To clarify the limiting factors for peak V̇o2 (V̇o2peak)〉 we measured muscle venous Po2 (Pvo2) and V̇o2, peak under various O2 delivery conditions (arterial O2 concentration X flow) via the alteration of blood flow and haematocrit (Hct) in anaesthetized dog gastrocnemius muscle (n= 11) stimulated by 1 Hz isometrically and tetanically.2. Two levels of flow (high and moderate) were maintained by using a pump (ISO and 100 mL/min per 100 g, respectively). Haematocrit was adjusted to 45 and 30% by isovolaemic haemodilution with dextran (MW 40 000).3. The decrease in Hct induced a 31% decrease in O2 delivery (P〈0.05), a 9–12% decrease in Pvo2, a 23% decrease in V̇o2, peak(P〈0.05) and a 12–14% increase in O2 extraction (V̇o2/O2delivery) at both flow levels. The decrease in flow induced a 24–25% decrease in O2 delivery (P〈0.05), a 15–17% decrease in Pvo2 (P〈0.05), a 9% decrease in V̇o2,peak (P〈0.05) and a 19–22% increase in O2 extraction (P〈0.05) at both Hct levels.4. The results suggest that the limiting factor of V̇o2peak, unrelated to O2 diffusion limitation (i.e. the change in the heterogeneity of V̇o2 to O2 delivery ratio among capillaries), due to lowering Hct may be different from that due to lowering blood flow in maximally contracting muscle.

It has previously been shown that perfusion with high O2-affinity-erythrocytes decreases venous PO2 (PVO2) and decreases O2 uptake (V.O2) in contracting muscle at the same O2 delivery (arterial O2 concentration × flow). A linear V.O2–PVO2 relationship has been obtained with a V.O2-axis intercept, suggesting that, during this type of hypoxia, V.O2 is composed of a PVO2-dependent and -independent V.O2. However, the V.O2– PVO2 relation during hypoxic hypoxia has not been examined.2. To clarify this relation, PVO2 and V.O2 have been measured in contracting gastrocnemius (1 Hz trains of 0.2 s isometric tetani) under both normoxic and hypoxic conditions during 5 min of stimulation.3. Venous O2 changes proportionally with O2 delivery. Each V.O2–PVO2 relation was linear, with the mean described by the equation V.O2 = 5.06 + 0.41 × PVO2 (n = 6, r = 0.81, P 〈 0.05). The V.O2-axis intercept was significantly different from zero (P 〈 0.05).4. These results were similar to those obtained during hypoxia induced by high O2-affinity-erythrocytes. We conclude that there is a linear relationship between PVO2 and V.O2 above the V.O2-axis intercept, regardless of the type of hypoxia.

    Notizen: Abstract: We have examined whether mouse plasma erythropoietin (EPO) can be measured by an improved enzyme-linked immunosorbent assay (ELISA) using milk proteins (Block Ace) both as a blocking reagent and as a diluent for standard recombinant human EPO (rHuEPO) and for plasma samples. Block Ace brought about high slope sensitivity of the standard curve, with a low background. The dose–response curves of normal or anaemic mouse plasma and of rHuEPO were linear and parallel to each other. The anaemic plasma had an additive effect with rHuEPO by increasing the absorbance at 405 nm. The coefficients of variation in the intra- and interassays ranged from 4.2% to 15.3%. The plasma EPO levels in 22 normal mice were 18.3 ± 10.3 mU/ml. An inverse relationship between the logarithm of plasma EPO concentrations and blood haemoglobin concentrations, red blood cell counts or packed cell volumes was found in normal mice and in mice with iron deficiency anaemia (IDA). These results show the validity for the use of the new improved ELISA method for measuring circulating murine EPO.

    Notizen: Abstract Plasma erythropoietin (EPO) activity was determined by an erythroid colony-forming microassay during the development of and recovery from iron deficiency anaemia (IDA) in mice. Growing mice were made iron deficient by feeding a low-iron diet. During the development of IDA, the plasma EPO level increased as the haematocrit (Hct) value decreased in the time-dependent fashion. Likewise, reticulocytosis was enhanced depending on the severity of anaemia or the plasma EPO level. After 9 weeks of the feeding, the development of IDA was confirmed by blood analyses. During the recovery from IDA after switching to an iron-rich diet and/or parenteral iron administration, the plasma EPO levels rapidly declined to normal level, while the Hct values increased and completely returned to normal on day 6. Administration of recombinant human EPO (rHuEPO) in addition to the iron-rich diet and iron brought about the supranormal levels of Hct after day 6. Thus, rHuEPO had no therapeutic role in IDA. The plasma EPO titres were inversely correlated with the Hct values during the progression and reversal of IDA. This result supports a generally accepted concept that the EPO production in anaemia is primarily regulated by Hct feedback control mechanism.
