    Notes: Summary The cytotoxicity of 5-fluorouracil (5-FU) is due in part to the incorporation of the base into RNA molecules. We assessed the cytotoxicity of 5-FU in human colonic carcinoma HT-29 cells and examined mRNA activity (measured by protein biosynthesis in vivo and in vitro) and the maturation of rRNA precursors as two possible modes of action of 5-FU. The rRNA processing pathways were studied using rDNA sequences as probes in blot hybridisation protocols and were specific for both the precursors and mature rRNA species of the maturation pathways. The conclusion from the studies was that although differences in mRNA activity were detected in vivo and in vitro, the significance of these changes are as yet unknown. In contrast, the effects on the pre-rRNA processing pathways proved to be highly significant cytotoxic consequences of 5-FU administration. We discuss the implications of this finding for an understanding of the mode of action of the drug and for the future monitoring of tumour sensitivity to 5-FU.

    Notes: Abstract The accuracy of CARS N2 thermometry has been assessed using carefully measured experimental data at high pressures for the temperature range 295 to 870 K and pressure range 1–20 bar. Excellent agreement between calculated and experimental spectra was achieved using statistically based fitting laws to model rotational relaxation. Temperature errors of within ±60 K were achieved using the MEG fitting law, and within ±30 K using an extension to the model (XMEG). Results of separate ab initio IOS calculations with ECS corrections for rotational relaxation gave good overall agreement but did not model the structure of the motionally narrowed spectra as well as the empirical models. The various models were also compared with some complementary data measured by DLR, Stuttgart in their high pressure burner for pressures up to 40 bar. This study also investigated the sensitivity of CARS thermometry to pressure; in certain regions a significant reduction in sensitivity was found. Other key factors investigated include nonlinearities in the intensified diode array detector.

    Notes: Abstract Single shot degenerate four wave mixing (DFWM) images of the distribution of nitrogen dioxide (NO2) doped into a propane/air flame at concentrations of the order of 7000 ppm have been obtained. These images indicate the relative concentration of NO2 in different parts of the flame with an estimated spatial resolution of 150 μm. Initial experiments were performed using NO2 in a glass cell with nitrogen buffer gas. DFWM signals were generated using both the frequency doubled output of a pulsed ND:YAG laser and the tunable blue output of an excimer pumped dye laser. The signal was investigated as a function of laser power, NO2 concentration and buffer gas pressure. In addition, spectra of NO2 in the region 450 to 480 nm were obtained. Signals were then sought in both a cold air/NO2 gas flow and an ignited mixture of propane and air seeded with NO2, using a DFWM imaging geometry. The resulting images from the flame demonstrate the disappearance of the NO2 molecules in the flame interaction zone.

    Notes: The self-broadening of the Q-branch vibration-rotational lines in the CARS spectrum of HCl was studied over the pressure range 0.002-1.000 bar and the temperature range 298-1048 K. The broadening coefficients, σp, are dependent on J at 298 and 550 K but are independent of J at higher temperatures; the σp value of 1048 K is only about 25% of its value for Jmax at 298 K.

    Notes: The Raman spectrum of the ν1 band of water vapour at pressures up to 300 τ and for five temperatures between 300 and 1200 K was measured using high-resolution inverse Raman spectroscopy. Self-broadening coefficients were determined for 59 distinct lines from J = 0 to J = 11. The temperature dependence of these coefficients was also determined for most of these lines and was compared with previously published calculations. Spectral regions where deviations have been previously observed between measured and calculated CARS spectra of the ν1 band were studied in detail. Corrections to line positions and relative intensities are suggested for improving the current water vapour spectral model.

    Notes: Calibrated coherent anti-Stokes Raman (CARS) spectra have been recorded for the resolved Q branches of the vibrational fundamentals and first hot bands of 14N2, 14N15N and 15N2 at a temperature of 1200 K. The spectra show a convolved linewidth of 0.13 cm-1, and have been used to derive new values of spectroscopic constants, for the species observed, with specific reference to the position of the Q branch lines where J 〉 20 for v = 1-0, and J = 0-30 for v = 2-1. The critical overlap region of the fundamental and the hot band may now be computer simulated with confidence, which should be of considerable help in combustor thermometry using CARS.
