated by 30 fold in chronic wounds (median 22.8 μg MMP Eq/ml) compared to acute wounds (median 0.76 μg MMP Eq/ml) (p 〈 0.001). The addition of the matrix metalloproteinase inhibitor Illomostat decreased the matrix metalloproteinase activity by approximately 90% in all samples, confirming that the majority of the activity measured was due to matrix metalloproteinases. Gelatin zymograms indicated predominantly elevated matrix metalloproteinase-9 with smaller elevations of matrix metalloproteinase-2. In addition tissue inhibitor of metalloproteinase-1 levels were analyzed in a small subset of acute and chronic wounds. When tissue inhibitor of metalloproteinase-1 levels were compared to protease levels there was an inverse correlation (p = 0.02, r = – 0.78). In vitro degradation of epidermal growth factor was measured by addition of 125I labelled epidermal growth factor to acute and chronic wound fluid samples. There was significantly higher degradation of epidermal growth factor in chronic wound fluid samples (mean 28.1%) compared to acute samples (mean 0.6%). This also correlated to the epidermal growth factor activity of these wound fluid samples (p 〈 0.001, r = 0.64). Additionally, the levels of proteases were assayed in wound fluid collected from 15 venous leg ulcers during a nonhealing and healing phase using a unique model of chronic wound healing in humans. Patients with nonhealing venous leg ulcers were admitted to the hospital for bed rest and wound fluid samples were collected on admission (nonhealing phase) and after 2 weeks (healing phase) when the ulcers had begun to heal as evidenced by a reduction in size (median 12%). These data showed that the elevated levels of matrix metalloproteinase activity decreased significantly as healing occurs in chronic leg ulcers (p 〈 0.01). This parallels the processes observed in normally healing acute wounds. This data also supports the case for the addition of protease inhibitors in chronic wounds in conjunction with any treatments using growth factors.

    Notes: Abstract A vast array of structurally diverse aromatic compounds is continually released into the environment due to the decomposition of green plants and as a consequence of human industrial activities. Increasing numbers of bacteria that utilize aromatic compounds in the absence of oxygen have been brought into pure culture in recent years. These include most major metabolic types of anaerobic heterotrophs and acetogenic bacteria. Diverse microbes utilize aromatic compounds for diverse purposes. Chlorinated aromatic compounds can serve as electron acceptors in dehalorespiration. Humic substances serve as electron shuttles to enable the use of inorganic electron acceptors, such as insoluble iron oxides, that are not always easily reduced by microbes. Substituents that are attached to aromatic rings may serve as carbon or energy sources for microbes. Examples include acyl side chains and methyl groups. Finally, aromatic compounds can be completely degraded to serve as carbon and energy sources. Routes by which various types of aromatic compounds, including toluene, ethylbenzene, phenol, benzoate, and dihydroxylated compounds, are degraded have been elucidated in recent years. Biochemical strategies employed by microbes to destabilize the aromatic ring in preparation for degradation have become apparent from this work.

    Notes: This paper reports the results of a Management History Division survey within the Academy of Management which investigated the current status and future direction of management history teaching in the management curriculum and the role and direction of the Management History Division in general. Comparisons were made to a similar 1989 survey. While management history as a separate course remains elusive, management history continues to be taught in other mainstream management courses. The role of the Management History Division is seen as critical in encouraging others to teach management history. Significant accomplishments have been made in this area since the earlier survey including an expanded Executive Committee, a revised newsletter, new awards for service in the field, and the initiation of the Journal of Management History as an outlet for publication in the field.

    Notes: Abstract Ammonia is the inorganic nitrogen source preferentially used by most cyanobacteria. Moreover, even in nitrate utilizers or N2-fixers, ammonia is an obligate intracellular intermediate in nitrogen assimilation. It also affects the synthesis and the activation of several key enzymes in nitrogen metabolism such as nitrogenase, glutamine synthetase and nitrate reductase. The mechanism by which NH3/NH4+ enters different cyanobacteria (N2-fixers, non-fixers, neutrophilic) was thus studied. Using 14CH3NH3+, the convenient radioactive analog of ammonium, we have shown that the neutrophilic A. nidulans R-2 possibly possesses an active transport system for this cation. The conditions leading to repression and depression of this transport system have been studied; it appears that de novo protein synthesis is required for the acquisition of the transport ability. We have also provided evidence that methionine sulfoximine affects ammonium uptake only through its inhibition on glutamine synthetase, and found no support for the possible interaction of this inhibitor with the ammonium transporter. In the alkalophilic cyanobacterium Spirulina platensis, an active mechanism to translocate ammonium is probably not needed. Our data suggest that net uptake of ammonia to support optimal growth could be explained by a pH driven diffusion process.In all the different strains tested net ammonia uptake was observed only when conditions permit continuous amidation through the activity glutamine synthetase and inhibition of this enzyme by methionine sulfoximine caused the excretion of ammonia to the external medium. The weight of experimentation suggests that ammonia leaks from cells because of the inherent permeability of NH3, and that no specific carrier is involved in its release.

    Notes: The unicellular cyanobacterium Synechococcus R-2 (PCC 7942) took up 14CH3NH+3 at least 10 times faster after growth with NO−3 than with NH+4 as nitrogen source. When NH+4-grown cells were transferred to medium containing NO−3, the ability to accumulate 14CH3NH+3 required approximately 6 h for maximal expression. Chloramphenicol (50 μg · ml−1) completely prevented the rise in transport ability. Transport by nitrate-grown cells incubated in 2 mM NH4Cl or CH3NH3Cl, with or without NO−3, continued at high rates for 2–3 h, but fell to zero by about 5 h incubation in light. Methylammonium, or the γ-methyl glutamic amide derived from it, thus may also serve as a signal for repression; however, externally added glutamine stimulated rather than repressed 14CH3NH+3 transport. Changes in 14CH3NH+3 uptake rates brought about by NH+4 were confined to these slow effects, and to competition for entry into the cell.
