    Notizen: Abstract— Thelevel of the S-100 protein, a brain-specific antigen, wasdetermined by quantitative complement fixation in the brain stem and cerebrum of the rat during postnatal maturation. The content was minimal at birth in the brain stem and rose to its adult value by day 25. Although S-100 protein could not be detected in the cerebrum of the 2-day-old rat, adult values were also present by the 25th day of age. Neither single dose X-irradiation with 750 rd to the head at 2 days of age or single dose X-irradiation at 11 days of age affected the adult level of S-100 protein in the brain stem or cerebrum. Similarly, hypophysectomy at 20 days of age had no effect on the subsequent levels of S-100 protein.

    Notizen: Abstract— Polysomes were isolated from several different fractions of mouse brain tissue. After homogenization, the extract was centrifuged to yield a post-mitochondrial supernatant fraction and a pellet fraction. Sucrose gradient analysis of the material in the post-mitochondrial supernatant fraction indicated that 80 per cent of the ribosomes were present in polysomes and that little, if any, of the pellet fraction was present. Sucrose gradient analysis of the solution obtained after washing the pellet showed that very little polysomal material was present. The remaining pellet fraction was resuspended in a detergent mixture of deoxycholate-Tween 40. Sucrose gradient analysis of the resulting detergent-soluble solution indicated that large amounts of ribosomal material, in which 60–70 per cent of the ribosomes were associated in polysomes, were present.In brain tissue from young animals, 20 per cent of the polysomes were found in the post-mitochondrial supernatant fraction whereas 80 per cent of the polysomes were released from the pellet fraction by detergent treatment. In contrast, in brain tissue from adult animals, 40 per cent of the polysomes were found in the post-mitochondrial supernatant fraction, whereas 60 per cent of the polysomes were released from the pellet fraction by detergent treatment.

    Notizen: —The concentration of a protein specific to brain, the S-100 protein, was measured in various regions of the human foetal brain at gestational ages ranging from 10 weeks until term. The relative increase in concentrations of the S-100 protein during development of the human foetal brain proceeded in a caudal-rostral fashion. This observation is emphasized by the delayed appearance of the S-100 protein in the frontal cerebral cortex until the 30th week of gestation.

    Notizen: Abstract We have confirmed the observations of Tsunoo et al. (Toxicol. Lett. 4:253, 1979) that (a) DBA/2 mice are more resistant to cadmium mortality than C3H mice and (b) DBA/2 mice accumulate more 109Cd into hepatic metallothionein than do C3H mice in response to an injection of 30 µmol CdCl2/kg, a dose of CdCl2 which is lethal to C3H mice. We now report, using a nonlethal dose of 8 µmol CdCl2/Kg, that the rates of both the synthesis and the degradation of cadmium-induced hepatic metallothionein are increased in C3H mice. The rate of metallothionein synthesis, measured 6 hr after cadmium administration and expressed as the percentage of injected [35S] cysteine incorporated into metallothionein/g liver, was 0.33±0.04% (SD) in C3H mice, compared to 0.19±0.06% in DBA/2 mice (significantly different rates by Students' t test, P〈0.01). Also, at this dose, hepatic 35S-labeled metallothionein was degraded with a half-life of 22.5±0.7 hr in C3H mice, compared to 30.1±2.5 hr in DBA/2 mice (significantly different half-lives by F test, within 95% confidence limits). The increased accumulation of metallothionein in resistant DBA/2 mice compared to sensitive C3H mice after cadmium exposure appears to be due primarily to a difference in metallothionein degradation, rather than metallothionein synthesis.

    Notizen: Adaptation of monolayer cultures of a clonal line of rat glial cells to suspension culture resulted in the nearly complete loss of certain surface antigens. This change in surface antigenicity was paralleled by the loss of the ability of the cells to accumulate in vitro a protein specific to the nervous system (“S100-protein”). In contrast, when glial cells were co-cultivated in monolayer culture with another cell line apparently lacking these surface antigens, the number of these antigens was markedly increased. The possibility of a causal relationship between the changes in the surface antigenicity and the expression of differentiated function is considered.

    Notizen: The synthesis of a protein unique to the nervous system, the “S100-protein,” has been studied in a clonal line of rat glial cells. It has been shown that these cells do not begin to accumulate “S100-protein” until the cultures enter a phase of density-dependent inhibition of cell proliferation. Further experiments indicate that the regulation of “S100-protein” accumulation resides at least in part in an interaction involving the cell surface.

    Notizen: Antisera have been prepared to clonal cultured rat astrocytic cells (C6 cells) and clonal human astrocytic cells (CHB4 cells). When cultured glial cells are grown in the presence of neuraminidase the serological activity of the cell membrane with homologous antiserum is increased extensively. A similar activation of serological activity can be effected by incubating washed harvested cells with this enzyme. Membrane bound N-acetyl-neuraminic acid is extensively reduced by either incubation. The activity responsible for promoting the enhanced serological activity co-chromatographs and co-electrophoreses with neuraminidase. Proteolytic digestion, unlike neuraminidase treatment, was unable to enhance the serological activity of cultured glial cells.

    Notizen: The rat PC12 pheochromocytoma cell line exhibits biological responses to both nerve growth factor (NGF) and epidermal growth factor (EGF). The existence of receptors and biological responses on a common cell for these two well-characterized polypeptide growth factors makes this an attractive system for comparison of ligand binding and processing. Both NGF and EGF are bound to PC12 cells in a competable form at 4°C. At 37°C both ligands are “sequestered”, but at different rates and to different extents. While sequestration happens rapidly and nearly quantitatively for bound EGF, the dissociation reation appears to compete favorably with NGF sequestration. Both EGF and NGF are degraded by PC12 cells. Sequestered EGF, however, is degraded to a greater extent than sequestered NGF.
