    Notes: Summary The present findings show that both elastic system fibers and collagen markedly resisted change in tissues more than 2000 years old. The distribution of elastic fibers and elastic-related fibers (namely, oxytalan and elaunin fibers) in mummified tissues coincided with the observations made on the modern human tissues used as controls. The collagenous structures present in tissue sections obtained from the Egyptian mummy studied took on a deeply red colour when stained in the Picrosirius solution indicating that, as well as in the fresh controls, the basic groups in the collagen molecules were available for reacting with the strongly acidic dye Sirius Red. When viewed with polarized light, the collagen in the same tissue sections displayed an increased birefringence, which shows that the collagen molecules in mummified tissues maintain the oriented disposition which is typical of the modern human tissues used as controls. The methods employed have proved to be useful for the delineation of the elastic system fibers and of the collagenous scaffolding, which may be used as valuable landmarks in the study of the histoarchitecture of organs that have undergone considerable distortion.

    Notes: Zusammenfassung Es wurde der zeitliche Verlauf des Einbaus radioaktiver Aminosäuren (Phenylalanin-3-C14, Alanin-1-C14, Histidin-2-C14 und Methionin-S35) in die Proteine des Pankreassaftes der Ratte untersucht. Ein Maximum des Einbaus kommt zwischen der zweiten und vierten Stunde nach Injektion der Aminosäuren vor, während in der ersten Stunde praktisch keine radioaktiven Proteine nachgewiesen werden konnten. Die Ergebnisse werden verglichen mit denen früher beschriebener Versuche mit Glyzin-C14.

    Notes: Summary The influence of tissue section thickness on the color and intensity of birefringence displayed by collagen in tissue sections studied by means of the Picrosirius-polarization method, is reported in this paper. When dermal collagen sections of different thicknesses (ranging from 0.25 to 11 μm) were studied by this method, it became evident that not only did the intensity of birefringence increase proportionally to tissue section thickness, as was to be expected, but also a gradual shift in color from green through yellow to red could be observed as tissue section thickness increased. The limitations of the Picrosirius-polarization method for the localization of collagen types I, II, and III in routinely used histological slides is discussed, showing that this method is useful for the study of the distribution of the different types of interstitial collagen in normal adult vertebrate organs.

    Notes: Summary Normal skin biopsy specimens obtained from 5 representative species of mammals, including man, were studied by the silver impregnation technique for reticulin fibers, by the histochemical Picrosirius-polarization method which is specific for collagen, and by transmission electron microscopy. The finely woven meshwork of argyrophilic reticulin fibers present in the adventitial dermis showed characteristics which are typical of collagen type III, when studied by aid of the Picrosirius-polarization method and by electron microscopy. On the other hand, the coarse collagen fibers of the deeper layers displayed ultrastructural and histochemical aspects which are characteristic of collagen type I. The foregoing observations support previous biochemical and immunohistologic findings, strongly suggesting that collagen type III is found mainly in the upper dermis while collagen type I predominates in the deeper layers.

    Notes: Summary The results presented in this paper show that collagen fibers can be clearly distinguished from reticular fibers using the picrosirius-polarization method. A morphologic and morphometric study of these two types of fibers with electron microscopy shows that reticular fibers are characterized by the smaller diameter of their fibrillar components and the higher content of interfibrillar material, resulting in a loose arrangement of the fibrils. The evidences presented suggest that the amorphous matrix in which fibrils are embedded is responsible for the silver impregnation of reticular fibers. Our results show that the matrix of reticular fibers is characteristically rich in heparitin sulfate, and that the glycosaminoglycans present show a high interaction with the fibrillar component of these fibers.

    Notes: Zusammenfassung 1. Die operative Anlegung einer Pankreas-Fistel bei der Ratte wird beschrieben. 2. Nach einer Hungerzeit von 24 Std wurde eine Hunger-Sekretion gefunden im Mittel von 0,60 λ/min/kg Körpergewicht ± 0,089. Der Proteingehalt des extruierten Hungersaftes betrug im Mittel 52 γ/λ Sekret ± 11. Saftmenge und Proteingehalt liefen während der Hungersekretion nicht parallel. Die Saftmengen wurden mit Befunden an Hunden, Menschen und Kaninchen verglichen, mit dem Ergebnis, daß größere Säugetiere offenbar per Kilogramm Körpergewicht mehr Hungersaft ausscheiden als kleinere. 3. Die Ursachen der Hungersekretion werden erörtert. Nach früheren Untersuchungen wird die Hungersekretion nicht nervös und nicht hormonal reguliert. Die Lebendbeobachtung des Pankreas zeigt aber, daß die Zellen je eines Acinus sich während des Hungerns bis zu 1/3 des Zellvolumens mit Granula füllen und sich dabei vergrößern. Dadurch entsteht ein Überdruck in den Zellen. Dann lösen die Zellen etwa 80% ihrer Granula in 1/2 Std auf und extruieren den so gewonnenen Saft. Dieser rhythmisch sich wiederholende Vorgang ist die Ursache der Hungersekretion. Die Anzahl der extruierenden Acini ist während des Hungerns klein; nach einem Reiz groß. So ist es auch zu erklären, daß der Manometerdruck in Ductus pancreaticus während des Hungerns fast ebenso hoch ist wie nach einer Reizung des Pankreas.
