    Notes: Quantitative trait loci (QTL) analysis of aluminium (Al) tolerance was performed using Ler/Cvi recombinant inbred (RI) lines of Arabidopsis thaliana. Relative root length (RRL) (root length with 4 µm Al/root length with no Al at pH 5.0) on day 5 was used as the Al tolerance index for QTL analysis. Al tolerance judged by RRL was well correlated to tolerance judged by other indexes, including accumulation of callose, reactive oxygen species in the root apex and growth performance on acid soil containing a large amount of exchangeable Al. Using data sets with an hb2 of 0.91, two QTLs were detected at the top of chromosome 1 and bottom of chromosome 3. These QTLs explained 40 and 16% of the phenotypic variation of Al tolerance, respectively, and the positive effect of the Cvi allele. The QTL on chromosome 1 overlapped with a major QTL in another recombinant inbred population, and is possibly related to malate excretion. A complete pair-wise search revealed 11 sets of epistatic interacting loci pairs, which accounted for the transgressive segregation among the RI population. Several epistatic interactions shared the same chromosomal region, indicating the possible involvement of regulatory proteins in Al tolerance in Arabidopsis.

    Notes: The citrate efflux of a carrot mutant cell line (IPG; insoluble phosphate grower) with enhanced citrate excretion is concomitant with an electrically equivalent H+ release. Furthermore, plasma membrane H+-ATPase activity (EC 3.6.1.35) in the IPG cell line is known to be greater than in the wild-type cell line. In the present study, molecular characterization of plasma membrane H+-ATPase in the IPG cell line was conducted to examine the involvement of this enzyme in enhanced citrate excretion. Four type-II (DcPA 1, 2, 4 and 5) and two type-I (DcPA 3 and 6) full-sized cDNAs encoding Daucus carota plasma membrane H+-ATPase were cloned. The transcript level of the most abundant isoform, DcPA 1, was three times higher in the IPG cell line than in the wild-type cells. The transcript levels of the other isoforms showed no differences between cell types. The plasma membrane H+-ATPase protein level in the IPG cell line was 1.6-fold that of the wild-type cell line when analysed by western blot analysis. These results indicate that the higher activity of plasma membrane H+-ATPase in the IPG cell line is due to up-regulation of DcPA 1. Antisense inhibition of the DcPA 1 gene also led to decreased citrate excretion from the transgenic IPG cell line, suggesting that the up-regulation of plasma membrane H+-ATPase is involved in enhanced citrate excretion.

    Notes: Summary Tomato plants were grown in water-culture with a different supply of Ca (10, 100 ppm) and B (0, 0.2 ppm), and the effects of B deficiency on the translocation and subcellular distribution of Ca in tomato plants were studied by using45CaCl2 as a carrier of Ca. Boron deficiency slight increased the total Ca uptake by the plant and inhibited the Ca translocation to the upper leaves. The incorporation of45Ca into the cell wall in the upper leaves was increased by B deficiency at both Ca levels. As Ca supply decreased, the distribution of45Ca in the 1N NaCl fraction of the cell wall increased only at 0.2 ppm B. As B supply decreased, the distribution of45Ca in the 0.6N HCl fraction increased at both Ca levels. These results suggest that B deficiency inhibit the translocation of Ca, and induce the abnormal changes of the Ca metabolism in the cell wall.
