    Notes: Summary Effects of pH on β-galactosidase expression and stabilization were investigated using recombinantE. coli harboring an expression vector with a thermally-inducible PL promoter. Expression of β-galactosidase was strongly promoted by lowering culture pH when culture temperature was raised to the induction temperature. Optimal pH for induction ranged from 5.4–5.8. The degradation of expressed β-galactosidase could be reduced by lowering the culture temperature while at the same time slightly increasing the culture pH in the β-gal degradation stage.

    Notes: In glutamic acid fermentation, the molasses feeding policy and time of penicillin addition significantly affected glutamic acid production, and a fuzzy supervisory control system was developed for their quasi-optimal regulation.From the trend of the experimental data, production rules and membership functions of fuzzy inference were devised to determine the quasi-optimum molasses feeding policy and penicillin addition time. A computer with multitasking operating system was used for the construction of the control system with fuzzy inferencing, which decided the control policy every minute, and the feed rate was controlled automatically. The pattern of residual sugar concentration was almost the same as that of maximum glutamic acid production under manual operation. Using the computer control system, stable production was maintained at the highest level of 71 to 75 g/L. © 1994 John Wiley & Sons, Inc.

    Notes: Abstract In order to establish an easy and quick construction method for obtaining a stable and highly productive gene-amplified recombinant Chinese Hamster Ovary (CHO) cell line, variouskinds of stepwise methotrexate (MTX) selection were carriedout. The specific growth and production rates of the cell were compared with each other, and the distribution of the amplified gene location was determined using fluorescence in situ hybridization (FISH). The specific growth andproduction rates of the cell pool reached the highest levels under the selection condition in which the stepwise increase in the MTX concentration was most gradual; about 82% of amplified genes were observed near the telomeric region. During long-term cultivation without MTX, the percentage ofamplified genes near the telomeric region hardly changed, butthat of amplified genes at other regions decreased. Based on these results, stable and highly productive cell pools could be easily and quickly constructed and amplified and gradual stepwise increase of the MTX concentration. In addition, the FISH technique was powerful tool to evaluate highly productiveand stable gene-amplified cells based on the chromosomal location of the amplified gene.

    Notes: A modified simulation procedure based on a statistical approach was investigated. The procedure predicts the time course of fed-batch culture for glutamic acid production by a temperature-sensitive strain of Brevibacterium flavum. The statistical approach requires only a data base of state points obtained in experiments, and not perfect identification of fermentation models. The simulation procedure is based on regression analysis to estimate specific rate parameters of system equations using the data points selected with reference to the Euclid distance. It was modified in that the data selection procedure included the use of the Maharanobis distance as well as a modified distance defined in this study. Simulation results using the modified procedure allow reasonable prediction of the time course of fed-batch culture for glutamic acid compared to that involving the Euclid distance alone.

    Notes: In many cases of fermentation processes, it is difficult to measure all of the state variables needed to indicate the culture state and to predict state changes. We propose the use of time-series data of measurable state variables instead of unmeasurable to describe the fermenatation process. We also employ a discrimination method of independent variables of regression analysis using statistical measures, PSS and AIC. These statistical procedure were applied to fed batch culture for yeast production under aerobic conditions, and the effectiveness was ascertained experimentally.

    Notes: Production of cellulase using solid culture systems of Trichoderma reesei QM9414 and Sporotrichum cellulophilum on wheat bran was studied. By using moisture-controlled solid culture equipment, the effect of water content of wheat bran on cell growth and cellulase production was investigated. Cellular biomass grown on solid substrate was estimated by measuring oxygen consumption rate and glucosamine content of the cells. These parameters were shown to have a good linear correlation with the specific growth rate. This reliable method of estimating the cell growth rate enabled us to simulate the enzyme production in a solid culture system by means of multiple linear regression analysis which takes into account of the water content, cell mass, and the oxygen consumption rate as variables. The cell growth and cellulase production were maximized at different water content of the medium. A high water content, 57% for T. reesei and 70% for S. cellulophilum, favored mycelial growth, while the maximum cellulase activity was obtained at a lower water content such as 50% for both fungi. It was observed that cellulase production by T. reesei depended on the culture conditions that support the optimal growth rate for the maximum enzyme production.

    Notes: During fed-batch cultivation of a recombinant Escherichia coli AT2471 harboring plasmid pSY130-14 for phenylalanine production, a large amount of acetic acid was excreted by the cells and accumulated in the culture medium. Acetic acid concentration reached 30-35 g/L at the end of a process conducted without special precautions for the reduction of this excretion. Cell growth stopped when acetic acid concentration was about 15 g/L, resulting in poor growth, 16 g/L cell concentration, and poor production  -  8 g/L phenylalanine. A novel control strategy, called a balanced DO-stat. was developed to prevent acetic acid excretion. It represents a model-independent two-loop control structure, which is simple, reliable, and convenient for computer application. Using the balanced DO-stat, implemented in a computer control system, acetic acid concentration was kept at zero during the entire cultivation period. As a result, the cell concentration increased to 36 g/L and phenylalanine concentration reached 24 g/L. Aside from the phenylalanine fermentation, the proposed control approach might be applied to cultivation of other bacterial and yeast strains which have similar mechanism of the excretion of fermentative by-products.
