    Notes: Summary We describe two patients with sporadic amyotrophic lateral sclerosis (ALS), who had developed progressive external ophthalmoplegia of a predominantly supranuclear type while they survived on respirators, and displayed histopathological abnormalities both typical and atypical of ALS. Patient 1 was a 43-year-old man with ALS of 5-year duration, who had initially exhibited fulminant ALS, and remained on a respirator for 4 years. Patient 2 was a 51-year-old man with ALS of 13-year duration, who remained on a respirator for 8 years. Both patients died in a “totally locked-in state”. Autopsy of both patients revealed not only histopathological abnormalities consistent with ALS, but also multisystem degeneration which involved the pontine tegmentum, substantia nigra, Clarke's dorsal nuclei and spinocerebellar tracts. In addition, Patient 2 displayed intracyto-plasmic neuronal basophilic inclusion bodies which exhibited marked immunoreactivity to anti-ubiquitin antibodies. Our case reports indicate that the longer survival which is possible through the use of respirators may make one subgroup of ALS patients prone to develop atypical clinical and neuropathological features which are not observed during the natural cours of ALS.

    Notes: Summary We have cloned and sequenced the two mouse preproinsulin genes. The deduced amino acid sequences of the mature mouse insulins are identical to the published protein sequences. However, the nucleotide sequence indicates that the mouse I C-peptide has a deletion of two amino acids compared with the mouse II C-peptide. We used an S1 nuclease assay to confirm the presence of the deletion and to measure the ratio of transcripts from gene I to transcripts from gene II. The mouse preproinsulin I gene, like the rat gene I, is missing the second intervening sequence that normally interrupts the C-peptide region in other insulin genes. Comparison of the 5′ flanking sequences of the mouse and rat genes II indicates that they are homologous for at least 1000 base pairs. The preproinsulin I genes also share homology in their 5′ flanking DNAs; however, their homology to the preproinsulin II genes extends for only about 500 base pairs.

    Notes: — Coffee, black tea and artificially flavored and artificially sweetened orange and grape soft drinks were prepared from solutions of 8 minerals each at 750 ppm. Additionally, coffee and tea were brewed, using 6 natural drinking waters which ranged from 42–1,725 ppm of total dissolved solids. Beverages made from solutions containing carbonates were the least desirable, having flat, insipid characteristics. Although distilled water resulted in an acceptable soft drink, coffee and tea prepared from it were excessively sour and astringent, respectively. The recommended formula of 53.3 g of coffee per liter of water was considered too strong and “burnt’ for the panel of trained judges, so that the experiments were repeated at a lower concentration of 47 g coffee/liter water. Large differences in the direction and magnitude of the visual characteristics of the coffee and tea brewed from the various waters also were observed in the marked changes in luminous transreflectance, purity and shifts in dominant wavelengths measured by thin-layer reflectometry.

    Notes: The global minimum on the water trimer potential energy surface has been investigated by means of second-order Møller-Plesset (MP2) perturbation theory employing the series of correlation-consistent basis sets aug-cc-pVXZ (X = D, T, Q, 5, 6), the largest of which contains 1329 basis functions. Definitive predictions are made for the binding energy and equilibrium structure, and improved values are presented for the harmonic vibrational frequencies. A value of 15.82±0.05 kcal mol−1 is advanced for the infinite basis set frozen core MP2 binding energy, obtained by extrapolation of MP2 correlation energies computed at the aug-cc-pVQZ MP2 geometry. Inclusion of core correlation, using the aug-cc-pCV5Z basis set, has been found to increase the binding energy by 0.08 kcal mol−1, and after consideration of core correlation and higher-order correlation effects, the classical binding energy for the water trimer is estimated to be 15.9±0.2 kcal mol−1. A zero-point vibrational correction of −5.43 kcal mol−1 has been computed from aug-cc-pVTZ MP2 harmonic vibrational frequencies. The accuracy of different computational schemes for obtaining the binding energies of the water dimer and trimer has been investigated, and computationally feasible methods are suggested for obtaining accurate structures and binding energies for larger water clusters.© 1999 American Institute of Physics.

    Notes: The long-standing problem of the topography, energetics, and vibrational dynamics of the ground-state surface of SiC2 is systematically investigated by means of the gamut of state-of-the-art electronic structure methods, including single-reference correlation techniques as extensive as the coupled-cluster singles and doubles method augmented by a perturbative triples term [CCSD(T)], the Brueckner doubles method (BD) with analogous contributions from both triple and quadruple excitations [BD(TQ)], and second-through fifth-order Møller–Plesset perturbation theory (MP2–MP5), as well as the multiconfigurational complete-active-space self-consistent-field [CASSCF(12,12)] approach. The one-particle basis sets for these studies ranged from Si[6s4p1d], C[4s2p1d] toSi[7s6p4d3f2g1h], C[6s5p4d3f2g1h]. The methodological analysis resolves the polytopism problem regarding the mercurial potential energy surface for the circumnavigation of Si+ aboutC2− in silicon dicarbide, whose topography is shown to exhibit almost all conceivable variations with level of theory. It is concluded that the X˜ 1A1 global minimum of SiC2 is a T-shaped(C2v) structure connected monotonically to a linear transition state 5.8 kcal mol−1 higher in energy, thus ruling out any metastable linear isomer. Previously undocumented bent transition states and L-shaped minima are encountered at relatively high levels of theory, but ultimately these stationary points are shown to be spurious. High-level focal-point thermochemical analyses yieldD0(Si–C2)=151 kcal mol−1, and hence a substantial revision is made in the heat of formation, viz.,ΔHf,0&convolu;(SiC2)=+155 kcal mol−1. A complete quartic force field about the T-shaped minimum is determined at the CCSD(T) level with the aug-cc-pVTZ (Si[6s5p3d2f],C[5s4p3d2f]) basis set and then employed in a preliminary probe of contours for large-amplitude motion, anharmonicity of the vibrations, and zero-point effects on the molecular structure. © 1997 American Institute of Physics.

    Notes: Excitatory amino acid transporters (EAATs) maintain the balance between pathological and physiological conditions by limiting the extracellular concentration of glutamate within the CNS and thus preventing excitotoxic injury. The loss of EAAT2 has been associated with the development of neurological diseases such as amyotrophic lateral sclerosis. It has therefore been suggested that the over-expression of specific EAATs may provide some degree of neuroprotection. However, the inability to isolate and study the function of the different EAAT isoforms in a cell type-specific manner has made it difficult to determine the exact contribution of individual EAATs toward neuroprotection or neurodegeneration in the context of excitotoxic injury. To address this question, we transduced hippocampal slice cultures from 1-week-old C57B/6 mice with recombinant adeno-associated virus carrying an EAAT2 gene expression cassette. EAAT2 gene expression was driven in neurons with the neuron-specific enolase promoter. Using this model system, we were able to induce a significant increase in the expression of functional EAAT2. Consequently, a significant increase in CA1 neuronal damage was observed in slices over-expressing EAAT2 in neurons following an acute exposure to exogenous glutamate. These data suggest that the increased expression of EAAT2 within neurons may contribute to neurodegeneration.

    Notes: Risk factors for citation errors in peer-reviewed nursing journals Background. Citation errors are common among nursing journals. But, there are no data regarding the factors that predispose to these errors. Objective. To determine the risk factors that predispose to citation error in peer- reviewed nursing journals. Methods. Five hundred and fifty references were selected randomly from articles published in eleven nursing journals for the year 1998. The incidences of major and minor citation errors were determined by comparing with the original articles. The relative odds of citation errors for the number of authors, collaborating institutions and the length of the reference list were calculated. The correlation between the scientific quality of the journal (by means of journal impact factor and immediacy index) and the incidence of citation error were also determined. Results. The incidence of citation errors is comparable to those reported previously. Long reference lists in articles written by a single author predicted strongly the occurrence of minor citation errors. Journals with a high impact factor and immediacy index tend to contain fewer minor mistakes. None of these factors affect the incidence of major errors. Conclusion. Contributors to journals should be aware of the various risk factors for citation errors. Citation accuracy may be improved by modifying these factors.
