    Description / Table of Contents: Résumé Nous rapportons ici une procédure de purification en une étape de la laccase deNeurospora crassa. Elle consiste à employer la chromatographie sur Celite qui permet la concentration de l'enzyme exo-cellulaire à partir de grands volumes de milieu de culture. Cette méthode permet d'isoler n'importe quelle laccase fungique ou de plante ainsi que d'autres protéines à cuivre. Dans le présent travail, nous avons aussi tiré parti de l'induction au centuple de l'enzyme par de faibles concentrations en cycloheximide.
    Abstract: Resumen Aquí se reporta un procedimiento de purificación en un solo paso para le enzima lacasa deNeurospora crassa. Consiste en el empleo de la cromatografía en Celite que permite la concentración de la enzima exocelular de grandes volúmenes de cultivo. Este método es útil para aislar cualquier lacasa de hongos o plantas así como otras proteínas que contienen sobre. En este trabajo, hemos aprovechado también la inducción de la enzima en más de 100 veces por bajas concentraciones de cicloheximida.
    Notes: Summary A single-step purification procedure forNeurospora crassa laccase is reported. It used Celite chromatography which permits the concentration of the extracellular enzyme from large culture volumes. This method is useful to isolate any fungal or plant laccase as well as other coppercontaining proteins. This work also takes advantage of the 100-fold induction of the enzyme by low concentrations of cycloheximide.

    Notes: Summary Molecular properties of Neurospora soluble protein kinase and cyclic AMP binding activities have been determined. Cyclic AMP-dependent Peak I kinase activity with a molecular weight of 118 000 is composed of two subunits, catalytic and regulatory, having molecular weights of 55 000 and 57 000, respectively. Cyclic AMP-independent Peak II kinase activity has a molecular weight of 56 000. Cyclic AMP-independent Peak III kinase has a molecular weight of 209 000.

    Notes: Summary Incubation of Neurospora crassa mycelia with low doses of cycloheximide induces the expression of several genes. After 6 h in the presence of cycloheximide, mycelia become tolerant to further additions of the drug and the rate of protein synthesis exhibits a lower sensitivity to it. The polypeptide pattern is indicative of a stress situation.

    Notes: Abstract Escherichia coli recombinant strains bearing the thr operon have been previously selected for threonine production and phenotypically classified according to antibiotic resistance properties (Nudel et al. 1987). Further analysis of those strains permitted the isolation and restriction mapping of two different plasmids of 13 kb and 18.6 kb. The smaller one, which expressed tetracycline resistance gave better results on threonine accumulation but it was rather unstable when grown without antibiotic pressure. Therefore, other hosts were transformed with those plasmids to improve stability. A threonine-auxotrophic strain was a better host for plasmid maintenance and expression of thr operon. Host influence in plasmid-mediated threonine production was studied in terms of specific yields (the ratios of threonine accumulated to biomass values) and of plasmid maintenance (percent of AprTcr clones after cultivation in non selective media). We also determined that semisynthetic media of defined composition were better than rich media for threonine expression, due to feed-back controls exerted by undesired catabolites accumulated in complex media.
