    Notes: Misonidazole, a hypoxic cell radiation-sensitizer, has been used in vivo to analyze its effects on normal and x-radiation epidermis. The drug action was evaluated 3 days after being implanted subcutaneously. The response of the epidermis and especially of the basal cells to the drug alone and in combination with x-radiation was studied. The ultramorphological analysis as well as quantitative data on epidermal thickness and keratinocyte volume showed that high doses of misonidazole impair the usual post-irradiation epidermal reaction, thus enhancing the involutional effects of radiation and inhibiting radiation-damage repair, even under conditions of normal oxygenation.

    Notes: The combined use of scanning electron-microscopy and chemical separation of epidermis permitted the study of the irradiated and normal hair follicle surface characteristics. Irradiation reduced the length and density of the hair follicles, producing at the same time marked alterations and deformities of the individual follicles. As most of these changes were dose- and time-dependent, these structures, studied by these techniques, provide an interesting model as biological indicators of radiation dose.

    Notes: The number of epithelial cells per unit volume of tissue (Nv) and the relative volume (Vv) occupied by the spinous and basal layers of X-irradiated and non-irradiated rat-tail epidermis were studied using techniques of stereologic cytology. By a simple calculation, the absolute volumes of basal and spinous keratinocytes were determined in irradiated (and in non-irradiated, control) epidermis, 24, 48, 72 and 96 h after exposure to a single dose of 16 Krads. During this post-irradiation period, the absolute volumetric values of both basal and spinous keratinocytes increased gradually, reaching a three-fold increase 96 h after irradiation. At the same time, the relative volume of the spinous layer increased from 58% to 75% of the total epidermis, whereas the volume fraction occupied by the basal layer decreased from 20% to 16%. These findings support previous evidence that lethally and sublethally damaged keratinocytes are capable of a metabolic effort resulting in a cell hypertrophy which in some circumstances could lead to repair and recovery from radiation damage.

    Notes: Standardized techniques of stereologic cytology were applied to normal human epidermis, in order to comparatively describe the suprabasal and the dark and clear basal keratinocytes. The results, expressed mainly in surface and volume densities of epidermal components, showed that some important stereologic parameters of dark basal keratinocyte constituents exhibited values which could be interpreted as intermediate between those of the clear basal keratinocytes and those of the suprabasal keratinocytes. Although the degenerative nature of dark cells cannot be ruled out, the stereologic characterization of their cell architecture also could indicate that dark basal cells are already differentiating keratinocytes still remaining in the basal layer.

    Notes: Specific-pathogen-free rats kept on a vitamin A-deficient diet for approximately 5 months developed cutaneous alterations, especially in the periorbital and eyelid skin. These lesions were characterized grossly by erythema, partial alopecia, and crusting, and histologically by hyperorthokeratinization, acanthosis, and a three-fold increase in incorporation of labeled DNA precursors into the epidermal basal layer. Electron microscopy showed an increased number of Langerhans cells with and without Birbeck granules, as well as the presence of cytoplasmic dense bodies of probable lipid nature in keratinocytes of all epidermal strata. The similarities of these ultrastructural alterations with some features of vitamin A-sensitive human dermatoses point to probable common mechanisms in the pathogenesis of these skin diseases.

    Notes: Abstract The immunoperoxidase method for involucrin detection was applied to the study of the maturation of epithelial lesions of the oral mucosa that included specimens of leukoplakia, lichen planus, verrucous carcinoma, carcinoma in situ and invasive carcinoma. Areas of orthokeratinized, parakeratinized, and non-keratinized normal mucosa were also studied. Normal orthokeratinized epithelia showed intracytoplasmic or pericellular staining in the suprabasal epithelia showed intracytoplasmic or pericellular standing in the suprabasal epithelial layers in a pattern similar to that of the normal epidermis. Parakeratinized and non-keratinized epithelia were less stained. Intense staining was observed in leukoplakia, whereas the staining of lichen planus was lessintense but exhibited a more homogeneous pericellular staining pattern than leukoplakia. Verrucous carcinoma was markedly and very irregularly stained. Carcinomas in situ and invasive carcinoma exhibited a slightly positive and patchy reaction. The distribution patterns of involucrin in the lesions correlated very well with the degree of epithelial differentiation. In addition, irregularpatchy distribution correlated with the degree of atypia, and was especially evident in carcinomas.

    Notes: Dark basal keratinocytes, characterized by a strong affinity for basic dyes and by electron density of cytoplasm and nucleus, could be recognized in eleven oral leukoplakias. The percentage of dark cells was higher in the group comprising leukoplakias verrucosa, and erosiva (28% of the basal cells) than in the leukoplakia simplex group (10%). The presence of these cells is a good indicator of the degree of histological dysplasia and correlates well with the preneoplastic potential of these lesions.
