    Characterization and crystallization of a novel Sarcocystis muris lectin, SML-2 (2001)

    Notes: A novel lectin (SML-2) consisting of 138 amino acids was isolated from cyst merozoites of Sarcocystis muris and sequenced by Edman degradation and mass spectrometry. All 12 cysteinyl residues are involved in disulfide bridges, four of which are attributed to a characteristic pattern of cysteines as found in the so-called PAN-module superfamily. Crystals of SML-2 diffracting to 2.1 Å resolution at a synchrotron were grown by the hanging-drop vapour-diffusion technique. They belong to the space group P212121, with unit-cell parameters a = 53.6, b = 128.8, c = 158.2 Å and eight molecules in the asymmetric unit. SML-2 cocrystallized with Au galactose results in two different crystal forms. The first form is isomorphous with the native crystals and the second form adopts space group C2221, with unit-cell parameters a = 74.7, b = 82.0, c = 131.0 Å, and diffracts to 2.4 Å at a rotating-anode X-ray generator.

    Notes: Abstract The mature major microneme protein of Sarcocystis muris cyst merozoites, which is known as a dimeric lectin with high affinity to galactose and some of its derivatives, was expressed in Escherichia coli as a histidine-tagged fusion protein. The recombinant polypeptide, which was recognized by a monoclonal antibody directed against the native lectin, was purified from inclusion bodies after solubilization and refolding, using a combination of metal chelate and lactose affinity chromatography. The apparent molecular mass of the refolded polypeptide as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoreses was 16 kDa, whereas gel filtration chromatography clearly demonstrated that the recombinant protein, like its native counterpart, exists as a homodimer of two non-covalently associated subunits. Inhibition of haemagglutination suggests that the combining site of the recombinant lectin recognizes N-acetyl-galactosamine as the dominant sugar, thus confirming the correct folding of the monosaccharide combining site in the renatured lectin. To the best of our knowledge, this work represents the first reported detailed characterization of a recombinant lectin from apicomplexan parasites, and may contribute to a better understanding of the process of host cell recognition and invasion by these obligate intracellular protozoa.

    Description / Table of Contents: The Electron Impact Induced Elimination of Water from 3-Hydroxy-5β-steroids3-Hydroxysteroids of the 5β-series eliminate water very readily, in particular the 3α-alcohols. Deuteriation of 5β-cholan-3α-ol at C-9 proved that the hydroxyl group abstracts the deuterium in the course of the dehydration, after ring A has changed to the boat conformation. A different mechanism must operate in the corresponding decomposition of [9α-D1]-5β-cholan-3β-ol, because only H2O is ejected. In general, the spatial distance of the hydroxyl group to an activated hydrogen seems to govern the extent of the water elimination. At least in this instance, the ring conformation has the same importance in reactions of radical ions in the gas phase as it has in reactions of molecules in solution.
    Notes: 3-Hydroxysteroide der 5β-Reihe spalten auffallend leicht Wasser ab, insbesondere die 3α-Alko-hole. Durch Deuterierung der 9α-Position im 5β-Cholan-3α-ol ergab sich, daß die Hydroxyl-gruppe bei der Dehydratisierung nach Umklappen des Ringes A in die Bootform das Deuterium abstrahiert. Für das entsprechende [9α-D1]-5β-Cholan-3β-ol muß ein anderer Mechanismus wirksam sein, da hier nur H2O eliminiert wird. Der Abstand der Hydroxylgruppe zu einem aktivierten Wasserstoff scheint ganz allgemein für das Ausmaß der Wasserabspaltung maßgebend zu sein. Die Konformation spielt bei der Reaktion von Ionen offenbar eine ähnliche Rolle wie bei Umsetzungen von Molekülen in Lösung.

In general the CD of these diketones follows the octant rule. In some cases strong vicinal effects have been observed, however.
    Notes: Ausgehend von 4.8-Dioxo-adamantan-carbonsäure-(2) (1) wurden einige solche monosubstituierte Adamantandionderivate in optisch aktiver Form neu dargestellt, deren Substituenten π-Elektronensysteme enthalten. Der CD dieser Diketone folgt im Prinzip der Oktantenregel. In einigen Fällen treten aber beträchtliche Vicinaleffekte auf.

Removal of the nitro group reduction of the ester groups, cleavage of the acetal group, and transformation of the alcohol functions into tosylates yielded compound 14. Cyclisation of 14 resulted in the tetracyclic compound 17. Addition of bromine and reaction with NaOH gave 21, the first known benzenoadamantanone derivative. The stereochemistry of the intermediates was established by NMR investigations.
    Notes: Michael-Addition von Methylenmalonester an das Tetrahydro-10-nitro-5,9-methano-7H-benzocyclohepten-7-on-ethylenacetal 9 lieferte die Ausgangsverbindung 11. Abspaltung der Nitrogruppe, Reduktion der Estergruppen, Entfernung der Acetalgruppe und Umwandlung der Alkoholfunktionen in Tosylate ergab die Verbindung 14. Eine Cyclisierung dieses Produktes führte zum Tetracyclus 17. Bromaddition und Umsetzung mit NaOH ergab 21, das erste bekannte Benzenoadamantanon-Derivat. Die Stereochemie der Zwischenprodukte wurde durch NMR-Untersuchungen ermittelt.

    Solving moving boundary problems with an adaptive moving grid method: Rotary heat exchangers with condensation and evaporation (1996)

    Description: A numerical method for the treatment of moving discontinuities in the model equations of chemical engineering systems is presented. The derived model describing the effects of condensation and evaporation in a regenerative air to air heat exchanger yields an illustrative example for these so called moving boundary problems. The presented adaptive moving grid method is based on the algorithm {\sc Pdex} for parabolic partial differential equations. It is shown that the method is suited for problems where the arising discontinuities cause low rates of convergence if the equations are solved with a static grid.
