Notes: Summary Potential polymeric herbicides have been synthesized by reacting poly(acrylic acid)s with 3-aniino-1,2,4-triazole and 1-acetyl-5-amino-1,2,4-triazole, respectively. The produced copolymers contained the active moieties as pendant groups in the range of 3 to 27 per cent of weight and with a content of carboxyl groups up to 95 mol-%. The hydrolytic release of 3-amino-1,2,4-triazole was investigated.

Notes: Zusammenfassung 1.

Notes: Summary Ultrastructural demonstration of NAD-pyrophosphorylase activity (E.C. 2.7.7.1) in isolated mouse liver nuclei was investigated with the use of an electronhistochemical procedure based on the precipitation of pyrophosphate ions with lead ions under conditions permitting simultaneous ATPase inhibition by formaldehyde/ethanol prefixation. In isolated mouse liver nuclei activity of NAD-pyrophosphorylase was found in nucleoli, in interchromatin granules, coiled bodies and strand-like structures in nucleoplasm.

Notes: Summary N-Phosphorylated 3(5)-methylpyrazoles, prepared from the antimiorobially active 3(5)-methylpyrazole by acylation with aryl phosphorodichloridates as well as with phosphoryl chloride, respectively, were reversibly linked to poly(vinyl alcohol). By changing the structure of the phosphorus-containing linkage between bioclde and carrier, it is possible to regulate the hydrolytic release of the active agent. Comparative hydrolysis studies demonstrated that the release rates were essentially independent of the degree of substitution of the polymeric carrier.

Notes: Summary Escherichia coli TG1 (pHRW500) permanently expressed the human interferon α1 gene (ifnα1) directed by the tryptophan promoter (trpP.O) during continuous and fed-batch cultivation with a limited supply of glucose. The expression of ifnα1 could be improved after insertion of the catabolite activator region (cap) upstream to trpP.O during cultivation of the modified E. coli TG1(pHRW500cap) in glucose-controlled continuous and fed-batch cultures. The cap-mediated stimulatory effect on the expression of cap-trpP.O-ifnα1 increased with decreasing dilution rate. These results are in line with the increase in the level of cAMP with declining dilution rate and the well-known positive effects of cAMP-catabolite gene activator protein (CAP) at the transcriptional level. In addition, expression of the galactokinase gene (trpP.O-galK) in E. coli TG1(pDR720) could be improved in the same way with cap-trpP.O-galK in E. coli TG1(pDR720cap). Determinations of plasmid copy numbers, cellular amounts of galactokinase-mRNA, activity of galactokinase (AGalK) and the concentration of galactokinase at various dilution rates (D) strengthen the conclusion that the increase in AGalK with decreasing D was indeed due to the cap-mediated enhancement of transcription of the galK gene. We suggest that expression of other recombinant genes directed by various promoters that allow permanent transcription during growth with limited glucose supply in chemostat and fed-batch fermentors can be improved by appropriate insertion of the cap region.

Notes: Summary B. subtilis A 18, a producer of exocellular amylase, was found to carry covalently closed DNA plasmid molecules (pMI 10). The pMI 10 was isolated and characterized by electron microscopy, electrophoretic mobility and restriction endonuclease cleavage pattern. The pMI 10 was absent in all α-amylase low productive or nonproductive clones. The pMI 10 DNA was transformed together with pUB 110 DNA into B. subtilis RM 125 arg-leu- recipient cells, and, hence, compatibility of these plasmids could be demonstrated.

Notes: Summary Some physico-chemical properties of the DNAs released from the actinophages SH3, SH10, SH11, and SH12 are described. The four phage DNAs have a linear double-stranded secondary structure and are unique with respect to their high G·C contents which, from melting studies and buoyant density experiments, were found to be in the range of 68–73 mol-%. The DNA molecular weights were determined by sedimentation velocity experiments and by electron microscopic length measurements, the mean values of the two corresponding data sets being 34.0·106 (SH3), 26.7·106 (SH10), 26.1·106 (SH11), and 28.7·106 (SH12) with a mean relative error of ±5%. From different observations it was concluded that SH10 DNA, and possibly also SH11 and SH12 DNA, have cohesive ends and can undergo intramolecular or intermolecular association to form ring-like monomers or linear and ring-like multimers. Cleavage of the DNAs of SH3, SH10, SH11, and SH12 by EcoRI restriction endonuclease delivered two, one, zero, and two cleavage sites, respectively, and by BamHI restriction endonuclease eight, zero, zero, and zero cleavage sites, respectively.

Notes: Summary DNA of the temperate Streptomyces phage SH5 (DNA molecular weight 27x106) is subject to restriction-modification mediated by S. hygroscopicus 0477, S. levoris 1331 and 2340. The restriction endonuclease ShyI (isoschizomeric with SacII) isolated from S. hygroscopicus 0477 is involved in restriction of SH5·1331 and SH5·2340 DNAs in S. hygroscopicus 0477.

Notes: Summary The DNA of the temperate Streptomyces phage SH3 contains 100 base-pair long inverted repeats separated by a 940 base-pair long segment of DNA as revealed by electromicroscopic analysis of snapback structures formed after rapid intrastrand reannealing of denatured DNA. The inverted repeat structure was found preferentially at map unit 22 of the circular physical map, in rare cases also in other positions, suggesting a movable character of this genetic element.
