    Notes: Abstract Measurements of trajectories of individual proteins or lipids in the plasma membrane of cells show a variety of types of motion. Brownian motion is observed, but many of the particles undergo non-Brownian motion, including directed motion, confined motion, and anomalous diffusion.     Notes: Current models for cellular plasma membranes focus on spatial heterogeneity and how this heterogeneity relates to cell function. In particular, putative lipid raft membrane domains have been postulated to exist based in large part on the results that a significant fraction of the membrane is detergent insoluble and that molecules facilitating key membrane processes like signal transduction are often found in the detergent-resistant membrane fraction. Yet, the in vivo existence of lipid rafts remains extremely controversial because, despite being sought for more than a decade, evidence for their presence in intact cell membranes is inconclusive. In this review, a variety of experimental techniques that have been or might be used to look for lipid microdomains in intact cell membranes are described. Experimental results are highlighted and the strengths and limitations of different techniques for microdomain identification and characterization are assessed.

    Notes: A means of determining quantitative maps of the tractions exerted by locomoting cells on a substratum has been developed. This method is similar to the Harris silicone substratum assay [Harris et al., 1980: Science 208:177-179], but uses an improved non-wrinkling film that deforms more predictably in response to traction forces. The method also utilizes a mathematical analysis of rubber deformation to produce the final map of the distribution of tractions. The resulting maps consistently showed that fish keratocytes exert a steady-state “pinching” on the substratum, perpendicular to the cell's direction of locomotion. No significant rearward tractions were detected at or near the front edge of the cell. Likewise, no significant forward tractions associated with peeling of adhesions were found at the back of the cell. A second assay uses deflection of a lightly attached glass microneedle to measure the total force exerted by locomoting cells. Forces of approximately 4.5 × 10-3 dyn were required to “stall” locomoting keratocytes. The implications of these findings for cell movement are discussed.

    Notes: Lateral diffusion measurements, using the photobleaching techniques, have provided unique and quantitative data on the random translational motions of proteins and lipids of membranes. Proper interpretation of this body of data can yield new insight into the structure of biomembranes. A comparative review of the lateral diffusion of membrane components in artificial lipid bilayers and of the same components in natural membranes is presented to demonstrate the effects of protein concentration and peripheral constraints on lateral mobility. Recent data on the effects of cell-substrate and cell-cell contact on lateral diffusion are reviewed. Finally, some experimental perspectives are offered in terms of emerging biophysical and biological technology.
