    Notizen: A coring technique (6), that may be used for estimating the mean amount of herbage on closely grazed pastures, has been adapted to include visual scoring. Two methods are described, both of which use sets of pasture cores from the sward as reference standards. The observers score the herbage on view at random sites against the standards. In the first method the scores are converted to herbage yields directly by reference to the yields of the standards. In the second method a double sampling regression technique is used and the set of standards is used as a visual guide only.

    Notizen: Extras samples were collected from ocsophageally fistulated sheep that had been fed mixtures of known proportions of a legume and a grass. The botanical composition of these samples was estimated by two microscope point techniques. When estimates were made by recording hits on each species the legume content of the ingesta was consistently underestimated by up to 23%. Much of this underestimate was attributed to the weight per unit area of the leaves of the legume being approximately twice that of the grass leaves. When the microscope hits were adjusted for the weight per unit area of each of the plant components (leaves, petioles, flowers and seed pods) the relationship between the estimated and actual legume consumed was improved, the greatest difference between actual and estimated legume being approximately 5%.

    Notizen: Extrusa samples were collected from two groups of oesophageally fistulated sheep grazing together. One group had foam plastic oesophageal plugs fitted below the fistula to assist in the complete collection of extrusa; the second group grazed without plugs. The extrusa were collected in four periods chosen to cover as wide a range of herbage conditions as possible varying from herbage with a high proportion of green leaves to predominantly dry material with a high proportion of dead leaves and stems. The plant components of the extrusa were determined and there was no evidence that stems or any other plant component preferentially by-pass the fistula in grazing sheep. It was concluded that it is not necessary to insert foam plastic plugs in the oesophagus of grazing sheep to obtain representative samples of ingested herbage.

    Notizen: The botanical composition of extrusa collected from oesophageally fistulated sheep fed diets of known proportions of a legume and a grass was estimated by manual separation and a microscope point technique. The microscope hits were adjusted for the weight per unit area of each plant component in order to estimate composition by weight. The microscope point technique was more accurate than manual separation. The deviations between the actual lucerne percentages and percentages estimated by manual separation were explained by higher losses of organic matter prior to separation from the legume than the grass and by the presence of unidentifiable particles that resulted in only a portion of the samples being separated. After adjustments for these sources of error the estimates determined by manual separation were similar to the actual botanical composition of the ingesta.

    Notizen: Abstract Several Drosophila receptor-linked protein tyrosine phosphatases (R-PTPs) are selectively expressed on axons of the developing embryonic central nervous system. The extracellular domains of these axonal R-PTPs are homologous to neural adhesion molecules. Thus, R-PTPs may directly couple cell recognition to signal transduction via control of tyrosine phosphorylation. To examine the function of these molecules during nervous system development, we wished to generate mutations in R-PTP genes. It was unclear whether a mutation in a single R-PTP gene would confer lethality, however, because the similarities in sequence and expression pattern between the axonal R-PTPs suggest that they may have partially redundant functions. To circumvent this problem, we developed a directed mutagenesis strategy based on local transposition of P elements, and used this approach to isolate a null mutation in the DPTP99A gene. This strategy, which we describe in detail here, should be applicable to any Drosophila gene within a lettered division of an appropriately marked P element. Flies lacking DPTP99A expression are viable and fertile, and we have been unable to detect any alterations in the embryonic nervous system of DPTP99A embryos using a variety of antibody markers.
