    Description / Table of Contents: Zusammenfassung Die anatomische Untersuchung von 5 Fällen von prolongiertem posttraumatischem Coma und von 2 Fällen von sogenanntem “Hirntod” derselben Ätiologie erlaubt die folgenden Feststellungen: Außer den Läsionen durch Kontusion oder cerebrale Anoxie und den Veränderungen durch arterielle Gefäßstörungen, die nicht genügen, um die bei diesen Kranken vorhandenen Bewußtseinsstörungen zu erklären, bestehen die am meisten konstanten Veränderungen in folgendem: 1. Diffuse Markablassung, mehr oder minder akzentuiert im Centrum ovale. 2. Lokalisierte Nekroseherde im Hemisphärenmark. Das anatomische Bild entspricht dem Syndrom der funktionellen Dekortikation, das an den Kranken in vivo zu beobachten ist. Diese Läsionen erscheinen wohl durch mechanische Faktoren bedingt (Faserzerreißung und Ödem); sie unterscheiden sich von den Läsionen des Hirnstamms, die gewöhnlich bei jenen posttraumatischen Comata zur Beobachtung gelangen, die rasch zum Tode führen. Es ist begreiflich, daß diese Läsionen sich in Richtung auf die globale Großhirnatrophie der posttraumatischen Demenzen entwickeln.
    Notes: Résumé L'étude anatomique de 5 cas de comas prolongés post-traumatiques et de 2 cas de «mort du cerveau» de même étiologie nous a permis les constatations suivantes: en dehors des lésions de contusion ou d'anoxie cérébrale et des altérations d'origine vasculaire artérielle, qui ne suffisaient pas à expliquer les troubles de conscience présentés par nos malades, les modifications les plus constantes étaient représentées par: 1. Une pâleur myélinique diffuse, plus ou moins accentuée, du centre ovale. 2. Des foyers de désintégration localisés à la substance blanche hémisphérique. Ce tableau anatomique correspond au tableau de décortication fonctionnelle observé du vivant des malades. Ses lésions paraîssent bien liées à des facteurs mécaniques (dilacération des fibres et oedème); elles s'opposent aux lésions du tronc cérébral habituellement observées dans les comas post-traumatiques rapidement mortels. On conçoit que de telles lésions puissent évoluer vers l'atrophie cérébrale globale des démences post-traumatiques.

    Notes: Summary The concentration-effect relationships of pinacidil, a peripheral vasodilator, have been measured in 12 healthy adults who received placebo or pinacidil 25 mg daily for 1 week in a cross-over experiment. Diastolic blood pressure (DBP) and heart rate (HR) were recorded and blood samples were taken on days 1 and 7. Plasma drug concentration-time data were fitted by a biexponential function with zero-order input. The pharmacokinetic model was incorporated into a combined pharmacokinetic-dynamic model (PK-PD) using the Hill equation, which has three parameters: n, the sigmoidicity parameter, Emax the maximum effect and EC50 the concentration which gives 50% of Emax. For ΔDBP, the parameter medians were estimated as n=5, EC50=44.6 ng · ml−1 and Emax=13.5 mm Hg. A hysteresis loop was found when ΔHR was plotted against concentration, which could be fitted by a linear effect compartment model. Simulations showed that experimental ΔDBP points on Day 7 could be predicted from a simulated curve computed by the model using parameters estimated on Day 1. Using the simulation, it was possible to suggest an optimal dosage regimen for pinacidil tablets.

    Notes: Summary The hypotensive and peripheral vascular effects of two different oral formulations of pinacidil were investigated in twelve healthy male volunteers. Arterial blood pressure, heart rate and calf blood flow were measured on Days 1 and 7. The vasodilator activity of both formulations was confirmed by a significant increase in calf blood flow on both days, which was correlated with a significant decrease in diastolic blood pressure. There was no differences between the tablet and the pellet.

    Notes: Summary Three ranges of doses of growth hormone releasing factor (2.5–80 µg, 80–320 µg and 75–600 µg) were intravenously administered to healthy young volunteers in three double blind studies. Serum circulating GRF levels were determined by radioimmunoassay. Experimental concentration curves were fitted, using the extended least squares method, to a biexponential model for the structural model and power function for the variance model. The power variance model, compared to the constant variance model greatly reduced the coefficient of variation of the biexponential parameters. The power of the variance model was estimated to be 1.95. The distribution half-life was 6.6 min and the elimination half-life was 39.0 min (harmonic means). Total clearance was 0.12±0.01 µg/l/min. No difference between these parameters was found for the various doses. GRF kinetics was linear established in the range 10 to 600 µg which means that elimination was not altered by the increased doses.

    Notes: The purpose of this in vivo study was to investigate, non-invasively on human subjects, xerotic skin and its physiological evolution over time, compared to normal skin. Two groups of 17 female subjects were studied during the winter season, one made up of subjects with normal skin and the other subjects with xerotic skin. A clinical assessment and biometrological measurements of hydration and transepidermal water loss (TEWL) were performed on the same area of the external antero-lateral surface of the leg at the start of the study then after three weeks. At the end of the study, the ultrastructure of stratum corneum samples taken from the same area was examined by transmission electron microscopy.Subjects with xerotic skin were selected according to their impaired cutaneous barrier function, reflected in a TEWL higher than 12 g/m 〈inlineGraphic alt="inline image" href="urn:x-wiley:01425463:ICS196570:ICS_196570_mu1" location="equation/ICS_196570_mu1.gif" extraInfo="missing"/〉/h. Compared to normal subjects, they presented a hydration level more than 25% lower. After an interval of 21 days, no significant change in the hydration level or clinical appearance of the xerotic skin was observed. Changes in the ultrastructure of the stratum corneum were also observed in the subjects with xerotic skin. Unlike normal skin, corneosomes could be detected right up to the surface layers, accompanied by intercellular lipids in an amorphous form. These observations confirm the important roles played by both corneosomes and lipid organization in the cohesion/desquamation processes.In the subjects with normal skin, the hydration level and barrier function remained unchanged during the three week study but an onset of skin dryness was observed, the mean clinical score increasing by +1.3 (p = 0.01). These results confirm that there is no direct relationship between TEWL and the severity of skin dryness. It appears that a clinical evaluation is more sensitive than biometrological measurement for describing early state of cutaneous dryness. This study highlights the importance of a regular cosmetic or dermopharmaceutical treatment during the winter to prevent xerosis apparition on legs.

    Notes: Measurements were carried out on 19 commercial cosmetic samples. Initially, their anti-infrared properties were quantified, in vivo, on healthy volunteers, by measuring variations in the Colorimetric Erythema Index ΔCEI, before and after application of a normalized quantity of product. Subsequently, the physico-chemical response of the product was tested in vitro by measuring the infrared reflection coefficient ΔIR at 800 nm.The correlation established between these two parameters enables products to be ranked in three categories. A good correlation was demonstrated between reflective power (ΔIR) and anti-erythemic properties (—ΔCEI) of the products in the first two groups.A qualitative examination of the diffraction of X-rays confirms the presence of titanium dioxide, TiO2 rutile, in most products with high IR protection properties whereas TiO2 anatase was detected in a sample with no IR reflection properties.

    Notes: A new method allowing accurate determination of the primary electron energy corresponding to the zero charge deposition on insulators is presented. It consists in evaluating the surface voltage variations under an electron-beam charge deposition, by means of a classical secondary electron spectrometer. Experimental data on lithography photoresists are reported. Comparison is made with classical metrology measurements, i.e., imaging profiles, and a good agreement is found. Experimental results show the angular dependence of the highest noncharge energy. A formulation of the secondary emission coefficient angular dependence is proposed and discussed versus experimental data.
