    Notes: Abstract: There is increasing evidence that members of the natriuretic peptide family display sympathoinhibitory activity, but it remains uncertain which receptor pathway is implicated. We performed cyclic GMP production studies with chromaffin cells treated with either atrial natriuretic factor (ANF) or C-type natriuretic peptide (CNP) and found that these cells specifically express the ANF-R1C but not the ANF-R1A receptor subtype. Evidence for the existence of ANF-R2 receptors was obtained from patch-clamp experiments where C-ANF, an ANF-R2-specific agonist, inhibited nicotinic currents in single isolated chromaffin cells. Involvement of ANF-R2 receptors in the modulation of nicotinic currents was further supported by the significant loss of this inhibitory activity after the cleavage of the disulfide-bridged structure of C-ANF. This linearized form of C-ANF also displayed a lower binding affinity for ANF-R2 receptors. Like the patch-clamp studies, secretion experiments demonstrated that both CNP and C-ANF are equally effective in reducing nicotine-evoked catecholamine secretion by cultured chromaffin cells, raising the possibility that this effect of CNP is predominantly mediated by the ANF-R2 and not the ANF-R1C receptors. Finally, this response appears to be specific to nicotinic agonists because neither histamine- nor KCI-induced secretions were affected by natriuretic peptides. In the present study, we report (1) the presence of ANF-R1C and ANF-R2 receptor subtypes in bovine chromaffin cells, (2) the inhibition by natriuretic peptides of nicotinic whole-cell currents as well as nicotine-induced catecholamine secretion, (3) the possible mediation of these effects by the ANF-R2 class of receptors, and (4) the specificity of this inhibition to nicotinic agonists. Because bovine chromaffin cells release ANF, BNP, and CNP together with catecholamines, all three peptides might exert negative feedback regulation of catecholamine secretion in an autocrine manner by interacting with the nondiscriminating ANF-R2 receptor subtype.

    Notes: Abstract Weaning from mechanical ventilation is usually associated with an increase in oxygen consumption (VO2), which may stress the cardiovascular system. We studied relative changes in the cardiac index and oxygen extraction ratio (EO2) during successful weaning in patients after cardiac surgery (n = 52), cardiac transplantation (n = 17), or abdominal aortic surgery (n = 11). Cardiac index was determined by the thermodilution technique and arterial and mixed venous blood gases were obtained before and 30 min after the start of weaning through a T-piece. The cardiovascular changes were evaluated in 42 patients in whom VO2 (calculated by Fick's equation) increased by more than 10 %. Cardiac index increased more after abdominal aortic surgery (from 3.27 ± 0.77 to 4.44 ± 0.58 l min–1 m–2, p 〈 0.01) than after cardiac surgery (from 2.53 ± 0.59 to 2.87 ± 0.46 l min–1 m–2, p 〈 0.01) or cardiac transplantation (from 2.99 ± 0.64 to 3.33 ± 0.74 l min–1 m–2, p 〈 0.05). EO2 remained stable in patients after aortic surgery (from 25.9 ± 7.1 to 25.2 ± 5.6 %, NS) but increased slightly after cardiac surgery (from 33.3 ± 6.1 to 37.3 ± 6.4 %, NS) and significantly after cardiac transplantation (from 25.8 ± 4.1 to 28.2 ± 4.0 %, p 〈 0.05). Hence the cardiovascular response to weaning from mechanical ventilation may vary according to the type of surgery.

    Notes: Summary Nickel dithiocarbamate complexes derived from some sympathomimetic drugs are examined on silica Radial-Pak columns using binary solvents containing a small percentage of an organic polar modifier. Both the type and concentration of this modifier was found to influence the separation of the ternary from the parent binary complexes. When the two ligands in a ternary complex are racemic to each other, separation of the ternary complex is only possible when certain structural requirements of the molecule are fulfilled. Ternary complexes which contain structurally similar, but nonracemic ligands, are shown to be readily separated from binary complexes. When two such complexes differ only in that one of the ligands in one is enantiomeric to a ligand in the second complex, then it can be shown that the ternary complex with the (+) enantiomer ligand elutes faster from the silica column than the one with the (−) enantiomer ligand. An example of the use of ternary complexes for the identification of optical and structurally related impurities in pharmaceutical products is also given.

    Notes: Summary A reversed-phase chromatographic method combined with an on-line matrix-elimination procedure is described for the determination of thorium and uranium in the nitric acid digest of phosphate rocks. Phoslphate and other anions were fiirst removed from the sample solution using a cation-exchange pre-column to bind cations in the sample and a dilute nitric acid eluent to elute the anions. In the second stage of the procedure, a short C18 column was employed with a phenylhydroxyacetic acid solution used as the washing eluent. Thorium(IV) and uranyl were trapped on the C18 column whilst the transition and lanthanide metals present were flushed to waste due to their weaker retention. In the final step thorium(IV) and uranyl were transferred onto a reversed-phase C18 column, where they were sepoarated and quantified using α-hydroxyisobutyric acid eluent with visible detection at 658 nm asfter post-column reaction with Arsenazo III. The entire procedure was performed automatically using a programmable HPLC pump.
