Zusammenfassung An Hand ausgedehnter experimenteller Untersuchungen wird über die Eigenschaften von Glasmikroelektroden bei der intracellulären Membranpotentialmessung berichtet. 1. Je negativer das tip-potential der Elektrode, umso stärker wird das Membranpotential verfälscht. Für jeweils 10 mV tip-potential ist eine mittlere Verminderung des Membranpotentials um 2,8 mV zu erwarten. 2. Die Ursachen des tip-potentials liegen in einer elektrischen Aufladung der inneren Wand der Elektrodenspitze in Verbindung mit dem Konzentrationsgradienten von Elektrodenfüllung zur Versuchslösung. 3. Der Übergang von Natrium- auf Kaliummilieu beim Einstechen der Elektrodenspitze in die Zelle erklärt nur einen Teil der in Abhängigkeit zum tip-potential tatsächlich auftretenden Verfälschung des Membranpotentials. 4. Da das tip-potential der Elektrode vom pH-Wert der Testlösung abhängt und durch den pH-Wert des Milieus die Aufladungen an der Glaswand der Elektrodenspitze verändert werden, wird diskutiert, daß bei eingestochener Mikroelektrode die elektrischen Ladungszustände an der Elektrodenspitze vom elektrischen Feld des Membranpotentials beeinflußt werden. 5. Um das störende tip-potential auszuschalten bzw. möglichst niedrig zu halten, wird die Verwendung von 0,5 molarer KCl-Lösung als Elektrodenfüllung empfohlen.

Abstract A study has been made of the behaviour of glass-microelectrodes for intracellular measurements. The following facts have been obtained on a large number of experiments: 1. The more negative the tip-potential of the electrode the greater the difference of the membrane potential from the actual value. For each 10 mV of the tippotential the membrane potential decreases by about 2.8 mV. 2. The tip-potential is caused by the electrical charge of the inner surface of the elctrode-tip in connection with the concentration gradient between the solutions within and outside the electrode. 3. The passage of the electrode from a high Na+- to a high K+-medium outside and inside the muscle cell respectively is only partly responsible for the error in the membrane potential values. 4. It has been shown that the tip-potential of the electrode depends on the pH of the testsolution, i.e. the pH affects the charge on the inner surface of the glass-wall at the tip of the electrode. It is discussed that when the tip of the electrode is introduced into the cell, the electrical charge on the glass wall is affected by the electrical field of the membrane potential. 5. For reducing the interfering tip-potential it is recommended to fill the electrode with 0.5-molar KCl-solution.
