    Notes: Background Annexin-1 (ANXA1, lipocortin 1) is a pleiotrophic protein produced by many cell types including peripheral blood leucocytes. Although it has been shown to inhibit ‘macroscopic’ inflammatory processes in animal models, its direct effects on antigen-activated human T cells have not been studied.Objective To test the hypothesis that ANXA1-derived peptides inhibit antigen-driven prototype Th1 and Th2-type human T cell responses of clinical relevance and lectin-driven responses in vitro.Methods Peripheral blood mononuclear cells (PBMC) were isolated from 14 atopic subjects sensitized to house dust mite allergen (Dermatophagoides pteronyssinus, Der p) and purified protein derivative (PPD) of Mycobacterium tuberculosis. PBMC (1 × 106/mL) were cultured with phytohaemagglutinin (PHA; 5 µg/mL; 4 days), Der p (25 µg/mL; 6 days), PPD (10 µg/mL, 6 days) or medium control. Two ANXA1-derived peptides, Ac2–26 and AF-2 (5–500 µm), were assessed for possible inhibition of PHA-and antigen-induced T cell proliferation (measured by 3H-thymidine uptake), while Ac2–26 was assessed for inhibition of Der p-induced interleukin (IL)−5 release and PPD-induced interferon-γ (IFN-γ) release (measured by ELISA). Comparison was made with dexamethasone as an established inhibitory control. Endogenous production by PBMC of cell surface-associated and intracellular ANXA1 in response to PHA, Der p and PPD in the presence and absence of dexamethasone was measured by specific ELISA.Results Both PHA- and antigen-induced T cellular proliferation were inhibited by dexamethasone. Although neither ANXA1-derived peptide significantly altered PHA-induced proliferation, both effected concentration-dependent reductions in antigen-induced proliferation, Ac2–26 being the more potent. Peptides of identical amino acid composition to Ac2–26 and AF-2, but of random sequence, were ineffective at equivalent concentrations. In addition, Ac2–26 and dexamethasone inhibited Der p-induced IL-5 release and PPD-induced IFN-γ release in a concentration-dependent fashion. Endogenous ANXA1 was detectable in PBMC, but at concentrations approximately 104-fold lower, in molar terms, than the effective concentrations of the exogenously added, ANXA1-derived inhibitory peptides. Endogenous production was not significantly altered by any of the T cell stimuli employed in this study, in the presence or absence of dexamethasone.Conclusion In prototype Th1 and Th2-type human T cell responses, ANXA1-derived peptides can inhibit antigen-driven cellular proliferation and cytokine production.

    Notes: Abstract.  Shock Waves (SW) were produced in air by focusing the (0.25 J, 6 ns) second-harmonic (λ=532 nm) Nd : YAG laser light into a stainless-steel cylindrical cell at a pressure from 200 to 760 Torr. The laser fluence at the focal point is 〉5 GW/cm2. The spatial variation and consequently the time evolution of the radial propagation velocity U of the generated shock waves were measured via a simple optical system utilizing a HeNe laser beam triply intersecting the propagating shock wave at three successive positions. Using a reflector, we were able to probe the traveling SW in six consecutive positions during its round trip. Good agreement was obtained between the experimental results and the predictions of the point strong explosion theory. It is shown that this method is simple with a fairly good precision. It therefore appears to be useful for the determination of the SW dynamic parameters, namely its Mach number, the pressure at the SW front, the thickness of the compressed air layer and the energy consumed in producing this layer.

    Notes: The trabeculae cranii are at first quite separate from each other, after few days their anterior two fifths are connected by a trabecular plate which is obliterated throughout development. The paired origin of the parachordal plate is not observed. The fused posterior orbital cartilages chondrify in the form of a wide short plate, traversed by the oculomotor and trochlear nerves. The basicranial fenestra and fenestra ovalis are formed by the degeneration of pre-existing cartilage. The cochlear portion is completely fused with the parachordal plate from the very beginning. The elements of the pterygoquadrate are fused together. The quadrate and Meckel's cartilage are in close contact from the very beginning. While the lower part of the interorbital septum is derived from the trabecula communis, its upper part is derived from the anterior orbital cartilages. The lateral parts of the fused posterior orbital cartilages give rise to most of the taeniae and pilae of the orbitotemporal region. There is only one commissure between the auditory capsule and parachordal plate. A cartilaginous connection between the distal portion of the columella auris and ceratohyal persists for some time. The parietotectal and paranasal cartilages are fused together from the very beginning. The processus paroticus originates from the columella auris.In the fully formed stage the notochord is completely embedded in the occipital condyle. The union between the condyle and odontoid process persists. The auditory capsules and occipital arches contribute to the formation of the tectum synoticum plus posterius. The prefacial commissure and facial foramen lie in front of the cochlear portion. The columella auris possesses a processus internus (connected with the quadrate), but the processes a dorsalis has completely disappeared. The orbitotemporal region is quite complete. A medial fenestra is formed in the planum supraseptale. A fenestra is observed in each of the interorbital and nasal septa. The lamina transversalis anterior is fused with the parietotectal cartilage. A complete zona annularis is present. The outer wall of the paranasal cartilage is perforated by a large fenestra lateralis. The parietotectal and paranasal cartilages and the posterior process of the lamina transversalis anterior contribute to the formation of the concha nasalis. There is a contact between the planum antorbitale and nasal septum. The pterygoid process has disappeared. The common characters of the lacertid chondrocranuium are deduced.

    Notes: The fully formed chondrocranium of the Egyptian snake, Psammophis sibilans has been described in detail. The tectum synoticum is very broad and there is apparently no tectum posterius. The notochord is completely embedded in the occipital condyle. The connection between the condyle and the odontoid process persists to the fully formed stage. There is only one commissure, representing the anterior basicapsular commissure, connecting the auditory capsule to the basal plate. The fisura metotica is continuous postero-dorsally with the fissura occipitocapsularis. The distal bent end of the shaft of the columella auris represents the processus dorsalis, and the nodule that separates from it and fuses with the quadrate is the intercalare. The interorbital septum, orbitosphenoid cartilages, basitrabecular processes and planum supraseptale are completely absent. The concha nasalis is of the simplified type, the active factor in its formation is the folding of the olfactory epithelium. The solum nasi is greatly reduced and comprises the floor of the cupola anterior, the composite lamina transversalis anterior, cartilago ectochoanalis, cartilago hypochoanalis and ectochonal plate. The branchial arches are completely absent. The pterygoquadrate is represented only by the quadrate cartilage. The ceratohyals are extremely long.

    Notes: The development of the chondrocranium of the Egyptian snake, Psammophis sibilans has been thoroughly studied in six developmental stages. The neurocranium and viscerocranium develop at the same time. In early stages the mesocephalic flexure is well represented, but later it is gradually reduced. The notochord is completely embedded inside the parachordal plate in the early and moderate stages, and the paired origin of the plate is not observed. From the very beginning the quadrate cartilage is in close contact with Meckel's cartilage. In early stages the two cartilages lie nearly at the same straight line, and throughout development the quadrate rotates freely until the angle between it and Meckel's cartilage becomes nearly a right one. It was impossible to find a stage at which the auditory capsules and their cochlear portions were separate from the parachordal plate. There is no interorbital septum and the platytrabic character of the chondrocranium is preserved. The basicranial fenestra, fenestrae X and the fenestra ovalis are formed by the resorption of pre-existing cartilage. The planum antorbitale has no separate center of chondrification. The rudiment of a simplified type of concha nasalis is formed in the last stage. The lamina transversalis anterior is formed of two separate portions. The tectum synoticum is of purely otic origin.
