    Notes: Abstract. The effects on normal and mildly burnt oral epithelium of rat palate of commonly used fixatives differing in osmolality and composition were examined quantitatively by the application of a stereological method at the electron microscope level. With five out of six different fixative solutions the intercellular spaces of the burnt epithelium occupied a significantly larger relative volume than in normal epithelium, and the tissue did not respond in the same way as normal epithelium does to differences in fixative solution. This may reflect changes in tissue properties resulting from burning. The differences in size of intercellular spaces between tissues fixed in different solutions were such that burnt tissue fixed in one solution did not always show larger intercellular spaces than normal tissue fixed in another solution. Observations of the ultrastructural changes of inflammation in oral epithelium should therefore always he checked by observations on identically fixed normal tissue.

    Notes: Abstract. The fine structural characteristics of five adenoid cystic carcinomas of human minor salivary glands and a quantitative assessment of the relative volumes occupied by morphologically defined cell types in these tumors are reported. We observed that the cyst-like spaces which give the characteristic cribriform pattern to the adenoid cystic carcinoma contain replicated basement membrane-like material. Material comprising aggregates of fine tubules having a median diameter of 270 A, and rounded, electron dense bodies were noted within duct-like lumena of one tumor. In addition, dilatation of the intercellular spaces and squamous metaplasia were noted. From 500 electronmicrographs obtained by standardized techniques and used for the morphological part of the study, 175 were selected by a random sampling method and analyzed by the stereological technique of point counting. This method demonstrated that duct type cells occupied 75 % by volume of the tumor in these glands; myoepithelial cells occupied 3 %, acinar cells occupied 2 %, and other tissues occupied 22 % of the tumors. These proportions differ significantly (P 〈 0.001) from our previously published figures for normal specimens of these glands.

    Notes: Abstract. Mucosa from the anterior palate of the rat was fixed in a variety of commonly used fixatives for electron microscopy having different osmolalities and chemical compositions. The volume of the intercellular space of the epithelium was assessed using a stereological technique in which a test grid was superimposed over electron micrographs of sections through the epithelium, and the relative areas of that test grid overlying cellular and intercellular components were measured. Epithelium fixed in solutions isotonic with mammalian serum showed an intercellular space occupying approximately 4% of the total tissue volume. There was a tendency for the intercellular space to increase with increasing osmolality of the fixative, although this relationship was not a simple one; the chemical nature of the fixative solution may also influence the response.

    Notes: Teeth undergoing resorption were dissected from the maxillae of frogs and prepared for electron microscopy using standard methods and methods designed to minimize demineralization. The resorptive cells overlapped one another, forming palisades. A central ruffled border was surrounded by a relatively structureless cytoplasmic “seal.” Crystals of mineral were present in the folds of the ruffled borders, and demineralized collagen fibrils were seen in the adjacent matrix in sections treated to prevent demineralization. The surfaces of the cells opposite the ruffled borders displayed numerous small villi.The cytoplasm of the resorptive cells contained a variety of lysosomes in various stages of development, mitochondria, scattered vesicles of endoplasmic reticulum and free ribosomes. Well developed Golgi zones were frequently seen, and occasional sections passed through centrioles. Two centrioles were commonly seen in such sections, but never more. Occasional patches of cytoplasm contained mitochondria and rough-surfaced endoplasmic reticulum, resembling “blast” cell cytoplasm.We concluded that the resorptive cells of several mineralized tissues of various vertebrates are morphologically, indistinguishable.
