    Notizen: NB-2, a member of the contactin subgroup in the immunoglobulin superfamily, is expressed specifically in the postnatal nervous system, reaching a maximum level at 3 weeks postnatal. NB-2 displays neurite outgrowth-promoting activity in vitro. To assess its function in the nervous system, we generated mutant mice in which a part of the NB-2 gene was ablated and replaced with the tau-LacZ gene. The general appearance of NB-2-deficient mice and their gross anatomical features were normal. The LacZ expression patterns in heterozygous mice revealed that NB-2 is preferentially expressed in the central auditory pathways. In the audiogenic seizure test, NB-2-deficient mice exhibited a lower incidence of wild running, but a higher mortality rate than the wild-type littermates. c-Fos immunohistochemistry demonstrated that neural excitability induced by the audiogenic seizure test in the NB-2-deficient mice was prominently attenuated in both the dorsal and external cortices of the inferior colliculus, where enhanced neural excitability was observed in the wild-type mice. In response to pure-tone stimulation after priming, NB-2-deficient mice exhibited a diffuse and low level of c-Fos expression in the central nucleus of the inferior colliculus, which was distinctly different from the band-like c-Fos expression corresponding to the tonotopic map in the wild-type littermates. Taken together, these results suggest that a lack of NB-2 causes impairment of the neuronal activity in the auditory system.

    Notizen: Interleukin (IL)-18, a member of the IL-1 cytokine family, is an important mediator of peripheral inflammation and host defence responses. IL-1 is a key proinflammatory cytokine in the brain, but the role of IL-18 in the CNS is not yet clear. The objective of this study was to investigate the actions of IL-18 on mouse glial cells. IL-18 induced intracellular expression of IL-1α and proIL-1β, and release of IL-6 from mixed glia. Treatment of lipopolysaccharide-primed microglia with adenosine triphosphate (ATP), an endogenous secondary stimulus, induced IL-1β and IL-18 release. Although deletion of the IL-18 gene did not affect IL-1β expression or release in this experimental paradigm, IL-1β knockout microglia released significantly less IL-18 compared to wild-type microglia. In addition, ATP induced release of mature IL-1β from IL-18-primed microglia. These data suggest that IL-18 may contribute to inflammatory responses in the brain, and demonstrate that, in spite of several common features, IL-18 and IL-1β differ in their regulation and actions.

    Notizen: Various bacterial pathogens have been identified as mediators of apoptosis. Apoptosis reportedly shows both detrimental and beneficial effects on biological functions. We studied the role of liver apoptosis in lethal Listeria monocytogenes infection and the regulation of apoptosis by endogenous cytokines during infection. Apoptosis was observed in the spleen but not in the liver of infected mice, whereas the induction of liver necrosis was evident by rising levels of serum aminotransferases in these animals. Apoptosis was detected in the liver of L. monocytogenes-infected mice which had been treated with monoclonal antibody (mAb) against tumor necrosis factor-α (TNF-α) or interleukin-6 (IL-6), or in TNF-α−/− mice, but not in γ- interferon (IFN-γ)−/− mice or mice which had been treated with mAb against IL-4 or IL-10. Augmentation of liver apoptosis in mice treated with mAb against TNF-α or IL-6 or in TNF-α−/− mice correlated with the increase in bacterial numbers in the organ, while no augmentation of apoptosis was observed in the liver of IFN-γ−/− mice irrespective of the marked increase in bacterial numbers in the organs, indicating that augmentation of liver apoptosis may not be merely due to the increase in bacterial growth in the organs. These results suggest that TNF-α and IL-6 may play an important role in protecting the liver from apoptosis in lethal L. monocytogenes infection.

    Notizen: Summary As an effort to elucidate the control of quality and quantity of the DNA-dependent RNA polymerase in Escherichia coli, the rate of synthesis of the individual subunits was determined during shift-up and- down of nutrients. When the strain B/r grown in a succinate medium was imposed to a shift-up by adding a mixture of glucose and amino acids, rapid rise was observed of the differential rates of the synthesis of α, β and β′ subunits, the constituents of core enzyme, leading to the increase of core polymerase concentration. The differential rates decreased thereafter to the level characteristic of the post-shift rate of cell growth. Compared to the strain B/r, the adaptation was slow in the strain K12 W3350. On the other hand, upon transfer of the strain B/r from a glucose-amino acids medium to a glucose medium lacking amino acids, the differential rate of core polymerase synthesis decreased rapidly and then regained the rate characteristic of the new growth rate. Similar control was also observed on the rate of ribosomal protein synthesis suggesting the coordinate expression of genes for the core polymerase subunits and ribosomal proteins. Thus, the intracellular concentration of RNA polymerase as well as of ribosomes might be one of the most important factors that affect the rate of bacterial growth. The rate of σ subunit synthesis, however, exhibited little change during the shift-up but a considerable decrease was observed during the shift-down.

    Notizen: Summary In an effort to elucidate the control of synthesis of the DNA-dependent RNA polymerase in Escherichia coli, intracellular amounts of the individual subunits were determined by polyacrylamide gel electrophoresis of cell lysates and of precipitates formed with specific antibody against holoenzyme I. Polyacrylamide gel electrophoresis of cell lysates in the presence of sodium dodecyl sulfate has been believed to separate the two larger subunits, β and β′, of RNA polymerase from the bulk of protein in E. coli. However, a polypeptide unrelated to the polymerase was found to migrate in the immediate vicinity of the β′ subunit, interfering with the accurate measurement of this subunit. Taking account of the presence of this peptide, designated tentatively as χ, the quantity of RNA polymerase was estimated relying only on the β subunit. Subunits content was also measured by polyacrylamide gel electrophoresis of the precipitates formed by treating cell lysates with anti-holoenzyme I serum. Since the isolated individual subunits as well as the holo- and core enzyme could be precipitated by the antibody, the present procedure permitted to determine total amounts of the subunits within cells. The subunits α, β and β′, the constituents of core portion of RNA polymerase, were found to be produced coordinately during steady-state growth at different rates within the range examined (0.22 to 1.87 generations/hr) and to be metabolically as stable as the bulk of protein. The rate of synthesis of these subunits relative to the total protein was found to be balanced with the growth rate; the differential rate of synthesis of enzyme core (αp) can be represented by the following empirical equation: αp(%) = 0.7 μ + 0.45, where μ represents growth rate (generation/hr). In contrast, the content of σ subunit was considerably small, i.e. only about one third mole equivalent to enzyme core, and was almost unaffected by the rate of growth.

    Notizen: Abstract To analyze the effect of human T-cell leukemia virus type I (HTLV-I) on cellular gene expression and its relation to tumorigenesis, two lines of transgenic mice carrying the long terminal repeat (LTR)-env-pX-LTR regions of the HTLV-I genome were produced. The transgene was expressed in many organs, including the brain, salivary gland, spleen, thymus, skin, muscle, and mammary gland. We found that the expression of the c-fos and c-jun genes, but not of thelyn and c-myc genes, was augmented 2- to 20-fold in histologically normal skin and muscle of these mice. The augmentation was tissue specific, suggesting the involvement of a cellular factor in the transgene action. In these mice, a three to seven times higher incidence of tumors was seen as compared with the control mice. These tumors included mesenchymal tumors, such as fibrosarcoma, neurofibroma, and lipoma, and adenocarcinomas of the mammary gland, salivary gland, and lung. The c-fos and c-jun genes were also activated in these tumors. The possible roles of elevated c-fos and c-jun gene expression in tumorigensis are discussed.
