    Notes: Progression factors for periodontal diseases have been suggested by in vitro study of peripheral blood and gingival cells; however, those factors are not established in vivo. This investigation assessed biopsies of three groups of gingival tissues: those adjacent to a 1) 〈inlineGraphic alt="leqslant R: less-than-or-eq, slant" extraInfo="nonStandardEntity" href="urn:x-wiley:09042512:JOP101:les" location="les.gif"/〉3 mm (normal), 2) 4-6 mm, and 3) 〈6 mm gingival sulcus, to determine changes in the gingival microenvironment coincident to the progression of periodontal disease. Superoxide dismutase (SOD) and catalase activity, and IL-12 and bc1-2 levels, were decreased at 〉6 mm; total protein and IL-6 concentrations were increased adjacent to 〉6 mm, as compared to 〈3 and 4-6 mm, sites. Apoptotic cells were evident only within gingiva adjacent to 〉6 mm sites. These data suggest that IL-12 is an important factor in the shift from a TH1 to TH2 cell profile and that a favorable gingival microenvironment for hyper-inflammation may develop coincident to progression of periodontal diseases due to decreased bcl-2 and increased IL-6 concentrations within gingiva. These changes in the gingival microenvironment could impair apoptosis and promote enhanced release of reactive oxygen species (ROS) by phagocytes; decreased catalase and SOD activity could promote accumulation of ROS and result in additional tissue destruction.

    Notes: The elastic properties of the periodontal ligament have been attributed, in part, to oxytalan fibers, as no other types of elastic fibers are described there. It has been difficult to study the periodontal elastic meshwork by standard microscopic techniques because it is partially obscured by the adjacent periodontal ligament collagen fibers. Our study employed methods which either completely or partially removed mandibular molar periodontal ligament collagen fibers, exposing a previously undescribed periodontal elastic meshwork. The periodontal elastic meshwork was composed of many elastin lamellae containing both peripheral microfibrils of regular arrangement and central microfibrils of irregular arrangement, which could only be demonstrated in oxidized tissues. Peripheral, regularly arranged bundles of microfibrils resembled oxytalan fibers, which were often adherent to the border of the elastin lamella. Elastin lamellae containing irregular microfibrils resembled elaunin fibers. These fibers probably enclosed either blood vessels, nerves or collagen fiber bundles. Peripheral microfibrils attached elaunin to cementum, alveolar bone, blood vessels, and principal periodontal collagen fibers. Thus, the periodontal elastic meshwork is composed of both oxytalan and elaunin fibers. Microfibrils attach elaunin fibers to the adjacent non-elastic tissue and also form bundles which traverse the periodontal ligament space and are probably the oxytalan fibers demonstrable by light microscopic techniques. This meshwork of oxytalan and elaunin fibers probably contributes to tooth support and maintenance of periodontal homeostasis by dissipating chewing forces and maintaining patency of periodontal blood vessels.

    Notes: Abstract: Gingival overgrowth commonly occurs coincident to therapy with calcium channel blockers. The biologic mechanism for this condition is unknown; however, many clinicians suggest that poor oral hygiene may contribute to development of the overgrowth. This study tests the hypothesis that collagenous protein synthesis by gingival fibroblasts is synergistically enhanced when they are exposed to both nifedipine (N) and the pro-inflammatory cytokine, interleukin-1-beta, a cytokine expressed in inflamed gingiva. Human gingival fibroblasts were isolated from biopsies of normal gingiva and cells separated into two groups. Group 1 was exposed to media containing 0, 5, 50, or 500 pg/ml IL-1-beta, or 10−7 M N for 7 days; Group 2 was exposed to those concentrations of IL-1-beta +10−7 M N. [3H]-proline was added to the medium for the final 24 h. Cells and matrix were harvested and radioactivity determined by liquid scintillation analysis. Means (d.p.m./103 cells) were compared by factorial ANOVA and Scheffè comparisons. Collagenous protein synthesis was significantly reduced by 5 pg/ml IL-1-beta +10−7 M N and enhanced by 500 pg/ml IL-1-beta +10−7 M N as compared to N or IL-1-beta alone. Thus, patients may be more susceptible to gingival overgrowth coincident to nifedipine therapy as a result of the synergistic enhancement of connective tissue synthesis by these agents.

    Notes: The purpose of this study was to compare the efficiency of protein elution from several types of gingival crevicular fluid (GCF) collection papers when the volume of the inoculated protein and the elution methods were constant. Various concentrations of bovine serum albumin (BSA) and 14C-BSA were placed onto strips of Whatman #1 [W1], Whatman 3 MM chromatographic [W3]. Periopaper™ (ProFlow) [P] and Periopaper™ (Harco) [H], and recovered proteins measured following a non-optimized centrifugal elution technique. There were significant differences in % recovery of BSA and 14C-BSA from the papers, which was dependent on both the type of paper and the concentration of the inoculated protein; that is, proteins at the lowest concentrations were less efficiently eluted from GCF collection papers than those at higher concentrations. Equations for regression lines of elution efficiency were quadratic. Thus, our data suggest significant differences in the efficiency for elution of BSA from absorbent papers when the volume of the inoculated fluid and the elution technique were constant. Previous variable or conflicting experimental data between research groups may have resulted from incomplete elution of proteins from GCF collection papers, possibly due to entrapment within, or binding of GCF proteins to the paper.

    Notes: Abstract Patients scheduled to undergo tonsillectomy were administered 500 mg oral azithromycin as two 250 mg capsules given 12 h apart. Between 9 h and one week after the second dose, tonsil samples were taken during surgery and assayed for azithromycin. Mean concentrations in tonsillar tissue, 12 and 24 h after the second of the two 250 mg doses given 12 h apart, were 4.5 and 3.9 µg/g, respectively. Concurrent mean serum concentrations were approximately 0.03 and 0.01 µg/ml, respectively. The mean concentration in tonsillar tissue 7.5 days after the last dose was 0.93 µg/g. The apparent half-life of drug in the tissue was 76 h. The ratio of mean concentration in tissue to that in serum was greater than 150-fold for all time intervals. The presence of high azithromycin concentrations in tonsillar tissue suggests that a once-daily regimen over five days or less may be effective in treating tonsillo-pharyngitis.

    Notes: Abstract The aurorae are the result of collisions with the atmosphere of energetic particles that have their origin in the solar wind, and reach the atmosphere after having undergone varying degrees of acceleration and redistribution within the Earth's magnetosphere. The global scale phenomenon represented by the aurorae therefore contains considerable information concerning the solar-terrestrial connection. For example, by correctly measuring specific auroral emissions, and with the aid of comprehensive models of the region, we can infer the total energy flux entering the atmosphere and the average energy of the particles causing these emissions. Furthermore, from these auroral emissions we can determine the ionospheric conductances that are part of the closing of the magnetospheric currents through the ionosphere, and from these we can in turn obtain the electric potentials and convective patterns that are an essential element to our understanding of the global magnetosphere-ionosphere-thermosphere-mesosphere. Simultaneously acquired images of the auroral oval and polar cap not only yield the temporal and spatial morphology from which we can infer activity indices, but in conjunction with simultaneous measurements made on spacecraft at other locations within the magnetosphere, allow us to map the various parts of the oval back to their source regions in the magnetosphere. This paper describes the Ultraviolet Imager for the Global Geospace Sciences portion of the International Solar-Terrestrial Physics program. The instrument operates in the far ultraviolet (FUV) and is capable of imaging the auroral oval regardless of whether it is sunlit or in darkness. The instrument has an 8° circular field of view and is located on a despun platform which permits simultaneous imaging of the entire oval for at least 9 hours of every 18 hour orbit. The three mirror, unobscured aperture, optical system (f/2.9) provides excellent imaging over this full field of view, yielding a per pixel angular resolution of 0.6 milliradians. Its FUV filters have been designed to allow accurate spectral separation of the features of interest, thus allowing quantitative interpretation of the images to provide the parameters mentioned above. The system has been designed to provide ten orders of magnitude blocking against longer wavelength (primarily visible) scattered sunlight, thus allowing the first imaging of key, spectrally resolved, FUV diagnostic features in the fully sunlit midday aurorae. The intensified-CCD detector has a nominal frame rate of 37 s, and the fast optical system has a noise equivalent signal within one frame of ∼ 10R. The instantaneous dynamic range is 〉1000 and can be positioned within an overall gain range of 104, allowing measurement of both the very weak polar cap emissions and the very bright aurora. The optical surfaces have been designed to be sufficiently smooth to permit this dynamic range to be utilized without the scattering of light from bright features into the weaker features. Finally, the data product can only be as good as the degree to which the instrument performance is characterized and calibrated. In the VUV, calibration of an an imager intended for quantitative studies is a task requiring some pioneering methods, but it is now possible to calibrate such an instrument over its focal plane to an accuracy of ±10%. In summary, very recent advances in optical, filter and detector technology have been exploited to produce an auroral imager to meet the ISTP objectives.
