    Notes: Summary A rare form of endocardial cushion defect is an atrioventricular (AV) canal-type ventricular septal defect with cleft mitral valve in the absence of a primum atrial septal defect. In two infants with Down's syndrome, preoperative delineation of this entity was accomplished. M-mode echocardiographic features included presence of two distinct AV valves, attachment of the anterior portion of the auterier leaflet of the mitral valve to the interventricular septum, and normal septal motion. Two-dimensional echocardiography demonstrated the intact atrial septum, ventricular septal defect, presence of two distinct AV valves, and the cleft mitral valve. Angled (axial) cineangiography was employed to delineate the AV valve morphological characteristics, position of ventricular septal defect, and absence of a primum atrial septal defect. Since the surgical approach to this lesion is different from other forms of endocardial cushion defects and specific problems related to AV valve morphology may be encountered, the preoperative diagnosis of this entity is important.

    Notes: Combined morphometric and autoradiographic methods were used to analyze the postnatal growth of rat lung from 1-21 days after birth. Each cell population had distinct growth patterns with an increase in the number of fibroblasts and capillary endothelial cells largely determining the increase in interstitial volume and capillary surface, respectively. The height of proliferation activity in mesodermally-derived cells was concurrent with the outgrowth of secondary alveolar septa (between days 4 and 13). Analysis of the location of labeled cells on day 7 showed that the higher labeling index on septal crests could be ascribed to the proliferative activity of fibroblasts and endothelial cells.Within the alveolar epithelium only the type II alveolar cells had a detectable labeling index. Over the first week, the number of type I epithelial cells steadily increased while the number of type II cells remained constant. Subsequently the number of type II cells increased rapidly, reached a peak on day 13 and then decreased, whereas type I cells continued to increase in number. These facts led us to consider that type II epithelial cells may represent the stem cell population of alveolar epithelium. The height of proliferative activity of type II cells on day 7 coincided with the outgrowth of septal crests and was followed by the steepest increase in number of type I and II cells.Between the 10th and 21st day labeling indices rapidly declined, cell production became undetectable after day 13. Increase in alveolar and capillary surface area however continued, resulting in a thinning of the interstitial layer and of the epithelial and endothelial sheets.

    Notes: Autoradiographic methods were used to analyze cell kinetics in peripheral epithelium of the fetal mouse lung, days 15-18. The 3H labeling index decreased in proportion to increases in fetal lung and body weight from day 15 to day 18. Cell-cycle determination estimated from the percent of labeled metaphases at intervals after 3H thymidine injection showed the shortest cell-cycle time (12.25 hours) on day 15 during the precenal stage of growth. Total cycle time (Tc) increased by approximately one hour on each subsequent day. Between days 15 and 16 Tc increased due to a 1-hour increase in the post mitotic interphase (G1); thereafter increases in Tc could be ascribed almost entirely to lengthening of the DNA synthetic period. A comparison of cell-cycle time in central and peripheral tubules of the same lungs on day 15 showed a slower cell cycle in the central tubules than in the peripheral branches. An estimate of the growth fraction (GF) on each day based on calculated and observed cell-cycle times showed that GF was 1 on days 15 and 16, decreasing to 0.69 on day 17 and 0.27 on day 18. Thus cell-cycle time increases with lung maturation and it increases from peripheral to central zones of the bronchial tree. The greater part of the observed decline in growth rate as the prenatal phase of growth is completed was due largely to a decrease in the fraction of cells in the cell cycle.
