    Notes: Abstract  The sensitivity for DNA adduct formation by antitumor alkylating agents (mechlorethamine, cisplatin and adozelesin) of the postlabeling technique and thin-layer chromatography was studied. Three DNAs were used: a double-stranded 20-bp oligonucleotide of defined sequence, calf thymus DNA and murine leukemia L1210 cellular DNA. With high concentrations of mechlorethamine, there was a marked decrease in normal dGp, a lesser decrease in dAp and dCp and no change in dTp. Using 2D mapping PEI-cellulose thin-layer chromatography analyses, it was found that six mechlorethamine: DNA adducts were produced after a short exposure to mechlorethamine. After an extended time at relatively high drug concentrations there was an alteration in the mechlorethamine: DNA adduct pattern that may reflect the conversion of monoadducts to crosslinked adducts. Similar observations were made with cisplatin and adozelesin. When murine leukemia L1210 cells were treated with 50 μM mechlorethamine or 50 μM cisplatin for 1 h, six or more mechlorethamine: DNA adducts and five cisplatin: DNA adducts were detected. After allowing 6 h. for repair of potentially lethal damage, several adducts were no longer detectable and others appeared with diminished intensity. Nuclease P1 dephosphorylates normal nucleotides at relatively low enzyme concentrations with variation depending upon the nucleotide. In general, considerably lower concentrations of nuclease P1 were required to dephosphorylate the normal nucleotides than to dephosphorylate the antitumor alkylating agent: nucleotide adducts, thus allowing increased sensitivity of the postlabeling assay. The sensitivity of detection of antitumor alkylating agent: DNA adducts in DNA from treated L1210 cells approached one adduct per 107–108 nucleotides. These results suggest that the postlabeling technique may be sufficiently sensitive and specific for the study of the clinically effective levels of antitumor alkylating agents.

    Notes: Stress corrosion cracking (SCC) behaviors of pipeline steel X70 in various near-neutral pHsoil environments with characteristics of eastern China have been studied through electrochemicalmeasurements and slow strain rate tests (SSRT) at various electrode potentials in four solutionscontaining different typical soils in eastern China as well as in NS-4 solution. The SCC susceptibilityin the four soil solutions was generally higher than that in NS-4 solution. There was a general trendthat SCC susceptibility increased with decreasing the potential, suggesting that hydrogen inducedcracking probably plays a key role in the cracking at least at low potentials and the parameters ofcathodic protection in engineering should be carefully optimized to avoid SCC

    Notes: Effects of some environmental factors on stress corrosion cracking (SCC) of pipeline steel X-70 both in near-neutral pH environments, including NS4 solution and several solutions containing main types of soil in the eastern part of China, and in high pH solution were studied by means of electrochemical measurement and slow strain rate testing (SSRT). The anodic polarization curves showed different features in near-neutral pH and high pH solutions in terms of active-passive transition behavior. In near-neutral pH solutions, the cracking mode was transgranular with the feature of quasi-cleavage, the susceptibility to SCC increased with decreasing potential, pH and temperature as well as increasing CO2, indicating a dominant mechanism of hydrogen induced cracking (HIC). In high pH solutions, the cracking behavior was similar to that in near-neutral pH solutions when the specimens were polarized at cathodic potentials, but quite different at anodic potentials. A comparison of the electrochemical behavior with the SCC potential region indicated a dominant SCC mechanism associated with anodic dissolution (AD) of X70 in high pH solution at anodic potentials. A preliminary experimental potential (E)-pH-SCC diagram has been established for X70 in near-neutral pH environments

    Notes: Crack propagation of X-70 pipeline steel in near-neutral pH solution was studied under different modes of cyclic loading. A revised equation of crack growth rate vs. Δ K was obtained. Average crack growth rate increased with cycles under conditions of different R values. Linear shape notch specimen made cracks much easier to initiate and propagate than V-shaped notch specimen did. For different R values, the curves of crack growth rate with cycles were similar, but the platformpropagation period and quick propagation period were different obviously. Crack growth rate at both periods increased and thus failure time decreased markedly with decrease of R value. The propagation directions of cracks were different under different cyclic loading conditions. Under mode I (single tensile stress) cyclic loading, cracks were straight and perpendicular to the tensile stress axis, while under mixed-mode I/III (tensile/shear stress) cyclic loading, cracks were sinuousand did not propagate in the direction perpendicular to the main tensile stress axis

    Notes: Abstract The dynamic interaction problems of three-dimensional linear elastic structures with arbitrary shaped section embedded in a homogeneous, isotropic and linear elastic half space under dynamic disturbances are numerically solved. The numerical method employed is a combination of the time domain semi-analytical boundary element method (SBEM) used for the semi-infinite soil medium and the semi-analytical finite element method (SFEM) used for the three-dimensional structure. The two methods are combined through equilibrium and compatibility conditions at the soil-structure interface. Displacements, velocities, accelerations and interaction forces at the interface between underground structure and soil medium produced by the diffraction of wave by an underground structure for every time step are obtained. In dynamic soil-structure interaction problems, it is advantageous to combine the SBEM and the SFEM in an effort to produce an optimum numerical hybrid scheme which is characterized by the main advantages of the two methods. The effects of the thickness, the ratio of length and diameter of underground structure and the soil medium on dynamic responses are discussed.

    Notes: Spontaneously arising, TGFβ1-resistant colonies were isolated directly from the soft agarose plates of MOSER human colon carcinoma cells grown in the presence of TGFβ1 but in the absence of serum. The colonies were cloned by limiting dilution and screened in a monolayer proliferation assay for sensitivity to TGFβ1 and TGFβ2 isoforms. Cell clones selectively sensitive or resistant to these isoforms in the growth inhibition assay displayed similar differential sensitivities to TGFβ isoforms for production of the extracellular matrix proteins laminin and fibronectin, as well as for the expression of the colon cell differentiation marker carcinoembryonic antigen. Differential receptor binding profiles for TGFβ1 and TGFβ2 were observed among the clones. The isolation of cell clones selectively resistant or sensitive to TGFβ isoforms as well as the identification of differential receptor binding profiles among the clones indicate the heterogeneity of TGFβ responsiveness that exists naturally in human colon tumor cells and stress the importance of defining mechanisms underlying differential responsiveness to TGFβ isoforms. © 1995 Wiley-Liss Inc.
