    Anti-Leu 19 monoclonal antibody detects an antigen on autonomic nerves in human skin (1988)

    Notes: The monoclonal antibody anti-Leu 19 recognizes the NKH-1 antigen of active natural killer cells. In immunohistological preparations of the skin, the antibody reacts with cutaneous nerves and labels nerve fibers in the region of the sweat glands, the blood vessels and the hair arrector muscles. This staining pattern corresponds to the distribution of the autonomic sympathetic nervous system of the skin. On the other hand, anti-Leu 7 labels a myelin-associated glycoprotein which is to be found in the myelin-sheath of sensory cutaneous nerves. The reactivity of neural structures of the skin can thus be differentiated. Anti-Leu 7 labels the myelinated sensory nervous system, whereas anti-Leu 19 seems to constitute a new marker for the autonomic nervous system of the skin.

    Description / Table of Contents: Zusammenfassung An einem umfassenden Krankengut der progressiven Sklerodermie (n = 20) wurden gegenüber Kontrollen Hexosaminbestimmung und anschließende säulenchromatographische Auftrennungen durchgeführt. Verwendung fanden hierbei moderne Testmethoden zur Darstellung der Parameter des Mucopolysaccharidstoffwechsels (Hexosaminbestimmung nach Südhoff u. Petrovic sowie das säulenchromatographische Auftrennungsverfahren nach Greiling u. Stuhlsatz. Die mit der Hexosaminbestimmung dargestellte vermehrte Ausscheidung in Korrelation zu erheblichen Sulfateinbauraten in die Haut wurde im Sinne einer Acceleration des Proteoglykanstoffwechsels in der Haut gedeutet. Die entsprechenden Ausscheidungsprodukte konnten nicht als typische Vertreter der in der Haut befindlichen Proteoglykanbestandteile identifiziert werden. Zur Darstellung kam wiederum ein erhöhter Anteil von Glykopeptiden sowie ein erhöhter Anteil von Chondroitin oder besonders niedrig sulfatierter Chondroitinsulphat-Bruchstücke. Damit konnte aufgezeigt werden, daß die Hexosaminbestimmung im Urin in ihrer Aussagekraft und Rückschlüssen auf den Mucopolysaccharidstoffwechsel limitiert ist.
    Notes: Summary After filtration the urine was concentrated to about 1/10 of the original volume. The urine was centrifuged for 15 min at 5000 rev/min. The supernatant was chromatographed on Bio-Gel P-2—with a 10% aqueous solution of ethanol. Then the glycosaminoglycan fraction was separated into seven further fractions by chromatography on Dowex 1 × 2 and elution with increasing concentration of sodium chlorid. The desalted fractions were analysed. From the analytical data presented no definite correlation between excretion of glycosaminoglycans in urine und progressive scleroderma can be observed. These results were discussed with further data presented by Hexosamine determination.

The analysis indicates that the ratio of hexosamine to collagen was higher in the center of the tumor than in the outer areas. Our findings could result from dysregulation of the biosynthesis of glycosaminoglycanes.

Blutchemische Veränderungen einschließlich der Enzymaktivitäts-bestimmungen sollten nie alleine zur Sicherung der Diagnose beitragen. Die Möglichkeiten und Grenzen der konventionellen histologischen Diagnostik sowie des EMG wurden ebenfalls aufgezeigt. Die bisher bei uns an der Klinik durchgeführte perorale Kreatinbelastung konnte zusätzliche Informationen für eine Schädigung der Muskulatur auch in den Fällen zeigen, bei denen die herkömmlichen Methoden, wie Histologie und EMG, bisher versagten, da die Aufnahmefähigkeit der Gesamtheit der Muskulatur unter der Belastung einer Beurteilung unterliegt.
    Notes: Summary The possibilities and limitations of the conventional histo-morphological procedures and of the electromyogram in the diagnosis of dermatomyositis are discussed. Changes in enzyme activities (transaminases, creatin-kinase0 in blood may be observed. With the peroral creatine tolerance test it is possible to find injuries of the muscular system, even in those cases where the conventional methods (histological examination and electromyogram) have so far failed. The advantage of this test is that the capacity of the whole muscular system to accumulate creatine can be determined.

    Description / Table of Contents: Zusammenfassung An einem Krankengut von 80 Patienten (Kontrollenn = 20, progressive Sklerodermien = 20, Dermatomyositisn = 23, Pseudoxanthoma elasticumn = 10, Psoriasis vulgaris, guttata, geographican = 7) wurden vergleichende Collagen-like-Protein-Bestimmungen im Serum durchgeführt. In Abhängigkeit zum klinischen Verlauf konnte vor allen Dingen bei der progressiven Sklerodermie und besonders bei den schweren Verlaufsformen der Dermatomyositis sowie bei der Psoriasis vulgaris eine Erhöhung des CLP-Spiegels nachgewiesen werden. Diese Werte standen in guter Übereinstimmung mit den bisher im Schrifttum mitgeteilten Untersuchungsbefunden. Eine Beziehung zum Kollagenstoffwechsel erscheint trotz noch nicht sicher aufgeklärter Beziehungen bzw. Einzelschritte des CLP aufgrund korrelierender erhöhter Hydroxyprolinausscheidung im Urin zu bestehen.
    Notes: Summary The collagen-like-protein (CLP) in sera of patients with skleroderma (n = 20), dermatomyositis (n = 23), Pseudoxanthoma elasticum (n = 10), Psoriasis vulgaris (n = 7) and control subjects has been determined. The increase of collagen-like-protein roughly parallels the activity of the diseases except for Pseudoxanthoma elasticum. In later phases a decrease has been observed. A relationship to collagen biosynthesis or metabolism is discussed.
