    Notes: In the present paper, the precision forging of an impeller was studied by means ofnumerical simulation and test forming. Based on the structure and dimension of the impeller, thecombination structure was used in the forging die to obtain the extrusion deformation. The formingprocesses were simulated with DEFORM-3D for different billet dimensions and processingparameters. The parameters, which could ensure the forming quality of the impeller, were determinedby the calculations and analysis. The die structure and the billet dimensions were determinedaccording to the simulation results, and the forging die was designed and manufactured. The billetwith semi-solid microstructure was produced by means of the direct heating-isothermal treatment.The forming was conduced in an YX-315F hydraulic press, and the precision forgings of the impellerwere produced successfully. Both of the simulation and the forming test show that the impellerforging can be formed with the combination structure die and the extruding forming stalesatisfactorily. The ideal parameters to produce the precision forgings of the impeller are: billettemperature at 625ºC, die temperature at 450ºC and punch speed at 20mm/s. Under these conditions,forgings of the impeller can be produced with plump blades, smooth outer surface, and good flowline. This can match the requirements of the precision forging of impellers

    Notes: Abstract Using human telomeric repeats and centromeric alpha repeats, we have identified adjacent single copy cosmid clones from human chromosome 22 cosmid libraries. These single copy cosmids were mapped to chromosome 22 by fluorescence in situ hybridisation (FISH). Based on these cosmids, we established contigs that included part of the telomeric and subtelomeric regions, and part of the centromeric and pericentromeric regions of the long arm of human chromosome 22. Each of the two cosmid contigs consisted of five consecutive steps and spanned approximately 100–150 kb at both extreme ends of 22q. Moreover, highly informative polymorphic markers were identified in the telomeric region. Our results suggest that the telomere specific repeat (TTAGGG) n encompasses a region that is larger than 40 kb. The cosmid contigs and restriction fragment length polymorphism markers described here are useful tools for physical and genetic mapping of chromosome 22, and constitute the basis of further studies of the structure of the subtelomeric and pericentromeric regions of 22q. We also demonstrate the use of these clones in clinical diagnosis of different chromosome 22 aberrations by FISH.

    Notes: In the obligate Intracellular parasitic bacterium, Rickettsia prowazekii, the molar ratio of σ73 to core RNA polymerase, that is, the degree of saturation of the core polymerase by the catalytically active sigma factor, was very low. This ratio was determined from the radioactivity in rickettsial RNA polymerase immuno-precipitated from crude extracts of infected L929 cells in which the parasite was exponentially growing. If we assume that, as Is true for the σ subunit, in R. prowazekii and Escherichia coli the β’and β subunits of the RNA polymerase have similar methionine and cysteine contents (the radiolabelled amino acids), the molar ratio of σ73 to core polymerase in R prowazekii would be 0.1. This is in striking contrast to E. Coli where the ratio is typically 0.4. it remains to be established whether this low sigma saturation results In a limitation of active RNA polymerase in R. prowazekii and contributes to its slow growth.
