    Notes: Abstract Purpose: Pancreatic cancer is widespread, associated with high mortality, and rapidly fatal. Most cases are diagnosed too late for surgical treatment, and the disease responds poorly to systemic chemotherapy. Nevertheless, pancreatic cancer cells are sensitive to fluorouracil (5-FU) in a time- and dose-dependent manner, suggesting that improved retention of drug in the tumor may improve patient prognosis. In this study, we evaluated a novel drug delivery system, 5-FU/epinephrine injectable gel (5-FU/epi gel), designed to improve drug retention in tumors. Methods: We used a BxPC-3 human pancreatic cancer xenograft model in athymic mice to examine drug levels in tumor, liver, and kidney tissue following administration of: (a) 5-FU/epi gel (30 mg 5-FU/ml) intratumorally (i.t.); (b) 5-FU solution i.t.; and (c) 5-FU solution intraperitoneally (i.p.). [3H]5-FU was added as a radiolabeled marker to all test formulations. Animals were sacrificed at designated times, and the tumor, liver, and one kidney from each animal were excised and processed for radioactivity analysis. Drug concentration was quantified by both storage-phosphor autoradiography (SPA) and liquid scintillation counting (LSC). Results: Higher and sustained i.t. drug levels were achieved following i.t. administration of 5-FU/epi gel (SPA AUC 18.4 mM · h, LSC AUC 13.0 mM · h) compared with 5-FU solution i.t. (SPA AUC 2.02 mM · h, LSC AUC 1.92 mM · h) or 5-FU solution i.p. (SPA AUC 0.07 mM · h, LSC AUC 0.04 mM · h). Use of the 5-FU/gel system was associated with lower drug levels in liver and kidney, indicating that it produces far less systemic exposure. Conclusion: In the human pancreatic cancer xenografts, i.t. administration of 5-FU/epi injectable gel provided significantly higher drug and/or metabolite concentrations for extended periods than was possible with either i.t. or i.p administration of drug solution. This i.t. drug delivery system could potentially be used to treat patients with pancreatic cancer to increase tumor exposure to drug and improve the therapeutic index in comparison to systemic drug administration.

    Notes: Summary The distribution of immunoreactive arginine vasotocin (AVT-ir) was determined in the brain of the lizard Anolis carolinensis. Cells and fibers containing AVT-ir were found in the medial septal region, lamina terminalis, lateral forebrain bundle, preoptic area, supraoptic nucleus, anterior hypothalamus, paraventricular nucleus, periventricular nucleus, arcuate nucleus, and ventromedial nucleus of the thalamus. Occasional AVT-ir cells were found in the interpeduncular nucleus. Fibers containing AVT-ir were found in the cortex, around the olfactory ventricle, in the diagonal band of Broca, amygdala area, dorsal ventricular ridge, striatum, nucleus accumbens, septum, ventromedial hypothalamus, lateral hypothalamus, medial forebrain bundle, median eminence, pars nervosa, nucleus of the solitary tract, locus coeruleus, cerebellar cortex (granular layer), dorsal part of the nucleus of the lateral lemniscus, substantia nigra, and myelencephalon. The intensity of AVT-ir staining was, in general, greater in males than in females. Comparison of AVT-ir distribution in A. carolinensis with those previously published for other reptilian species revealed species-specific differences in distribution of AVT.

    Notes: Summary Using traditional as well as whole-mount immunohistochemistry, we described the location of tyrosine hydroxylase-and dopamine beta hydroxylase-positive cells and fibers in the brain of the lizard Anolis carolinensis. Major catecholaminergic cell groups were in the ependyma in certain ventricular regions, alous coeruleus, anterior hypothalamic and lateral hypothalamic areas, and in the mesencephalic tegmental region, locus coeruleus, nucleus of the solitary tract, vagal motor nucleus, and rhombencephalic reticular formation. Major catecholaminergic fibers, tracts and varicosities included tuberohypophysial, mesolimbic, nigrostriatal, isthmocortical, medullohypothalamic, and coeruleospinal systems. Although the catecholaminergic systems in A. carolinensis are similar to those in the brains of other lizards studied, there are a few species differences. Our information about A. carolinensis will be used to help localize the hypothalamic asymmetry in catecholamine metabolism previously described in this lizard.

    Notes: Numerous tissue mast cells are present in the ovarian medulla and hilus and in the oviduct of rats. In the medulla, most of these mast cells are in the connective tissue of the stroma near blood and lymphatic vessels. During proestrus, many of the medullary mast cells totally degranulate and thus are not visible histochemically; they then regranulate during estrus. In contrast, the number of stainable mast cells in the ovarian hilus and oviduct does not change during the estrous cycle. Histofluorometric methods demonstrate that mast cells in the ovarian medulla and hilus, as well as the oviduct, contain histamine. In addition, the lining of small blood vessles in the ovarian medulla contains histamine. Thus, mast cell and blood vessel histamine secretion may play a role in ovarian function.

    Notes: The urinogenital blood vessels of Anolis equestris and A. carolinensis were studied using dissection, injection of silicone rubber, and histology. A new blood-vascular connection between small veins in the pampiniform plexus at the base of the ovary and an ostial extension of the secondary oviductal vein is described. Chemical messengers could thus be carried in the blood from the ovarian venous drainage to the oviduct. Furthermore, close apposition of the secondary oviductal vein and artery provides an anatomical basis for countercurrent exchange of these messengers. A circular venous arcade, involving the supracardinal vein and branches of the oviductal veins, also is described; this arcade could serve to deliver adrenal hormones to each oviduct.

    Notes: The house gecko (Hemidactylus frenatus) exhibits an ovarian cycle that can be divided into early follicular, vitellogenic, and luteal phases. Serial sections through the right ovary of animals in the three phases allowed us to quantify follicular size, condition, and number, as well as germinal bed activity. There are six to eight healthy, growing follicles in each ovary, arranged in a stepwise size hierarchy. This number does not vary among the three phases, even though one follicle becomes atretic and one ovulates during each cycle. Therefore, compensatory follicular hypertrophy occurs, leading to replacement of lost follicles and maintenance of the follicular size hierarchy.

    Notes: The architecture of follicular blood vessels in the ovary of lizards (Anolis equestris and Anolis carolinensis) was studied by standard histology and also after vascular perfusion with an orange silicone-rubber compound or with India ink. The theca of the follicular wall contains a netlike arrangement of anastomosing sinusoids, which increase in size as a follicle grows. An avascular stigma forms in very small, growing follicles when a portion of the follicular wall contacts the ovarian surface epithelium. Blood vessels then invade the theca except in the zone of contact. The diameter of the stigma is about 50% of follicular diameter, regardless of follicular size. Although the stigma of smaller follicles is avascular, that of vitellogenic follicles is hypovascular, i.e., a few vessels radiate into the stigma region. The antiangiogenic process involved in stigma formation may continue as the stigma enlarges. The development pattern of stigma formation found in Anolis is displayed by many other vertebrates.

    Notes: Ovarian follicular histology of the lizard Anolis carolinensis was studied in relation to follicular size and rank. The structure of the Anolis follicular granulosa is similar to that described in other reptiles. Changes in the granulosa during follicular hypertrophy are also similar to those described in other species. Follicles of low rank in the linear size-gradation exhibited atresia.Counts were made of erythrocytes per millimeter of thecal tissue in crosssections of follicles of different size and rank. Follicles greater than 2.6 mm in diameter exhibited a marked increase in the number of thecal erythrocytes. Also, follicles of a similar diameter tended to possess a greater number of thecal erythrocytes the highter the rank in the linerar size-gradation. These observations support the hypothesis that follicular selection involves appropriation of blood and blood-borne hormones by larger follicles.
