    Description / Table of Contents: Zusammenfassung Kürzlich wurde angegeben, die UV-Absorption von Thioharnstoff in Gegenwart von Cu(II) führe zu genaueren Ergebnissen als kolorimetrische Methoden. Wir konnten feststellen, daß die Zugabe von Cu(II) unnötig ist, daß die Messung bei 236 nm empfindlicher ist und genauere Bestimmungen von 0,1 μg bis 0,1 mg Thioharnstoff/ml ermöglicht. Ein zehnfacher Überschuß von Harnstoff stört nicht. Die durch Kupferzusätze verursachten Komplikationen wurden erörtert.
    Notes: Summary It has recently been suggested that determination of thiourea from its ultraviolet absorption in the presence of Cu(II) is more accurate than colorimetric methods mentioned in the literature. We have found that the addition of Cu(II) is unnecessary and measurement at the 236 nm absorption peak is more sensitive and more accurate allowing determination of thiourea in the concentration range 0.1μg per ml to 0.1 mg per ml. A 10-fold excess of urea does not interfere with the estimation of thiourea. The complications caused by the addition of copper are discussed.

    Notes: Abstract Sedimentation equilibrium centrifugation has been applied to determine the affinity and stoichiometry of the interaction between Fab fragments, derived from monoclonal antibodies NC10 and NC41, with influenza virus neuraminidase N9 isolated from either tern or whale. Although the two neuraminidase epitopes recognized by NC10 and NC41 Fab overlap, crystal-lographic studies have shown that the modes of binding of each Fab are different. The sedimentation equilibrium experiments described here reveal that the binding affinities are also different, with NC10 Fab binding more strongly to each neuraminidase. Furthermore, comparison of the affinity of binding of each antibody fragment reveals a stronger interaction with tern neuraminidase than with whale neuraminidase. Although the respective epitopes recognized by each antibody on the two antigens are similar, this technique shows that they do nevertheless possess sufficient differences to affect significantly the binding of antibody.
