    Notes: Summary Electron microscopic studies of ultrathin serial sections of the perikarya and adjacent neuropil of neurons of the supraoptic nucleus (SON) of the adult male rat revealed varying forms of two types of somatic spines. One type forms synapses with axons passing the cell, the other, without synapses, appears to serve as a buttress or clasp for adjacent neuronal and glial processes. The synapse-bearing spines lack the usual spine apparatus but contain the flocculent substance often seen in spines. The other spines do not exhibit either of these structures. These somatic spines were also seen in Golgi impregnated preparations but the types could not be distinguished. Certain axons synapse either on a somatic spine of the perikarya or penetrate the glial sheath of the neuron and synapse, usually repeatedly, on the soma in an en passant manner.

    Notes: A comparison of results of measuring supraoptic neuron activity was made in normal and in dehydrated rats by the following methods; (1) Measuring the nucleolar diameter of the neurons with a filar micrometer, with an image splitting eyepiece and, with the latter instrument, using the “comparative technique” - all three methods yielded statistically significant differences. (2) Measuring the concentration of cresyl violet in the nucleoli with a microspectrophotometer - a statistically significant difference was found. Also included for comparison were the results of a previous experiment in which the ribonucleic acid and succinic dehydrogenase content of the supraoptic nucleus was measured with a spectrophotometer following chemical extraction. The RNA was found to increase significantly but not the succinic dehydrogenase. Of the methods used for measuring nucleolar diameters the “comparative technique” appeared to be the most rapid from the practical viewpoint.

    Notes: With the object of investigating the so-called “feed-back” mechanism whereby hormones from the various endocrine glands presumably influence the hypothalamus in its regulation of the hypophyseal tropic hormones, the effect of hypophysectomy, ovariectomy, thyroidectomy, adrenalectomy and adrenal medullectomy on the nucleolar size of neurons in 16 hypothalamic nuclei was studied. A total of forty-five female albino rats was used. The nucleoli were measured under oil by means of a Cooke AEI image splitting eyepiece. For every rat 50 nucleoli in each of the 16 hypothalamic nuclei was measured, a total of 36,000 measurements. Hypophysectomy was found to affect all of the nuclei either after a short period (5 days) or after 60 days. Ovariectomy (20 days) caused nucleolar size changes in six nuclei, thyroidectomy (30 days) in eight, adrenalectomy (14 days) in all of the nuclei and adrenal medullectomy (30 days) in all of the nuclei. The results indicate that the absence of the various hormones does affect the activity of hypothalamic neurons as determined by nucleolar size changes. The use of the present methods also appears to “localize” centers for specific activities in the hypothalamus in some instances (e.g. ovariectomy) but not in others (e.g. adrenalectomy).
