    Notes: SYNOPSIS. Transection and regeneration of Blepharisma intermedium initiate complex macronuclear activity and micronuclear division. Reversible condensation of the macronucleus is achieved by contraction which involves primary and secondary coiling. Primary coiling, evident in vegetative cells, is enhanced by pronounced constriction of the macronuclear extremities during regeneration. Secondary coiling is restricted to the period of contraction and is initiated at the ends of the mauonucleus. Condensation terminates the coiling processes and gives rise to an amorphous structure. Re-elongation of the macronucleus is not followed immediately by coiling. Micronuclear division begins early in regeneration, but the peak of visible activity is not reached until the 3rd through 5th hours.

    Notes: SUMMARY. The mating behavior of 16 stocks of Paramecium multimicronucleatum from 12 states has been studied. Mating always follows a decline in nutritive conditions. The evidence indicates that there exists one set of four interbreeding mating types. The period of subculture strongly influences the mating reactions, many stocks mating only long after isolation from nature and culture in the laboratory under the restricted conditions employed. Selfing was observed to occur in many races, usually after long periods of subculture. It first occurred in only a small proportion of the population, later in a larger proportion. The similarities and differences between P. multimicronucleatum and other species of protozoa showing multiple mating types are discussed.

    Notes: SYNOPSIS. Transection and repair in Blepharisma intermedium modify the distribution of several enzymes. The number of active phagosomes is reduced and there are increased staining reactions for acid phosphatase, the nucleases and type-B esterases in the ground cytoplasm. 5'-Nucleotidase activity is localized in the cytoplasmic matrix. Except for ribonuclease, the enzymes may have a perimacronuclear deposition at some period during regeneration. Acid phosphatase may be involved in development of the oral membranelles. Adenosine triphosphatase and lactic dehydrogenase have a moderate staining intensity whereas malic and succinic dehydrogenases have a strong reaction in the cytoplasm; in addition, these enzymes may be localized in the kinetics involved in the neoformation of the oral concrescence. Nuclear reorganization is attended by an increased frequency of peripheral dehydrogenase activity in the late stages of regeneration. These observations are compared with catalytic systems known to function in protozoan metabolism and in vertebrate regeneration.

    Notes: SYNOPSIS. Individuals of Blepharisma intermedium (Rao A race) acclimated by prolonged growth in axenic medium containing 8X, 16X, 24X and 32X the concentration of salts in unit standard balanced salt solution (USBSS) used for fresh-water protozoans were tested for rate of regeneration at various salt concentrations, the time for 50% regeneration being used for comparisons. Blepharismas acclimated to axenic medium (of osmolality 10 × USBSS) containing 16X USBSS regenerated at about the same rate as those grown in axenic medium but were delayed at higher salt concentrations. The same was true for those grown in axenic medium containing 32X USBSS, except that the range of higher salt concentration tolerated was much greater. Those acclimated to higher salt concentrations were delayed in regeneration by exposure to lower salt concentrations tolerated by controls. Because growth was very slow in media of higher osmolal concentration, it was not possible to determine the internal concentration of salts in acclimated cells. The morphogenetic process as measured by rate of regeneration, appeared to be less sensitive to increased salt concentration than growth, as measured by cell division.

    Notes: SYNOPSIS. The regeneration of three species of Blepharisma—americanum, undulans and japonicum (grown in conslant yellow light in a constant-temperature room at 25d̀C) from specimens of which the hypostome had been removed, was studied under a variety of conditions. The reliability of the methods was checked by an extensive series of experiments with B. americanum and B. japonicum under normal conditions. The time of regeneration of 50% of the individuals was taken as a convenient means for comparing action of various agents. No diurnal rhythm of rate of regeneration was observed. Regeneration is readily affected by the state of nutrition of the cells, starved ones regenerating more slowly than well-fed ones. Regeneration is accelerated by maintaining the cells at 30d̀ but retarded at 35d̀. Even brief exposures to 40d̀ may retard regeneration. Regeneration is slowed by maintenance at temperatures below 25d̀, and stopped at 10d̀. Cut Blepharisma placed at 5d̀ lose pigment in a “shock” reaction and many die. The injury is more marked in lettuce medium than in balanced salt solution. Between 13 and 30d̀ the rate of regeneration increases with rise in temperature, the Q10′s for the ranges 13 to 18d̀, 18 to 25d̀ and 25 to 30d̀ being, respectively, 6.92, 3.93 and 1.56. The effect of temperature upon division of Blepharisma was found to be similarly sensitive to temperature, the Q10's for the ranges 13 to 18d̀, 18 to 25d̀ and 25 to 30d̀ being 3.87, 2.92 and 1.59, respectively. High and low temperature limits for division were also found to be similar to those for regeneration. Preliminary evidence indicates some degree of thermal adaptation in Blepharisma to both higher and lower temperatures than those usually tolerated. The studies indicate a general similarity in sensitivity of regeneration and division to environmental influences in the various races of the species studied, with some individual variations.
