    Notes: Abstract The performance of a tapered reactor for the continuous cultivation of bakers' yeast (SCP) from cane molasses has been compared with that of a conventional cylindrical reactor. It is found that the tapered reactor has less non-idealities (bypass and deadspace). Using the experimentally evaluated bypass and deadspace values, a model for predicting conversions of substrate (cane molasses), based on the RTD model proposed by Cholette and Cloutier has been developed. The experimental substrate conversions are found to match the model satisfactorily.

    Notes: Abstract Biosurfactants capable of emulsifying pesticides have great potential to assist in microbial degradation of the pesticides. Solid State Fermentation (SSF) due to several advantages, is one of the efficient ways of producing these surfactants and seldom receives attention for commercial exploitation. In this study, a packed column bioreactor with wheat bran as the raw material and Bacillus subtilis has been used to produce a biosurfactant specific to disperse Fenthion, an organophosphrous pesticide. The emulsifier activity (EA) and surface tension from the packed column bioreactor were compared with flask fermentation experiments, which served as control. Airflow rate in the packed column bioreactor was varied from 10–20 l/min. Maximum EA and minimum surface tension occurred at airflow rate of 20 l/min. Peak EA in the control was 1.2 at 29 h while it was 1.9 in the bioreactor. The least surface tension of 24 dynes/cm was noticed at 54 h in the bioreactor, which was 33% better than the control at the same time period. The results indicate that the packed column bioreactor can become a more acceptable solid state fermentation system for commercial exploitation of Fenthion specific biosurfactant production.

    Notes: Abstract A simple mathematical model for the interaction of mass transport with biochemical reaction in solid state fermentations (SSF) in static tray type bioreactors under isothermal conditions has been developed. The analysis has enabled scientific explanations to a number of practical observations, through the concept of critical substrate bed thickness. The model will be most useful in the prediction of the concentration gradients as also in efficient design of these bioreactors.

    Notes: Abstract Aqueous two phase extraction (ATPE) in combination with ultrafiltration was employed for concentration and purification of amyloglucosidase produced by solid state fermentation. After extraction (with water) from dry moldy bran the dilute enzyme extract was concentrated by ATPE in a polyethylene glycol (PEG)/maltodextrin (MDX) system. The enzyme in the top PEG rich phase was then extracted into a Na2HPO4 rich bottom phase and further concentrated by ultrafiltration. The partitioning behavior of amyloglucosidase was examined in PEG/MDX, PEG/Na2SO4, PEG/Na2HPO4, PEG/KH2PO4 aqueous two phase systems. Effect of buffering salts such as NaCl, Na2HPO4, KH2PO4 and Na2SO4 on the partitioning behavior of enzyme was studied in PEG/MDX system. Maximum partitioning of amyloglucosidase was seen with KH2PO4 (m = 18.1). A two stage ATPE employing PEG/MDX (buffered with KH2PO4) and PEG/Na2HPO4 systems, followed by ultrafiltration has resulted in an overall recovery of 78.4% with 3.1 fold purification and 9.4 fold concentration of the enzyme.
