    Notes: Summary Three DNA plasmids, designated pScrl-1, pScrl-2, and pScrl-3 have been found in a strain of the heterothallic yeast Saccharomycopsis crataegensis (NRRL Y-5902). pScrl-l, -2 and -3 are, respectively, 15, 7, and 5 kilobase pairs (kbp) in size. Based on the results of exonuclease digestions, all three plasmids appear to be linear molecules with blocked 5′ ends. All three plasmids also have a lower buoyant density than does nuclear DNA of S. crataegensis. The two lower molecular weight plasmids hybridize strongly with one another, but only weakly to the higher molecular weight plasmid. Two of four related S. crataegensis strains surveyed were found to contain two plasmids that are of the same size as the two larger plasmids of Y-5902. Evidence is presented indicating that the plasmids in strain Y-5902 reside in the cytosol since they were found not to be located within the major organelles (mitochondria and nuclei).

    Notes: Abstract The phylogenetic relatedness of Issatchenkia spp. was estimated from partial rRNA sequences in two regions of the large subunit and one region of the small subunit. I. terricola was the most divergent species of the genus, differing from other members by 18% nucleotide differences in the highly variable 25S-635 region. These data indicate Issatchenkia to be the most divergent ascomycetous yeast genus presently known.

Moderate (36–68%) DNA relatedness was detected between the formerPichia sargentensis and varieties ofW. saturnus again demonstrating that nitrate assimilation is not a reliable criterion for separating yeast species.

    Notes: Abstract An unknown yeast species was isolated from maize silage and was determined to be novel on the basis of morphological and physiological characteristics, nucleotide sequence of domain D1/D2 of LSU rDNA and from its electrophoretic karyotype. The name for the proposed new species is Saccharomyces bulderi Middelhoven, Kurtzman et Vaughan-Martini (type strain CBS 8638, NRRL Y-27203, DBVPG 7127). S. bulderi is closely related to S. barnettii and S. exiguus from which it can be distinguished by having a double vitamin requirement of biotin and thiamine and by no or slow aerobic growth on raffinose, a sugar that on the contrary is fermented rapidly. Gluconolactone is rapidly fermented with ethanol, glycerol and carbon dioxide being the main products.

    Notes: Abstract Approximately 500 species of ascomycetous yeasts, including members of Candida and other anamorphic genera, were analyzed for extent of divergence in the variable D1/D2 domain of large subunit (26S) ribosomal DNA. Divergence in this domain is generally sufficient to resolve individual species, resulting in the prediction that 55 currently recognized taxa are synonyms of earlier described species. Phylogenetic relationships among the ascomycetous yeasts were analyzed from D1/D2 sequence divergence. For comparison, the phylogeny of selected members of the Saccharomyces clade was determined from 18S rDNA sequences. Species relationships were highly concordant between the D1/D2 and 18S trees when branches were statistically well supported.

    Notes: Abstract Saturnospora ahearnii, a new, nonhyphal saturn-spored yeast, is described. The type strain (NRRL Y-7555, CBS 6121) of this species was isolated from the rhizosphere of marsh grass in Louisiana. Comparisons of nuclear DNA complementarity showedS. ahearnii to be closely related toS. saitoi, a species presently known only from Japan.

    Notes: Abstract A 230-nucleotide region of the large subunit (LSU) ribosomal RNA was examined to determine whether signature nucleotide sequences could be used for species identifications of basidiomycetous yeasts. Multiple strains of genetically defined heterothallic species ofRhodosporidium, Leucosporidium, Cystofilobasidium, andSporidiobolus demonstrated that nucleotide sequences within these species are homologous and that differences between species range from 1 to 20 or more bases. Also included in this study were several homothallic species of these teleomorphic genera and some anamorphs assigned toRhodotorula andCandida. Those results indicated close relationships among certain homothallic species, particularly in the genusMrakia, and potential relationships of homothallic and anamorphic strains to several teleomorphs. The data suggest that LSU sequences can be used for yeast identifications with the possible exception of closely related homothallic species.
