    Notes: Abstract Consumption of nitric oxide (NO) in a humisol was studied at 0.1–2 ppmv NO, a range representative of NO concentrations in ammonium-fertilized soil. Denitrification was not a major sink for NO. The principal NO-consuming reaction was a biological oxidation, leading ultimately to nitrate (NO3 –). Nitrogen dioxide (NO2) and nitrite (NO2 –) may have been intermediates in this pathway. An abiological reaction accounted for about 25% of the NO-consuming activity in soil at 90% H2O (d.w.) and 25oC, but contributed relatively more to total NO consumption at higher temperatures. Biological NO-consuming activity was highest at 25oC, while the abiological activity increased exponentially with temperature. The product of the abiological reaction was neither NO3 –, NO2 –, nor nitrous oxide (N2O), and the reaction did not require O2.

    Notes: Abstract A methanotrophic nitrifying consortium was previously obtained from a humisol which showed CH4-dependent nitrification. Although the methanotroph could not be obtained in pure culture, three other members of the consortium have been isolated: An obligately methylotrophic Methylobacillus (Is-1) which grows only on CH3OH and does not nitrify; a Pseudomonas (Is-2) which grows on Is-1 culture filtrate and produces NO2−, NO3− and N2O from NH2OH, and NO3− from NO2−; and a second Pseudomonas (Is-3) which produces NO3− from NH4+ or NO2−, and N2O from NH2OH. A model is proposed for the trophic relations and nitrogen transformations in the consortium which may apply to some natural systems.

    Notes: Abstract A denitrifying Cytophaga was isolated from soil enriched by anaerobic incubation with glucose, sulfide (S2−), nitrous oxide (N2O), and acetylene (C2H2). Such soil enrichments and pure cultures of the isolated Cytophaga reduced N2O rapidly even in the presence of a normally inhibitory concentration of C2H2 (4 kPa) providing S2− was present (8 μmol/g soil or 0.4 μmol/ml culture). Since C2H2 inhibition of the reduction of N2O is used as a tool in the assay of denitrification, the presence in large numbers of such a Cytophaga may influence the effectiveness of this assay especially in sulfidic environments.

    Notes: Abstract A consortium was enriched from a humisol incubated with 3.6 kPa CH4 and NH4+. This consortium oxidized NH4+ to NO2− and NO3− (NO3−/NO2− ratio about 20) with smaller amounts of N2O. This oxidation stopped in the stationary phase after depletion of CH4. CH3OH or CO2 did not support oxidation. Growth and resting cell experiments suggested that nitrification was associated with methanotrophic activity and that chemoautotrophic nitrifiers were absent.

    Notes: Abstract A gel-stabilized system with counter gradients of CH4 and O2 was used to grow methanotrophs from wetland, agricultural and forest soils and lake sediment. Columns of semi-solid nitrate- or ammonium-minerai salts medium were continuously flushed at opposite ends with CH4 and O2 to create opposing concentration gradients of the two gases. Methanotrophs grew from all samples except forest soil, and were visible as thin bands after 5 to 15 days of incubation. The position of growth was CH4 and O2 concentration-dependent and occurred at the point of maximum possible CH4 oxidation, where both substrates were completely consumed. Evidence was obtained for denitrification and nitrification activities concomitant with CH4 oxidation. This approach may be useful to isolate methanotrophs with different CH4 and O2 requirements and to study their interactions with other groups of bacteria in nature.

    Notes: Abstract: A component of Hamilton Harbour sediment prevented nitrous oxide (N2O) reduction in denitrification assays with a mixed population of endogenous bacteria and a pure culture (HH1) isolated from the sediment. A 5% (v/v) concentration of sediment in nutrient broth caused near maximum inhibition of N2O reduction. Sediment taken from a site closer to pollution sources (Site 906) was twice as inhibitory (as measured by N2O accumulation) as sediment from Site 910, further from pollution sources. N2O persistence was associated with the particulate sediment fraction only. Several heavy metals were tested at in situ concentrations, and ionic cadmium (Cd) and chromium (Cr) caused N2O accumulation. Ashed sediment did not cause N2O accumulation, but did decrease initial nitrate reduction rates with HH1.

    Notes: Abstract The subcellular distribution of nitrous oxide reductase was studied in the gliding soil bacterium Flexibacter canadensis. Nitrous oxide reductase activity, as measured by the methyl viologen-nitrous oxide oxidoreductase assay, was associated entirely with the membrane fraction of cell-free extracts. The enzyme was liberated from the membranes with use of detergents but not by high-salt concentrations, thus implying that nitrous oxide reductase is an integral membrane protein. The nitrous oxide reductase of F. canadensis is the first reported example of a membrane-bound form of this respiratory enzyme.

    Notes: When the effect of water content was minimized, soil CO2 evolution and soil organic matter content were good predictors of aerobic NO. uptake rate constants across a wide range of soil types. Field manure application to a Gleysol stimulated NO. uptake rate constants and lowered NO. compensation points compared to unfertilized or NH4NO3-fertilized soil. This effect lasted for months after manure application. In a laboratory experiment, addition of manure reduced the NO. efflux associated with nitrification of NH4 Cl fertilizer, and manured soils had a greater capacity to remove NO. from polluted air. Evidence is presented that these observations result from NO. oxidation during heterotrophic microbial activity in soil.
