    Description / Table of Contents: Zusammenfassung Dichtemessungen der Genauigkeit von 0,01 g/l wurden an menschlichen Blutproben sowie an Plasma und Erythrozytenkonzentrat mit Hilfe eines Dichtemeßgerätes nach der „Biegeschwingermethode“ vorgenommen. Zwischen Blutdichte und Hämatokrit wurde eine hochsignifikante lineare Beziehung gefunden. Bei Kenntnis der Plasma- und Blutzelldichte einer Blutprobe kann der Hämatokrit aus der Blutdichte genauer ermittelt werden als mit der Zentrifugationsmethode, was insbesondere für den Nachweis geringgradiger Änderungen, etwa durch Verschiebung des transkapillären Filtrationsgleichgewichtes, von Interesse ist. Aus der statistischen Verteilung der Plasma- und Erythrozytendichtewerte ergibt sich ferner der Schwankungsbereich der Blutdichte für bestimmte Hämatokritwerte und umgekehrt.
    Notes: Summary Density measurements were performed with an accuracy of 0.01 g/l by means of a density meter based on the “mechanical oscillator technique”, on human whole blood, on plasma, and on erythrocyte concentrates. A highly significant correlation between blood density and hematocrit was found. If plasma and red cell densities are known, hematocrit can be estimated more precisely than using the centrifugation method. This fact is important if small variations of the hematocrit are to be determined. Such variations can be produced, e.g., by alteration of the transcapillary filtration equilibrium. Furthermore, the statistical distribution of plasma and erythrocyte densities allows to determine the range of variation of blood densities for certain hematocrit values and vice versa.

Die ATP-Konzentrationen der Leber und des Blutes von Ratten, die 1–48 Std nach intramuskulärer Injektion von 150–370 mg/kg Butazolidin getötet wurden, unterscheiden sich nicht signifikant von den ATP-Konzentrationen normaler Kontrolltiere. 2. Einedirekte Beeinflussung der Bildung und Verwertung von energiereichen Phosphaten findet demnach in der Leber nicht statt. Die Übertragbarkeit dieses Befundes auf andere Organe wird diskutiert.

    Description / Table of Contents: Summary 0 1. An optical test according to Warburg for the determination of thiamine pyrophosphokinase is described. 2. Comparing extracts of baker's yeast, brewer's yeast, Escherichia coli, spinach and pig liver in our tests, an alkaline extract of dried baker's yeast produced maximal activity. 3. This extract of baker's yeast phosphorylates 168 · 10-9 moles of thiamine per gram of dry weight baker's yeast per hour. This activity is more than ten fold that required for the synthesis of TPP from thiamine under standard growth conditions in the factory. 4. D,l-2-(α-hydroxyethyl)-thiamine (D,l-HET), obtained by treating synthetical D,l-2-(α-hydroxyethyl)-thiamine pyrophosphate (D,l-HETPP “active acetaldehyde”) with phosphatase is phosphorylated by purified thiamine phosphokinase and ATP to D,l-HETPP.
    Notes: Zusammenfassung 1. Ein optischer Test nach Warburg zur Bestimmung von Thiaminpyrophosphokinase wird beschrieben. 2. Bei der Untersuchung von Rohextrakten aus Backhefe, Bierhefe, Escherichia coli, Spinat und Schweineleber zeigte ein alkalischer Extrakt aus getrockneter Backhefe in unserem Test die größte Aktivität. 3. Dieser Backhefe-Extrakt pyrophosphoryliert 168 · 10-9 Mol Thiamin je Gramm Trockensubstanz und je Stunde. Diese Aktivität ist mehr als 10 mal größer als für die Synthese von TPP aus Thiamin beim Wachstum unter normalen, in der Fabrik üblichen Bedingungen, die den optimalen nahekommen, erforderlich ist (etwa 13 · 10-9 Mol je Gramm Trockensubstanz und je Stunde). 4. Aus synthetischem D,l-2-(α-Hydroxyäthyl)-thiaminpyrophosphat (D,l-HETPP = “aktiver Acetaldehyd”) durch Phosphatase-Behandlung gewonnenes D,l-2-(α-Hydroxyäthyl)-thiamin (D,l-HET) wird von gereinigter Thiamin-pyrophosphokinase aus Backhefe mit ATP zu D,l-HETPP pyrophosphoryliert. Es verhält sich in dieser Hinsicht wie HET, das mit Phosphatase aus Enzymatisch gewonnenem HETPP bereitet wurde.

    Notes: Summary The activities of several enzymes involved in the oxidation of ethanol and in the formation of oxaloacetate from acetate were compared in the yeasts Saccharomyces cerevisiae, Hansenula anomala and Rhodotorula glutinis grown on glucose and acetate, respectively. The most striking differences in the regulation of enzyme activities were found for alcohol dehydrogenase, acetaldehyde dehydrogenase, and malate dehydrogenase. The activities of the other enzymes tested behaved rather similar; in all three yeast species the enzymes of the glyoxylic acid by-pass showed the most extensive increase of activity in cells grown on acetate.

    Notes: Abstract Treatment of a yeast suspension with ozone inactivates a number of cytosolic enzymes. Among 15 studied, the most drastic inactivation was found for glyceraldehyde-3-phosphate dehydrogenase and to lesser extents: NAD-glutamate dehydrogenase, pyruvate decarboxylase, phosphofructokinase-1 and NAD-alcohol dehydrogenase. Ozone treatment also effects the quantity of ATP and of other nucleoside triphosphates, reducing to about 50% of the initial value. The ATP missing in the cells appears in the medium. NAD and protein also accumulate in the medium suggesting that the yeast cells have been permeabilized. Permeabilization of the yeast cells by treatment with ozone preceeds the inactivation of glyceraldehyde-3-phosphate dehydrogenase and other cytosolic enzymes.
