    Notes: Sciatic sensory afferents that retrogradely transport and accumulate leukaemia inhibitory factor (LIF) within their soma were characterized in the adult rat in vivo. Twenty-four percent of neurons within the L4 and L5 dorsal root ganglia accumulated biotinylated LIF following intraneural injection of the cytokine into the sciatic nerve. Labelled cell bodies were predominantly of small diameter (20.1 ± 2 0.5 μm). Retrograde transport was eliminated by excess unlabelled LIF but not by the related cytokines, ciliary-derived neurotrophic factor (CNTF) and interleukin-6 (IL-6). Double labelling revealed that the majority (81%) of LIF-accumulating neurons were immunopositive for CGRP and 34% were immunopositive for the cell surface glycoconjugate IB4. Sixty-two percent of LIF-accumulating neurons were immunopositive for trkA. Our results demonstrate a group of small-diameter sensory neurons that retrogradely transport LIF, comprising cells that constitutively express neuropeptides and those likely to be peptide-deficient. LIF-accumulating neurons expressing trkA are also potentially responsive to nerve growth factor. It is likely that the LIF-accumulating neurons described in this study are nociceptive in function.

    Notes: The expression of the four fibroblast growth factor receptor (FGF-R) genes was examined in murine embryonic stem (ES) cells, embryonal carcinoma (EC) cells, and their differentiated derivatives. FGF-R1 and FGF-R4 were found to be expressed constitutively in all samples examined. The expression of FGF-R2 and FGF-R3 was, however found to increase significantly upon differentiation of both ES and EC cells. Examination of splice variants of the third immunoglobulin domain (Iglll) of the extracellular region of the FGF-R2 revealed that whilst Iglllc transcripts were expressed upon ES cell differentiation, Iglllb transcripts (which confer specificity for the ligand FGF-7) were expressed in both ES cells and their differentiated progeny. FGF-R3 transcripts were also expressed in ES cells, but variont FGF-R3 transcripts containing the Iglllb region were expressed upon differentiation. The findings suggest that the repertoire of FGF-R expression in embryonic cell types is developmentally regulated at the level of both gene expression, and alternative splicing and different members of the FGF-R family can exhibit distinct patterns of both gene and splice variant expression. © 1994 Wiley-Liss, Inc.

    Notes: The requirements for the serum-free culture of PC13 murine embryonal carcinoma cells were determined. Supplementation of a 50:50 mixture of Dulbecco's modified Eagles medium and MCDB104 with transferrin (5 μg/ml), human high-density lipoprotein (HDL) (100 μg/ml), and human low-density lipoprotein (LDL) (50 μg/ml) supported growth comparable to that observed with 5% foetal calf serum. Media supplementation with lipoproteins apparently substitutes for the effects of insulin, desoctapeptide insulin (DOP), or multiplication-stimulating activity (MSA) on EC cell multiplication. Clonal growth of PC13 EC cells in this serum-free medium could only be achieved in the presence of suitable feeder cell monolayers. These observations demonstrate that PC13 EC cells do not have an absolute requirement for exogenous mitogens to support multiplication.

    Notes: The consequences of parental exposure to epidermal growth factor (EGF), for progeny cell cycle times was investigated. Slowly dividing mouse 3T3 fibroblasts were exposed to EGF for 8 hr, the EGF was withdrawn, and the cell cycle times of parental and progeny cells were measured by time-lapse video microscopy. It was observed that exposure to EGF induced a round of cell division following a lag phase of approximately 8 hr. The progeny of these cells exhibited accelerated cell cycle times compared to cells that had not been exposed to EGF. Parental cell division time was significantly correlated with progeny cell cycle time. Sibling progeny cell cycle times were also significantly correlated. EGF can therefore apparently exert an effect on the cell cycle times of more than one generation of cells.
