    Notizen: Abstract Growth ofLactobacillus acidophilus was inhibited in the presence of deoxyadenosine-5′-monophosphate (dAMP). The other purine deoxyribonucleotides and -sides were only weak inhibitors, and pyrimidine deoxyribotides and -sides were inactive. dAMP did not act as an inhibitor if thymine, thymidine, or 5-methyldeoxycytidine-5′-monophosphate was present in the medium. The inhibition by dAMP was counteracted by increasing concentrations of deoxyuridine, deoxyuridine-5′-monophosphate and, to some extent, adenosine-5′-monophosphate. The effect of these substances was proportional to their concentration and competitive in character. The results support the assumption that dAMP inhibits the synthesis of thymine. Mutants ofL. acidophilus resistant to inhibition by dAMP were found.

    Notizen: Abstract Previously, we established experimental conditions allowing us to induce hypomethylation of tandem arrays of highly repetitive DNA sequences in the Nicotiana tabacum L. nuclear genome (M. Bezděk et al. 1991, Planta 184, 487–490). In this paper, we demonstrate that loci containing the highly repetitive sequences of the HRS60 family can maintain the induced hypomethylated state through protoplast regeneration, non-differentiated callus growth, and plant regeneration. The hypomethylation, induced with 5-azacytidine and monitored on these sequences, did not substantially alter the capacity of calli to produce plants. It can be concluded that, in contradistinction of multiple copies of transgenes or ectopic genes which are usually recognized as methylation targets, endogenous tandem repeats, such as the HRS60, present at 105 copies in the genome, can escape de-novo methylation.

    Notizen: Abstract The methylation status and 5-azacytidine-induced hypomethylation of CCGG sites within a family of tandemly organized, highly repeated DNA sequences of the Nicotiana tabacum L. nuclear genome (HRS60 family) were studied. As shown by in-situ hybridization experiments, the HRS60 family is clustered in a few regions of some tobacco chromosomes. The DNAs of leaf-derived calli, leaf-derived calli cultured on media with 5-azacytidine, and leaves were cleaved with restriction endonucleases differing in the sensitivity to the methylation of cytosine. After electrophoresis and Southern blotting they were hybridized with the HRS60 probe. We show that (i) CpG dinucleotides, and partially also CpCpG trinucleotides, of the HRS60 family are methylated in DNAs of the non-treated calli and leaves, and (ii) that these DNA repeats are sensitive to the action of a hypomethylating drug, 5-azacytidine.

    Notizen: Summary Chloramphenicol (CLP) at slightly inhibitory concentrations (3–5 μg/ml) suppresses the inhibition of DNA synthesis and cell division caused by the temperature sensitive mutation dnaE486 at the nonpermissive temperature. Some other mutations can also be phenotypically suppressed by CLP. A similar effect is shown by chlortetracycline. Phenotypic suppression is caused by both these drugs in different cases than by streptomycin.

    Notizen: Summary Synthesis of DNA in Lactobacillus acidophilus R-26 was investigated in the presence of rifampicin which inhibits RNA and protein synthesis. Increments in DNA of between 140 and 200% were found under these conditions. Indirect evidence is presented that these large increments are not due to the presence of several replication points in the bacterial chromosome at the time of addition of the drug. Incubation with rifampicin was found to result in a progressive decrease in the capacity for DNA synthesis. This decrease was independent of the amount of DNA synthesized during incubation with rifampicin but was dependent on the time of incubation in the presence of the drug. It is proposed that inhibition of protein and RNA synthesis in L. acidophilus does not inhibit initiation of new cycles of chromosome replication but results in a progressive loss of the capacity to replicate DNA.
